Background. In recent years, the incidence and mortality of colorectal cancer (CRC) are increasing, and the 5-year survival rate of advanced metastatic CRC is poor. Small mothers against decapentaplegic (SMAD) superfamily are intracellular signal transduction proteins associated with the development and prognosis of a variety of tumors. At present, no study has systematically analysed the relationship between SMADs and CRC. Methods. Here, R3.6.3 was used to analyse the expression of SMADs in pan-cancer and CRC. Protein expression of SMADs were analysed by Human Protein Atlas (HPA). Gene expression profiling interactive analysis (GEPIA) was used to evaluate the correlation between SMADs and tumor stage in CRC. The effect of R language and GEPIA on prognosis was analysed. Mutation rates of SMADs in CRC were determined by cBioPortal, and potentially related genes were predicted using GeneMANIA. R analysis was used to correlate immune cell infiltration in CRC. Results. Both SMAD1 and SMAD2 were found to be weakly expressed in CRC and correlated with the immune invasion level. SMAD1 was correlated with patient prognosis, and SMAD2 was correlated with tumor stage. SMAD3, SMAD4, and SMAD7 were all expressed at low levels in CRC and associated with a variety of immune cells. SMAD3 and SMAD4 proteins were also expressed at low levels, and SMAD4 had the highest mutation rate. SMAD5 and SMAD6 were overexpressed in CRC, and SMAD6 was also associated with patient overall survival (OS) and CD8+ T cells, macrophages, and neutrophils. Conclusions. Our results reveal innovative and strong evidence that SMADs can be used as biomarkers for the treatment and prognosis of CRC. © 2023 Ning Ding et al.

Objective. The objective of this study was to explore the medicinal properties of herbal medicines that can interfere with the copper death pathway. Methods. The Human Gene Database, Chemical Interactions in Comparative Toxicogenomics Database, Encyclopedia of Traditional Chinese Medicine, China Medical Information Platform, and Cytoscape software were used to find target and chemicals that interfere with copper death targets, as well as herbal medicines containing these chemicals and their four natures and five flavors (basic properties of herbal medicines). Results. 27 copper death-related targets were finally retrieved, as well as 2143 chemicals that could interfere with them, including 180 herbal compounds. The compounds with the highest degree values (number of nodes connected to this node) were folic acid, resveratrol, and quercetin. The 180 compounds were related to 278 herbs; those with the highest degree values (number of nodes connected to this node) were Jujubae Fructus, Ginkgo biloba L, and Acanthopanax senticosus. The 27 copper death targets were indirectly associated with 278 herbs; those with the highest degree values (number of nodes connected to this node) were Achyranthis Bidentatae Radix, Polygonum cuspidatum Sieb. et Zucc, and Mori Folium. Among the 278 herbs, 6 had incomplete information. A pharmacological analysis showed that among the 272 Chinese herbs, the most frequent meridians were the liver (133), lung (104), and spleen (91). Of the four natures, the most frequent were cold (73), warm (68), and flat (45). Of the five flavors, the most frequent were bitter (165), pungent (116), and sweet (99). Conclusion. This study preliminarily discussed the material basis and medicinal properties of herbs that can intervene in copper death, which can provide reference for the theoretical discussion, drug development, and clinical research of Chinese medicine regulating copper death. © 2023 Zhong Dayuan et al.

摘要:
婴幼儿谷类辅助食品是婴幼儿营养的重要能量来源, 其主要原料为大米, 而水稻极易从其生长的土壤和水中吸收重金属Cd积累到谷粒中, 导致大米中Cd的残留浓度相对较高, 从而对婴幼儿谷类辅助食品构成潜在威胁。 提出了电感耦合等离子体串联质谱(ICP-MS/MS)测定婴幼儿谷类辅助食品中重金属Cd的新策略。 样品经微波消解处理后, 采用ICP-MS/MS进行测定。 婴幼儿谷类辅助食品中Cd的测定所面临的质谱干扰主要来源于高浓度Mo所形成的Mo基多原子干扰离子(MoO+, MoN+, MoC+)以及Sn所形成的同量异位素(112Sn+, 114Sn+, 116Sn+)。 针对这些干扰, 在MS/MS模式下, 分别采用H2, NH3/He和O2为反应气, 考察不同Cd同位素的质谱干扰消除效果和分析灵敏度。 结果表明, 在H2和NH3/He反应模式下, H2和NH3均能与Mo基干扰离子发生质量转移反应, 而不与Sn+反应, 可选择110Cd+, 111Cd+和113Cd+进行测定, 但H2与Mo基干扰离子反应速度较慢, 难以彻底消除高浓度Mo基质中对Cd的质谱干扰; 在O2反应模式下, 所有干扰离子均能与O2发生质量转移反应, 虽然获得的分析灵敏度略低于NH3/He反应模式, 但背景等效浓度(BEC)明显比NH3/He反应模式低。 确定了以O2为反应气消除干扰测定Cd的最优分析方案。 利用标准参考物质评价了方法的准确性, 采用国标法(GB5009.268—2016)进行对比分析验证了方法的可靠性。 结果表明, Cd的LOD为2.03~13.4 ng·L-1, 标准参考物质的测定值与认定值基本一致, 在95%的置信水平, 对比分析结果之间无显著性差异。 方法的灵敏度高, 结果准确可靠, 适用于大批量婴幼儿谷类辅助食品中重金属Cd的高通量测定。

摘要:
植物精油是从芳香植物提取的天然复杂化合物, 作为芳香植物的次生代谢产物具有挥发性和浓郁香味, 其特有的多样化生物活性广泛应用于医药和化妆品行业。 植物精油具有高渗透性, 能以活跃的分子态渗透皮肤组织, 经淋巴腺吸收后进入血液, 其所含的重金属元素也极易随植物精油进入人体对健康构成潜在威胁。 采用硝酸-双氧水对植物精油进行微波消解, 在多模式样品导入系统(MSIS)的双重模式下, 采用电感耦合等离子体发射光谱(ICP-OES)测定其中可形成蒸气重金属元素As, Sn, Sb, Hg和不可形成蒸气重金属元素Cr, Ni, Cd和Pb的含量。 选择盐酸对样品进行酸化并预还原氧化态元素, 通过在线加入L-半胱氨酸/酒石酸提高可形成蒸气元素的蒸气发生效率, 利用硼氢化钠/氢氧化钠在MSIS中将As, Sn, Sb和Hg转变为蒸气状态; 针对分析过程中存在的多个或单个光谱重叠和背景干扰, 分别对空白溶液、 分析元素和预期干扰元素的纯溶液进行测定, 根据获得的光谱响应数据解卷积构建快速自动曲线拟合技术(FACT)模型, 将分析谱线从干扰谱线中分离出来, 从而实现光谱重叠干扰和背景干扰的实时校正; 采用加标回收并与电感耦合等离子体质谱(ICP-MS)进行对比分析评价方法的准确性。 各元素方法的检出限(MDL)为0.3811.2 μg·kg-1, 加标回收率为95.4%104%, 相对标准偏差(RSD)为1.9%4.9%, 对比分析的相对误差(RE)在-2.1%2.7%之间, 表明方法准确可靠, 精密度高。 对8种植物精油中的重金属元素进行了分析, 所有植物精油样品中重金属元素As, Hg和Pb的含量远低于GB/T 26516-2011制定的限量标准, 植物精油中重金属元素Cr, Ni, Sn, Cd和Sb的含量虽然没有制定限量标准, 但均处于极低水平。 MSIS兼具传统雾化和蒸气发生双重功能, 在分析可形成蒸气元素和不可形成蒸气元素时无需切换不同进样系统, 能满足大批量植物精油中微量重金属元素的高通量分析需要。

This work was aimed at exploring the efficacy of Ginkgo biloba extract combined with hormones in the treatment of sudden deafness and the influence on the reactivity of peripheral blood T cell subsets (PBTCSs). In this work, 64 patients with sudden deafness who were treated in The First Affiliated Hospital of Hunan University of Traditional Chinese Medicine from August 2019 to August 2022 were selected as the research objects. The patients were randomly divided into a hormone group (treatment with prednisone acetate, n=34) and a combination group (treatment with Ginkgo-Damole combined with prednisone acetate, n=30). After the two groups of patients were treated in different ways, their efficacy, symptom improvement, changes in blood rheology, and PBTCSs were compared. The total effective rates (TERs) of the hormone group and the combination group were 76.32% and 95.73%, respectively (P<0.05). The fibrinogen contents of the patients in the combination group were obviously lower than those in the hormone group after 5 d, 7 d, and 10 d of treatment (P<0.05). The high blood viscosity (HBV) values of patients in the combination group at 5 d, 7 d, and 10 d after treatment were greatly lower than those in the hormone group (P<0.05). The low blood viscosity (LBV) values after 3 d, 7 d, and 10 d of treatment in the combined group were much lower in contrast to those in the hormone group (P<0.05). The CD3+, CD4+, and CD4+/CD8+ in peripheral blood in the combination group were sharply higher while the CD8+ in the combined group was lower in contrast to the hormone group (P<0.05). There was no visible difference in the incidence of adverse reactions between the two groups of patients after treatment (P>0.05). In conclusion, the combined application of Ginkgo biloba extract and hormones could effectively improve the abnormal hemorheological indexes of patients with sudden deafness and effectively relieve the imbalance of PBTCSs, which was safe. © 2022 Zhenhua Zhu et al.

摘要:
The collection and extraction of tongue images has always been an important part of intelligent tongue diagnosis. At present, the collection of tongue images generally needs to be completed in a sealed, stable light environment, which is not conducive to the promotion of extensive tongue image and intelligent tongue diagnosis. In response to the problem, a new algorithm named GCYTD (GELU-CA-YOLO Tongue Detection) is proposed to quickly detect and locate the tongue in a natural environment, which can greatly reduce the restriction of the tongue image collection environment. The algorithm is based on the YOLO (You Only Look Once) V4-tiny network model to detect the tongue. Firstly, the GELU (Gaussian Error Liner Units) activation function is integrated into the model to improve the training speed and reduce the number of model parameters; then, the CA (Coordinate Attention) mechanism is integrated into the model to enhance the detection precision and improve the failure tolerance of the model. Compared with the other classical algorithms, Experimental results show that GCYTD algorithm has a better performance on the tongue images of all types in terms of training speed, tongue detection speed and detection precision, etc. The lighter model can contribute on deploying the tongue detection model on small mobile terminals. © 2022 Tech Science Press. CuO/Fe3O4/GO, as a Z-scheme heterojunction catalyst, was successfully synthesized and used as a photocatalyst for removing tetracycline from aqueous solution. The CuO/Fe3O4/GO heterogeneous catalyst combines the narrow bandgap semiconductor CuO, oxygen vacancies of Fe3O4, and oxygen-containing reaction sites of GO. Without the addition of activators (persulfate or H2O2), the photocatalytic performance on decomposing tetracycline is very excellent. Compared with GO, Fe3O4, and CuO, CuO/Fe3O4/GO exhibits superior photocatalytic performance. Under visible light radiation, CuO/Fe3O4/GO generates h+ and ⋅O2−, which are the mainly responsible active groups for TC degradation. The effects of various pH, catalysts, and reuse on the degradation performance are evaluated, and the optimal conditions for CuO/Fe3O4/GO removal of tetracycline are obtained at pH 7, catalyst dosage 20 mg⋅L−1, TC at a concentration of 30 mg/L, nearly 97.3% of tetracycline is decomposed. This study has great potential in the treatment of wastewater containing various antibiotics. © 2022 Elsevier Ltd

Zearalenone (ZEN) was a toxic compound secreted by the metabolism of fungi that exerted negative influence over human and animal health due to its carcinogenicity, mutagenicity, genotoxicity, nephrotoxicity, immunotoxicity and teratogenicity. The superiority of biosensors was recognized which held a great potential for ZEN detection because they were less costly and much friendlier in preparation and operation steps but further sensitive, specific, rapid, and portable. This review was designed to pose a comprehensive interpretation on ZEN detected biosensors through highlighting the nanomaterials for fabricating the sensing elements and their applications in assorted read-out types of biosensors. Besides, novel nanomaterials-based signal amplification strategies utilized to accomplish this goal were overviewed and analyzed in detail. Current challenges and future perspectives were discussed as well. © 2021 The Authors

何首乌有生首乌和制首乌之分, 生首乌能解毒, 润肠通便; 制首乌能补肝肾, 益精血, 乌须发, 强筋骨, 化浊降脂。 生/制首乌都含有二苯乙烯苷类、 蒽醌类、 磷脂类等化学成分, 但其含量存在一定差异。 生首乌炮制后其毒性减弱。 生/制首乌的功效、 主治和毒性都存在一定的差异, 在外观性状上差异较易识别, 但其磨成粉末后两者不易区分。 因此, 寻找一种快速简单的判别方法来区分生/制首乌十分必要。 中红外光谱法具有检测速度快, 对样品无损等优势, 已在中药质量控制和鉴别中得到广泛的应用。 该研究建立生/制首乌中红外光谱的指纹图谱, 并采用正交偏最小二乘-判别分析对其进行鉴别。 在4 000~700 cm-1范围内采集38批不同来源生/制首乌中红外光谱数据, 分析其主要的特征峰, 并用simca13.0软件对数据进行正交偏最小二乘-判别分析。 建立38批不同来源生/制首乌中红外指纹图谱, 并分析出主要化学成分包括蛋白质类、 核酸类、 脂肪酸类、 蒽醌类、 二苯乙烯苷类、 磷脂类。 对红外光谱峰形和峰强度进行分析, 生首乌和制首乌的峰形差异比较少, 但峰强度具有一定的差异。 为了建立生/制首乌红外光谱差异模型, 采用正交偏最小二乘-判别分析法, 其结果能很好地将生/制首乌分成两类, 左边为制首乌, 右边为生首乌。 根据变量在项目中的重要性(VIP值)筛选差异化学成分, 并利用SPSS17.0统计软件进行t检验, 以VIP>1及p<0.05确定差异性化学成分。 生/制首乌的差异性化学成分为二苯乙烯苷类、 蒽醌类、 磷脂类, 表明何首乌炮制后二苯乙烯苷类、 蒽醌类、 磷脂类的含量都发生变化。 中红外光谱结合模式识别筛选出差异性化学成分和文献报道基本一致, 表明此法可用于生/制首乌鉴别。 本研究成功采用中红外光谱对生/制首乌进行快速检测和整体质量评价, 结合模式识别能将其鉴别, 可为中药质量控制和快速鉴别提供依据。

Postoperative bacterial infection greatly threatens prognosis of patients following tissue engineering implantation and surgical operation. In this study, a novel antibacterial tissue engineering porous scaffold has been prepared, by blending a newly synthesized berberine-oleanolic acid (HL-9) with collagen (COL I) via freeze-drying. It showed that the HL-9 delivery COL I scaffolds displayed a uniform porous three-dimensional structure, high porosity and good water absorption. These HL-9 delivery COL I porous scaffolds (HL-9/COL I scaffolds) are able to release HL-9 steadily for 15 days. HL-9/COL I scaffolds, which contains, 4% and 5% HL-9 respectively, exhibited a similar antibacterial activity against Gram-positive and Gram-negative bacteria. Hence, the HL-9/COL I scaffolds containing 3% HL-9 is used to test the biocompatibility. By employing Alamar Blue assays and FDA staining, the proliferation of MG-63 cultured with 3%HL-9/COL I scaffolds is similar with that cultured with COL I scaffolds. In conclusion, the 3%HL-9/COL I scaffolds showed a good biocompatibility and antibacterial activity, suggesting that it may be applied in tissue engineering in the future. © Published under licence by IOP Publishing Ltd.

The occurrence and development of hepatocellular carcinoma (HCC) is related to the level of trace elements. The mechanism of changes in trace element concentration and the identification of HCC risks associated with them are not clear. Therefore, an accurate and reliable approach for quantification of the level of trace elements in the serum of liver cancer patients is of great significance. In this study, we proposed a novel strategy for accurate determination of trace elements such as Mn, Fe, Ni, Cu, Zn, and Se in the serum of HCC patients based on inductively coupled plasma tandem mass spectrometry (ICP-MS/MS). In the MS/MS mode, N2O was used as the reaction gas, and an octopole reaction system (ORS) was employed to provide interference-free conditions and a sufficiently low limit of detection (LOD) for the determination of analytical elements. The internal standard elements with similar masses and similar mass spectrometry behavior were selected to correct the matrix effect. The LODs of Mn, Fe, Ni, Cu, Zn, and Se were as low as 0.013, 0.25, 0.010, 0.020, 0.17, and 0.008 mu g L-1, respectively. Bovine serum (SRM 1598a) was adopted as the standard reference material to evaluate the accuracy of the method, and the developed method was successfully applied to determine the content of Mn, Fe, Cu, Zn, Ni, Se in serum from HCC patients. The obtained results were in agreement with that of sector field ICP-MS (SF- ICP-MS). The contents of Fe and Cu in the serum of HCC patients were higher compared to that of healthy controls, while the contents of Zn and Se were lower than that of healthy controls. Furthermore, no significant differences were observed in the contents of Mn and Ni in the serum of HCC patients and healthy controls. (C) 2020 Elsevier B.V. All rights reserved.

This study used the method of review to analyze the economic, tourism, health and social impacts of the first three PHEIC outbreaks, so as to identify the impact of COVID-19 epidemic on China in 2020 and put forward policy suggestions in response to the challenges. This study's main research subjects are the 2009 H1N1 outbreak, 2014 Ebola outbreak in West Africa, and 2016 ZIKV outbreak. The literature retrieval of the three PHEIC impacts and the related grey literature were reviewed. The comprehensive analysis of this review indicates that COVID-19 outbreak in China may cause impacts on consumption, import and export, and manufacturing. For a short time, from the demand side and supply side, the COVID-19 affects tourism industry in China. However, if the government attaches considerable importance to the tourism industry, which will lead to the development of new digital technology such as online travel industry. It harms people's psychological health and physical health, and produces unhealthy psychological emotions such as stress, discrimination, fear and a series of physiological and drug sequelae. Meanwhile, it leads to the decline of social trust, the decrease of educational opportunities, the augment of employment pressure, the potential 'coaxing'of prices and other harms, thus causing a series of pressures on China's economy, tourism, health and society. It is urgent for Chinese government to put forward effective policies, which are aimed for short term and long term impact separately. While addressing the damage caused by COVID-19 outbreak in China, we should seize the opportunity to promote the reform and innovation of China's health system. © 2020 ACM.

For fabrication of composite electrode, one-pot strategy is highly attractive for convenience and efficiency. Here, a self-supporting Co 3 O 4 /nanoporous gold (NPG) composite electrode was one-pot prepared via one-step in situ anodization of a smooth gold electrode in a CoCl 2 solution within 100 s. It worked as a bifunctional electrocatalyst for glucose oxidation and H 2 O 2 reduction in NaOH solution. Under optimized conditions, the electrocatalytic oxidation of glucose exhibits a wide linear range from 2 μM to 2.11 mM with a limit of detection as low as 0.085 μM (S/N = 3) and an ultrahigh sensitivity of 4470.4 μA mM −1 cm −2 . Detection of glucose in human serum samples are also realized with results comparable to those from local hospital. The electrocatalytic reduction of H 2 O 2 shows a linear response range from 20 μM to 19.1 mM and a high sensitivity of 1338.7 μA mM −1 cm −2 . The present results demonstrate that the facilely prepared Co 3 O 4 /NPG is a promising nonenzymatic sensor for rapid amperometric detection of glucose and H 2 O 2 with ultrasensitivity, high selectivity, satisfactory reproducibility, good stability and long duration. © 2019 Elsevier B.V.

The gold standard for diagnosing pulmonary Mycobacterium tuberculosis (TB) is the detection of tubercle bacillus in patient sputum samples. However, current methods either require long waiting times to culture the bacteria or have a risk of getting false-positive results due to cross-contamination. In this study, a method to detect tubercle bacillus based on the molecular typing technique is presented. This method can detect genetic units, variable number of tandem repeat (VNTR), which are the characteristic of tuberculosis (TB), and performs quality control using a mathematical model, ensuring the reliability of the results. Compared to other methods, the proposed method was able to process and diagnose a large volume of samples in a run time of six hours, with high sensitivity and specificity. Our method is also in the pipeline for implementation in clinical testing. Reliable and confirmed results are stored into a database, and these data are used to further refine the model. As the volume of data processed from reliable samples increases, the diagnostic power of the model improves. In addition to improving the quality control scheme, the collected data can be also used to support other TB research, such as that regarding the evolution of the tubercle bacillus. © 2019 Tienan Feng et al.

Human amniotic membrane (AM) offers unique advantages as a matrix to support the transplantation of limbal stem cells (LSCs) due to its inherent pro-regenerative and anti-inflammatory properties. However, the widespread use of AM in clinical treatments of ocular surface disorders is limited by its weak mechanical strength and fast degradation, and high cost associated with preserving freshly isolated AM. Here we constructed a composite membrane consisting of an electrospun bioabsorbable poly(ε-caprolactone) (PCL) nanofiber mesh to significantly improve the ultimate tensile strength, toughness, and suture retention strength by 4–10-fold in comparison with decellularized AM sheet. The composite membrane showed extended stability and conferred longer-lasting coverage on wounded cornea surface compared with dAM. The composite membrane maintained the pro-regenerative and immunomodulatory properties of dAM, promoted LSC survival, retention, and organization, improved re-epithelialization of the defect area, and reduced inflammation and neovascularization. This study demonstrates the translational potential of our composite membrane for stem cell-based treatment of ocular surface damage. Statement of Significance: Human decellularized amniotic membrane (dAM) has been widely shown as a biodegradable and bioactive matrix for regenerative tissue repair. However, the weak mechanical property has limited its widespread use in the clinic. Here we constructed a composite membrane using a layer of electrospun poly(ε-caprolactone) (PCL) nanofiber mesh to reinforce the dAM sheet through covalent interfacial bonding, while retaining the unique bioactivity of dAM. In a rabbit model of limbal stem cell (LSC) deficiency induced by alkaline burn, we demonstrated the superior property of this PCL-dAM composite membrane for repairing damaged cornea through promoting LSC transplantation, improving re-epithelialization, and reducing inflammation and neovascularization. This new composite membrane offers great translational potential in supporting stem cell-based treatment of ocular surface damage. © 2019 Acta Materialia Inc.

Using one-step anodization strategy, a nanoporous gold film (HNPG) with large surface area was rapidly fabricated on Au80Sn20 (wt%) alloy in just 80 s. The formation of highly surface-roughened nanoporous structures results from a complex process of electrochemical dealloying of Sn component from AuSn alloy, anodic electrodissolution, disproportion and deposition of Au component, and spontaneous redox reaction between electrodissolved Sn2+ and AuCl4 -species at the applied anodic potential. As-prepared HNPG/AuSn shows enhanced electrochemical performance for glucose oxidation in alkaline electrolyte. At a low potential of 0.1 V (vs. SCE), it offers a short response time of 4 s, a wide linear detection range of 2 μM to 8.11 mM, an ultralow detection limit of 0.36 μM (S/N = 3), an ultrahigh sensitivity of 4374.6 μA cm−2 mM−1, and satisfactory selectivity and reproducibility. Specifically, after 6 weeks, no obvious loss of glucose amperometric signal was observed on HNPG/AuSn. The facile preparation and excellent sensing performance of HNPG/AuSn electrode make sure that it is a promising candidate for advanced enzyme-free glucose sensors. © 2018 Elsevier B.V.

With the economic development, more and more people participate in medical insurance and enjoy the benefits of medical insurance. However, medical insurance fraud has brought tremendous losses to the medical insurance fund. This paper presents a two-stage outlier detection method for medical insurance fraud detection. In the first stage, weighted k-means algorithm is used to cluster the data set and prune the result set, where the weight w is calculated by using the particle swarm optimization algorithm to minimize the evaluation function of the weight index. The second stage adopts the improved outlier detection method to process the result set. Experiments show that the accuracy of this method is higher than that of using K-means algorithm or LOF algorithm alone. 设计新型的DNA探针,其主要功能区域包含适配体区域和分子信标互补区域;当无靶标存在时,羧基荧光素(FAM)的荧光被黑洞猝灭基团(BHQ)猝灭,加入靶标后,Ag~+与适配体区域特异性结合,使得分子信标与其互补区域发生结合,加入EXO III后,分子信标被酶解,从而释放Ag~+/DNA探针复合物用于下一轮反应,同时产生大量的FAM,因此,可以通过荧光的增强来定量检测靶标的含量.评价了该方法的灵敏度和选择性,探究它在实际样品中的表现能力.结果表明,适配体区域与分子信标间隔2个碱基的DNA探针与分子信标结合效果最好,检测的信噪比最大.该方法对Ag~+的检测最低下限(LOD)为0.1nmol/L,在0.5~200nmol/L范围内,荧光值与靶标浓度成线性关系(R~2 = 0.994).对实际样品的检测在20~200nmOl/L范围内呈线性关系(R~2=0.998),且能很好的从混合样品中区分10nmol/L Ag~+.该研究建立的快速检测Ag~+方法具有良好的线性定量能力和操作性能,且灵敏度高、特异性强,可满足现实的需求.

摘要:
两亲性聚合物纳米颗粒作为疏水性抗肿瘤药物载体因其能够增强化疗效率并降低毒副作用而受到广泛关注.采用双乳液溶剂挥发法制备了聚(3-羟基丁酸酯-co-3-羟基戊酸酯)(PHBV)/葡聚糖纳米颗粒,测得平均粒径为$205.0 \pm 6.9\;{\rm{nm}}$,Zeta电势为$ - 1.59 \pm 0.12\;{\rm{mV}}$,纳米颗粒具有明显的壳核结构,粒径均一,分散性良好.将疏水性化疗药物顺铂包载后,其粒径及电势均无明显变化,载药量达$19.3 \pm 2.9\% $.顺铂在模拟肿瘤细胞环境${\rm{pH}} = 5.5$的磷酸盐缓冲液(PBS)中比正常细胞环境${\rm{pH}} = 7.4$时释放更快,且累计释放周期均长达7d以上,表明该药物载体具有一定的pH响应性以及优异的缓释性能.细胞集落形成实验表明PHBV/葡聚糖纳米药物载体具有良好的生物相容性,而载药纳米颗粒对肿瘤细胞的毒性明显高于正常细胞,表明该纳米颗粒对肿瘤细胞具有更强的杀伤作用.综上所述, PHBV/葡聚糖纳米颗粒具有两亲性分子结构,合适的粒径及Zeta电势,显著的缓释效果,对肿瘤细胞具有pH响应性及更强的杀伤作用等优势,有望成为一种新型纳米药物载体,在癌症化疗中显著提高药物利用率并降低毒副作用.
