Brain plasticity refers to the brain's ability to change and adapt the environment and experience when the nervous system is damaged. Acupuncture can relieve the relevant pathological status due to stroke by enhancing brain plasticity. Specifically, acupuncture may finally achieve the balance between excitatory and inhibitory neurotransmitters by inhibiting the expression of neurotransmitter GABA, neurotrophic factor BDNF and proteins related to synaptic plasticity. This article analyzed and summarized that taking advantage of nervous plasticity the acupuncture could regenerate nervous cells and restore the related regulation of the movement by upper motor neurons, so as to relieving limb spasticity. It also summarized the research and application of acupuncture in regulating related signals, providing a systematic thought for the future study on acupuncture in the treatment of stroke induced limb.收起

Objective:To observe the changes in the neurological score, infarct size, and autophagy-and apoptosis-related indicators following the acupuncture treatment of rats with cerebral ischemia-reperfusion injury(CIRI)in order to investigate the effects of acupuncture on autophagy and apoptosis in the hippocampal tissue of CIRI rats Methods:In this study,170 Sprague-Dawley rats were randomly assigned to the normal group(n=34),sham group(n=34),and model establishment group(n=102).A CIRI rat model was established in the model establishment group. The rats of the model establishment group were then randomly and equally allocated to the middle cerebral artery occlusion/reperfusion group(MCAO/R group),acupuncture group(AC group),and edaravone group(ED group)(n=34 each). Each group was further subdivided into the 24-h and 72-h subgroups(n=17 each). The rats of the MCAO/R group were immobilized for 30 min and administered with an intraperitoneal injection of 0.9%sodium chloride solution. The rats of the AC group were immobilized, and they received acupuncture at the"Dazhui(GV 14)","Shuigou(GV 26)",and"Baihui(GV 20)"and an intraperitoneal injection of 0.9%sodium chloride solution, with the acupuncture needles left in place for 30 min.The rats of the ED group were immobilized for 30 min and administered with an intraperitoneal injection of edaravone(5 mg/kg).All interventions were performed once every 12 h.Investigations of the neurological score, infarct size using triphenyl tetra-zolium chloride staining, hippocampal Caspase-3 protein expression by immunohistochemical assay,hip-pocampal Beclin-1 expression by Western blotting, autophagosomes in the hippocampus by transmission electron microscopy, and apoptotic rate in the brain tissue of the ischemic side using the terminal de-oxynucleotidyl transferase dUTP nick end labeling assay were performed both before and after the inter-ventions.Results:Before intervention, the neurological score of the model establishment group was significantly lower than that of the normal and sham groups(P<0.01).After 24 h and 72 h,the neurological scores of the ED and AC groups were significantly increased(P<0.01),whereas the infarct size ratio were smaller(P<0.05 or P<0.01)and the apoptotic rates and Caspase-3 expression were lower(P<0.01)when com-pared with the MCAO/R group. The hippocampal Beclin-1 expression in the ED and AC groups was downregulated after 24 h of intervention(P<0.05)but upregulated after 72 h of intervention(P<0.05 and P<0.01,respectively).In the ED and AC groups,72 h of intervention led to significant increases in the neurological score(P<0.01)and significant reductions in the apoptotic rate and Caspase-3 expres-sion(P<0.05 or P<0.01),but no significant changes in the Beclin-1 expression(P>0.05)when compared with 24 h of intervention. Moreover, the AC group showed a significant decrease in the infarct size ratio(P<0.01).Electron microscopy revealed that after 24 h of intervention, only individual lysosome struc-tures could be observed in the tissues of the AC and ED groups. After 72 h of intervention,co-existence of autophagosomes and lysosomes was observed in the ED group, whereas co-existence of lysosomes and autolysosomes was observed in the AC group. Conclusion:Acupuncture significantly improved the neurological score and reduced the infarct size in the CIRI rats. The mechanisms of these effects could be attributed to the regulation of autophagy and inhibition of apoptosis in the hippocampal tissue as well as the duration of the intervention to a certain extent.

Objective: To observe the effect of acupuncture on local skin tissue metabolites of Tianshu(ST25) in rats with Crohn’s disease(CD) using metabolomics and to explore the mechanism of acupuncture treatment metabolites and pathway on local skin tissue of Tianshu(ST25).Materials and Methods: Thirty-eight Sprague-Dawley rats were randomly assigned to a normal group(n = 14) and a CD modeling group(n = 24). Rats in the CD modeling group were administered with 2,4,6-trinitrobenzene sulfonic acid and ethanol enema, in addition to the normal group. Four normal rats and four CD modeling rats were selected at random after model identification was established. Furthermore,CD modeling group rats were randomly assigned to two groups of 10 rats: the model group and acupuncture group. Rats in the acupuncture group underwent acupuncture of Tianshu(ST25) and Shangjuxu(ST37) for 7 days. After the intervention, the colon tissue was collected from each group of rats and the pathological changes were observed through hematoxylin and eosin(HE) staining. The local skin tissues at Tianshu(ST25) of the rats in each group were taken, and the proton nuclear magnetic resonance technique was used to detect the metabonomics of the local skin tissues of Tianshu(ST25) in rats. Results: Compared with the normal group, the following were observed in the model group after HE staining: irregular colon morphology and other pathological changes such as intestinal mucosal hyperemia, edema, ulcers, polyps, and inflammatory cell infiltration. In the acupuncture group, colon tissue structure was relatively complete and layered, the colon gland structure was restored, and inflammatory cell infiltration was significantly improved. The choline, glycerin, glycine, guanidoacetic acid, and proline levels were significantly higher in the model group than in the normal group(P < 0.05 or P < 0.01). Compared with the model group, the acupuncture group had significantly increased contents of alanine, leucine, L-phenylalanine, and tyrosine in the skin(P < 0.05 or P < 0.01).Valine, leucine, and L-isoleucine biosynthesis and L-phenylalanine metabolism were the main metabolic pathways involved in the changes in the local skin tissues of the rats in each group, biosynthesis of tyrosine, and tryptophan. Conclusion: The change in the metabolites in the local area of Tianshu(ST25) may be related to intestinal disease. The acupuncture of Tianshu(ST25) can improve intestinal inflammatory reaction in rats with CD, and this finding may be related to the regulation of amino acid metabolites and their pathways in the local acupoint tissues of Tianshu(ST25) by acupuncture.
