摘要:
婴幼儿谷类辅助食品是婴幼儿营养的重要能量来源, 其主要原料为大米, 而水稻极易从其生长的土壤和水中吸收重金属Cd积累到谷粒中, 导致大米中Cd的残留浓度相对较高, 从而对婴幼儿谷类辅助食品构成潜在威胁。 提出了电感耦合等离子体串联质谱(ICP-MS/MS)测定婴幼儿谷类辅助食品中重金属Cd的新策略。 样品经微波消解处理后, 采用ICP-MS/MS进行测定。 婴幼儿谷类辅助食品中Cd的测定所面临的质谱干扰主要来源于高浓度Mo所形成的Mo基多原子干扰离子(MoO+, MoN+, MoC+)以及Sn所形成的同量异位素(112Sn+, 114Sn+, 116Sn+)。 针对这些干扰, 在MS/MS模式下, 分别采用H2, NH3/He和O2为反应气, 考察不同Cd同位素的质谱干扰消除效果和分析灵敏度。 结果表明, 在H2和NH3/He反应模式下, H2和NH3均能与Mo基干扰离子发生质量转移反应, 而不与Sn+反应, 可选择110Cd+, 111Cd+和113Cd+进行测定, 但H2与Mo基干扰离子反应速度较慢, 难以彻底消除高浓度Mo基质中对Cd的质谱干扰; 在O2反应模式下, 所有干扰离子均能与O2发生质量转移反应, 虽然获得的分析灵敏度略低于NH3/He反应模式, 但背景等效浓度(BEC)明显比NH3/He反应模式低。 确定了以O2为反应气消除干扰测定Cd的最优分析方案。 利用标准参考物质评价了方法的准确性, 采用国标法(GB5009.268—2016)进行对比分析验证了方法的可靠性。 结果表明, Cd的LOD为2.03~13.4 ng·L-1, 标准参考物质的测定值与认定值基本一致, 在95%的置信水平, 对比分析结果之间无显著性差异。 方法的灵敏度高, 结果准确可靠, 适用于大批量婴幼儿谷类辅助食品中重金属Cd的高通量测定。

摘要:
植物精油是从芳香植物提取的天然复杂化合物, 作为芳香植物的次生代谢产物具有挥发性和浓郁香味, 其特有的多样化生物活性广泛应用于医药和化妆品行业。 植物精油具有高渗透性, 能以活跃的分子态渗透皮肤组织, 经淋巴腺吸收后进入血液, 其所含的重金属元素也极易随植物精油进入人体对健康构成潜在威胁。 采用硝酸-双氧水对植物精油进行微波消解, 在多模式样品导入系统(MSIS)的双重模式下, 采用电感耦合等离子体发射光谱(ICP-OES)测定其中可形成蒸气重金属元素As, Sn, Sb, Hg和不可形成蒸气重金属元素Cr, Ni, Cd和Pb的含量。 选择盐酸对样品进行酸化并预还原氧化态元素, 通过在线加入L-半胱氨酸/酒石酸提高可形成蒸气元素的蒸气发生效率, 利用硼氢化钠/氢氧化钠在MSIS中将As, Sn, Sb和Hg转变为蒸气状态; 针对分析过程中存在的多个或单个光谱重叠和背景干扰, 分别对空白溶液、 分析元素和预期干扰元素的纯溶液进行测定, 根据获得的光谱响应数据解卷积构建快速自动曲线拟合技术(FACT)模型, 将分析谱线从干扰谱线中分离出来, 从而实现光谱重叠干扰和背景干扰的实时校正; 采用加标回收并与电感耦合等离子体质谱(ICP-MS)进行对比分析评价方法的准确性。 各元素方法的检出限(MDL)为0.3811.2 μg·kg-1, 加标回收率为95.4%104%, 相对标准偏差(RSD)为1.9%4.9%, 对比分析的相对误差(RE)在-2.1%2.7%之间, 表明方法准确可靠, 精密度高。 对8种植物精油中的重金属元素进行了分析, 所有植物精油样品中重金属元素As, Hg和Pb的含量远低于GB/T 26516-2011制定的限量标准, 植物精油中重金属元素Cr, Ni, Sn, Cd和Sb的含量虽然没有制定限量标准, 但均处于极低水平。 MSIS兼具传统雾化和蒸气发生双重功能, 在分析可形成蒸气元素和不可形成蒸气元素时无需切换不同进样系统, 能满足大批量植物精油中微量重金属元素的高通量分析需要。

摘要:
CuO/Fe3O4/GO, as a Z-scheme heterojunction catalyst, was successfully synthesized and used as a photocatalyst for removing tetracycline from aqueous solution. The CuO/Fe3O4/GO heterogeneous catalyst combines the narrow bandgap semiconductor CuO, oxygen vacancies of Fe3O4, and oxygen-containing reaction sites of GO. Without the addition of activators (persulfate or H2O2), the photocatalytic performance on decomposing tetracycline is very excellent. Compared with GO, Fe3O4, and CuO, CuO/Fe3O4/GO exhibits superior photocatalytic performance. Under visible light radiation, CuO/Fe3O4/GO generates h+ and ⋅O2−, which are the mainly responsible active groups for TC degradation. The effects of various pH, catalysts, and reuse on the degradation performance are evaluated, and the optimal conditions for CuO/Fe3O4/GO removal of tetracycline are obtained at pH 7, catalyst dosage 20 mg⋅L−1, TC at a concentration of 30 mg/L, nearly 97.3% of tetracycline is decomposed. This study has great potential in the treatment of wastewater containing various antibiotics. © 2022 Elsevier Ltd

何首乌有生首乌和制首乌之分, 生首乌能解毒, 润肠通便; 制首乌能补肝肾, 益精血, 乌须发, 强筋骨, 化浊降脂。 生/制首乌都含有二苯乙烯苷类、 蒽醌类、 磷脂类等化学成分, 但其含量存在一定差异。 生首乌炮制后其毒性减弱。 生/制首乌的功效、 主治和毒性都存在一定的差异, 在外观性状上差异较易识别, 但其磨成粉末后两者不易区分。 因此, 寻找一种快速简单的判别方法来区分生/制首乌十分必要。 中红外光谱法具有检测速度快, 对样品无损等优势, 已在中药质量控制和鉴别中得到广泛的应用。 该研究建立生/制首乌中红外光谱的指纹图谱, 并采用正交偏最小二乘-判别分析对其进行鉴别。 在4 000~700 cm-1范围内采集38批不同来源生/制首乌中红外光谱数据, 分析其主要的特征峰, 并用simca13.0软件对数据进行正交偏最小二乘-判别分析。 建立38批不同来源生/制首乌中红外指纹图谱, 并分析出主要化学成分包括蛋白质类、 核酸类、 脂肪酸类、 蒽醌类、 二苯乙烯苷类、 磷脂类。 对红外光谱峰形和峰强度进行分析, 生首乌和制首乌的峰形差异比较少, 但峰强度具有一定的差异。 为了建立生/制首乌红外光谱差异模型, 采用正交偏最小二乘-判别分析法, 其结果能很好地将生/制首乌分成两类, 左边为制首乌, 右边为生首乌。 根据变量在项目中的重要性(VIP值)筛选差异化学成分, 并利用SPSS17.0统计软件进行t检验, 以VIP>1及p<0.05确定差异性化学成分。 生/制首乌的差异性化学成分为二苯乙烯苷类、 蒽醌类、 磷脂类, 表明何首乌炮制后二苯乙烯苷类、 蒽醌类、 磷脂类的含量都发生变化。 中红外光谱结合模式识别筛选出差异性化学成分和文献报道基本一致, 表明此法可用于生/制首乌鉴别。 本研究成功采用中红外光谱对生/制首乌进行快速检测和整体质量评价, 结合模式识别能将其鉴别, 可为中药质量控制和快速鉴别提供依据。

工业化和现代化进程的加快消耗了大量能源, 对能源的高度依赖性导致了全球化石能源需求的快速增长, 随着非再生化石能源的日渐枯竭, 迫切需要大力发展可再生能源以调整现有能源结构。 作为国际上研究最多的生物柴油, 麻疯树油是国内外公认的最有可能替代化石能源的再生能源, 具有极大的开发潜力。 麻疯树油中的微量元素在燃烧过程中会影响发动机的性能, 并在尾气排放过程中决定了对环境所造成的污染程度。 本文以获得麻疯树油中多元素的含量为目的, 建立应用电感耦合等离子体串联质谱(ICP-MS/MS)法准确测定麻疯树油中低水平Na, Si, P, S, Cl, K, Ti, V, As含量的分析方法。 采用微波密闭消解系统, 依次向麻疯树油样品中加入硝酸和双氧水进行消解。 详细地研究了各待测元素在不同分析模式下检出限(DL)和背景等效浓度(BEC)的变化情况, 在MS/MS模式下, 通过向碰撞/反应池(CRC)中加入反应气可以完全消除质谱干扰。 选择O2为反应气, P+, S+, Ti+, V+, As+与O2的反应均为放热过程, 能发生质量转移自发生成PO+, SO+, TiO+, VO+, AsO+, 利用O2质量转移法消除质谱干扰; 选择H2为反应气, Cl+与H2反应能自发生成ClH+2, 利用H2质量转移法消除质谱干扰, 而Na+, Si+, K+均不能与H2发生质量转移反应, 利用H2原位质量法消除质谱干扰。 选择Sc为内标元素校正了分析过程中的基体效应。 通过考察不同反应气流速下各元素的BEC变化, 优化了反应气流速, O2的最佳流速为0.45 mL·min-1, H2的最佳流速为7.5 mL·min-1。 在优化的实验条件下获得Na, Si, P, S, Cl, K, Ti, V, As的检出限分别为6.41, 37.3, 24.6, 118, 530, 7.96, 7.61, 0.34, 3.20 ng·L-1, 各元素在0~50 μg·L-1范围内的线性相关系数(R2)≥0.999 8, 方法具有良好的线性关系。 采用三水平加标回收实验来验证方法的准确性和精密度, 所有元素的加标回收率在91.2%~108%之间, 相对标准偏差(RSD)为1.9%~4.6%, 表明所建立的方法准确性好, 精密度高。 通过对来自中国不同地区的4个麻疯树油样品进行测定, 结果显示, 4个麻疯树油样品中P含量≤164 ng·g-1, S含量≤2310 ng·g-1, 碱(Na+K)含量≤1 690 ng·g-1, 三项指标均达到了中国生物柴油调和燃料国家标准, 欧Ⅳ生物柴油标准, 德国生物柴油标准和美国生物柴油标准。 这项研究为麻疯树油中多种微量元素的准确分析提供了一种方便可行的新方法, 为麻疯树油的质量控制和安全应用提供了科学的理论依据。

摘要:
Despite chemotherapy has been widely used for tumor therapy, the serious side effect is still a major challenge. Recently, two dimensional nanomaterial-based drug delivery systems have attracted wide concern due to their high drug loading and low side effect. In addition, some kinds of nanomaterials can directly act as a photosensitizer to induce cancer destruction. In this study, we developed a drug delivery system of mixture of high/low molecular weight branched polyethylenimine-polyethylene glycol-reduced graphene oxide (mBPEI-PEG-rGO) using reduced graphene oxide as matrix. A model drug of doxorubicin (DOX) was loaded on the nanocomposites with the efficiency of 81% and the release rate of more than 50% at acidic environment. In vitro experiments indicated that mBPEI-PEG-rGO-DOX with enhanced stability and biocompatibility efficiently delivered and released DOX into cells mainly through micropinocytosis and killed SMMC-7721 cells by inducing reactive oxygen species (ROS) and cell apoptosis. Furthermore, in vivo experiments indicated that the combination of intratumoral injection of mBPEI-PEG-rGO-DOX and local laser irradiation nearly ablated hepatocarcinoma. In conclusion, this new drug delivery system provided an alternative for combinational photothermal and chemotherapy against hepatocarcinoma.

摘要:
Simple, fast and effective detection methods of Escherichia coli (E. coli) are essential for human life and health. This article proposes a sensitive and rapid detection method of E. coli based on electrochemical biosensor using 16 S rDNA as target biomarker, and using oligonucleotide probes and nanogap network electrodes as transducer elements. The nanogap network electrodes are prepared by interconnecting of gold nanoparticles through thiolated peptide nucleic acid (PNA) probes on the substrate of Au interdigital electrode. In the presence of specific 16 S rDNA fragments, PNA capture probe hybridized to 5′ terminal of the fragments, and detection probe modified with horseradish peroxidase (HRP) hybridized to its 3′ terminal. HRP attached to the hybridized detection probe catalyzed the polymerization of aniline along target chain. Conductive connection was caused by polyaniline deposition between the interrupted gold nanoparticles and offered a conductimetric response of constructed sensor. Thus, E. coli could be successfully detected with the detection limit of 100 CFU/mL. The detection time was less than 3 h. It would be widely used for rapid detection of E. coli. © 2018 Elsevier B.V.

As an essential phosphate group hydrolase, alkaline phosphatase (ALP), whose level in serum is correlated with bone disease, liver dysfunction, and cancer, could be used as a biomarker for clinical diagnosis and biomedical studies. Hence, developing a convenient and sensitive method for ALP assay has importance in disease diagnosis, drug treatment, and prognosis assessment. In this work, using a hairpin DNA strand as the substrate, we developed an ultrasensitive and simple fluorescence method for quantitative ALP assay based on the binding difference of reduced graphene oxide (rGO) with different DNA strands coupled with λ exonuclease (λ exo) cleavage. Under the optimal conditions, the limit of detection (LOD) of ALP is estimated to be 0.01 U/L with the linear region from 0.5 U/L to 70 U/L. Furthermore, the proposed assay was used to detect ALP in complicated cell-free extracts and evaluate the inhibitory effects of two well-known inhibitors of ALP activity. Finally, the method was used to investigate the effect of natural compounds on ALP activity and five compounds with different inhibitory capability were screened. In summary, we propose that the new method for ALP assay can be applied for therapeutic drug monitoring (TDM) and high-throughput compound screening in combination with multiwell plate technology.

摘要:
The aim of this study was to construct recombinant adenovirus vector carrying a short hairpin RNA (shRNA) that can exclusively target cyclooxygenase-2 (COX-2) gene expression and observe its inhibitory effect on hepatic fibrosis. We designed and synthesized three oligonucleotide sequences, and cloned those into a shuttle vector, pYr-1.1-hU6-EGFP, after annealing. The restriction-enzyme digestion and sequencing analyses confirmed that the constructed recombinant eukaryotic expression vector was correct. A recombination reaction using LR Clonase was performed for the pYr-1.1-hU6-EGFP COX-2shRNA and the adenovirus vector pAd/BL-DEST to form Ad-COX-2shRNA. The adenoviruses containing the recombinant plasmids were transfected into hepatic stellate cells (HSCs). The transfection efficiency of the three COX-2 shRNAs exceeded 70%. Reverse transcription PCR (RT-PCR) and western blot confirmed that the target gene expression was decreased at the level of mRNA and protein, and the interference effect of COX-2 shRNA-1 was better than that of the other two COX-2 shRNAs. COX-2 shRNA-1 recombinant adenovirus vectors (1 ×10<sup>9</sup>PFU/mL) were injected via the tail vein into rats fed a high-fat diet with a 40% carbon tetrachloride peanut oil lavage, which induced liver fibrosis. Rats were euthanized at the end of the 12th week, and their liver was removed. Liver expression of COX-2 mRNA and protein was detected by RT-PCR and immunohistochemistry, respectively. RT-PCR and immunohistochemistry showed that COX-2 shRNA-1 inhibited COX-2 expression in liver tissue. Hematoxylin/eosin and Masson staining showed that COX-2 shRNA-1 ameliorated the severity of liver fibrosis. The COX-2 shRNA eukaryotic expression vectors were constructed successfully and COX-2 shRNA-1 ameliorated liver fibrosis.<br/> ©2018 The Author(s). In addition to being an important object in theoretical and experimental studies in enzymology, RNase A also plays an important role in the development of many kinds of diseases by regulating various physiological or pathological processes, including cell growth, proliferation, differentiation, and invasion. Thus, it can be used as a useful biomarker for disease theranostics. Here, a simple, sensitive, and low-cost assay for RNase A was constructed by combining a fluorogenic substrate with reduced graphene oxide (rGO). The method with detection limit of 0.05 ng/mL was first applied for RNase A targeted drug screening, and 14 natural compounds were identified as activators of this enzyme. Then, it was applied to detect the effect of drug treatment and Hepatitis B virus (HBV) infection on RNase A activity. The results indicated that RNase A level in tumor cells was upregulated by G-10 and Chikusetsusaponin V in a concentration-dependent manner, while the average level of RNase A in the HBV infection group was significantly inhibited compared with that in the control group. Furthermore, the concentration-dependent inhibitory effect of heavy metal ions on RNase A was observed using the method and the results indicated that Ba<sup>2+</sup>, Co<sup>2+</sup>, Pb<sup>2+</sup>, As<sup>3+</sup>, and Cu<sup>2+</sup>inhibited RNase A activity with IC<inf>50</inf>values of 93.7 μM (Ba<sup>2+</sup>), 90.9 μM (Co<sup>2+</sup>), 110.6 μM (Pb<sup>2+</sup>), 171.5 μM (As<sup>3+</sup>), and 165.1 μM (Cu<sup>2+</sup>), respectively. In summary, considering the benefits of rapidity and high sensitivity, the method is practicable for RNase A assay in biosamples and natural compounds screening in vitro and in vivo.<br/> ©2018 American Chemical Society.

摘要:
An efficient glucosylation of alcoholic hydroxyls in the structures of protostane triterpenoids catalyzed by fungus has been developed. Four protostanes 1–4 as the substrates have been transformed by Syncephalastrum racemosum AS 3.264 and generated corresponding 11-OH glucosylation derivatives. The regioselective characteristics were also identified by various highly oxygenated protostane substrates. The time-courses displayed the highest yields of 1a, 3a, and 4a at > 50%, and 1a had the highest yield of 72%. All of the protostane analogues displayed hCE-2 inhibitory effects in an in vitro bioassay. This report describes a rare biocatalytic alcoholic hydroxyl glucosylation with considerable yield and regioselective characteristics.

Isoflavones are natural substances that exhibit hormone-like pharmacological activities. The separation of isoflavones remains an analytical challenge because of their similar structures. We show that ultra-high performance supercritical fluid chromatography can be an appropriate tool to achieve the fast separation of 12 common dietary isoflavones. Among the five tested columns the Torus DEA column was found to be the most effective column for the separation of these isoflavones. The impact of individual parameters on the retention time and separation factor was evaluated. These parameters were optimized to develop a simple, rapid, and green method for the separation of the 12 target analytes. It only took 12.91 min using gradient elution with methanol as an organic modifier and formic acid as an additive. These isoflavones were determined with limit of quantitation ranging from 0.10 to 0.50 μg/mL, which was sufficient for reliable determination of various matrixes. ©2017 WILEY-VCH Verlag GmbH & Co. Bond coats play a significant role in manipulating the stability of 8YSZ thermal barrier coatings (TBCs) deposited on Mg alloy. In this study, the metal of Ni-Al mixture was sprayed as bond coat via reaction plasma spraying (RPS). Besides, Intermetallic compound NiAl was prepared by self-propagating high-temperature synthesis (SHS). The pure intermetallic compound NiAl was sprayed as bond coat via atmospheric plasma spraying (APS). The microstructure, composition, bond stability and corrosion resistance of the produced bond coats were investigated to find the optimal bond coat for the TBCs on Mg alloy. Results indicate that the bond coats composed of NiAl show excellent bond stability. High toughness of the RPS Ni-Al bond coat results in high tensile bond strength. The APS NiAl bond coat leads to outstanding thermal shock resistance mainly attributing to its moderate thermal expansion match, good mechanical properties and excellent corrosion resistance. As a highly conserved damage repair protein, RNase H can specifically hydrolyze RNA in DNA-RNA chimeric strands. DNAzyme, a synthetic single-stranded DNA consisting of binding and catalytic sites, can cleave RNA in the presence of cofactors. In this study, we establish a highly sensitive RNase H assay assisted with DNAzyme's cleavage property. A DNA-RNA chimeric strand, which contains DNAzyme sequences, is used as the hydrolysis substrate of RNase H. The RNase H hydrolysis of the chimeric substrate results in the release of DNAzyme. Subsegment DNAzyme digest, a molecular beacon, causes a "turn-on" fluorescence signal by disrupting its hairpin structure. Furthermore, the fluorescence signal is amplified by cyclic digestion of DNAzyme to the substrate of molecular beacon. Under the optimal conditions, the detection limit of RNase H is 0.01 U/mL, which is superior to those of several alternative approaches. Additionally, the method was further used for RNase H detection in heterogeneous biological samples as well as to investigate the effects of natural compounds on this enzyme. In summary, these results show that the method not only provides a universal platform for monitoring RNase H activity but also shows great potential in biomedical studies and drug screening. As a highly conserved damage repair protein, RNase H can hydrolysis DNA-RNA heteroduplex endonucleolytically and cleave RNA-DNA junctions as well. In this study, we have developed an accurate and sensitive RNase H assay based on fluorophore-labeled chimeric substrate hydrolysis and the differential affinity of graphene oxide on RNA strand with different length. This end-point measurement method can detect RNase H in a range of 0.01 to 1 units /mL with a detection limit of 5.0×10<sup>-3</sup>units/ mL under optimal conditions. We demonstrate the utility of the assay by screening antibiotics, resulting in the identification of gentamycin, streptomycin and kanamycin as inhibitors with IC<inf>50</inf>of 60±5 µM, 70±8 µM and 300±20 µM, respectively. Furthermore, the assay was reliably used to detect RNase H in complicated biosamples and found that RNase H activity in tumor cells was inhibited by gentamycin and streptomycin sulfate in a concentration-dependent manner. The average level of RNase H in serums of HBV infection group was similar to that of control group. In summary, the assay provides an alternative tool for biochemical analysis for this enzyme and indicates the feasibility of high throughput screening inhibitors of RNase H in vitro and in vivo. ©2016 Elsevier B.V.

The tender leaves of Rubus corchorifolius L. f. have been consumed as tea for drinking in China since ancient times. In this study, a novel ent-kaurane diterpenoid was isolated and identified from R. corchorifolius L. f. leaves as ent-kaur-2-one-16β,17-dihydroxy-acetone-ketal (DEK). DEK suppressed the growth of HCT116 human colon cancer cells with an IC<inf>50</inf>value of 40 ±0.21 μM, while it did not cause significant growth inhibition on CCD-18Co human colonic myofibroblasts at up to100 μM. Moreover, DEK induced extensive apoptosis and S phase cell cycle arrest in the colon cancer cells. Accordingly, DEK caused profound effects on multiple signaling proteins associated with cell proliferation, cell death, and inflammation. DEK significantly upregulated the expression levels of pro-apoptotic proteins such as cleaved caspase-3, cleaved caspase-9, cleaved PARP, p53, Bax, and tumor suppressor p21<sup>Cip1/Waf1</sup>, downregulated the levels of cell cycle regulating proteins such as cyclinD1, CDK2, and CDK4 and carcinogenic proteins such as EGFR and COX-2, and suppressed the activation of Akt. Overall, our results provide a basis for using DEK as a potential chemopreventive agent against colon carcinogenesis. ©2017 American Chemical Society.

The leaves of Rubus corchorifolius L. f. have been consumed as a herbal tea for a long time. In this study, two novel (1 and 5) and four known (2, 3, 4 and 6) terpenoids were isolated from the leaves of Rubus corchorifolius L. f. Structural analysis was performed using various spectroscopic methods (1H NMR, 13C NMR and MS) to identify the following six compounds: (16α)-16,17,18-trihydroxy-ent-kauran-18-O-β-d-glucoside (1), ent-16β,17-dialkyl-3-oxygen-kaurane (2), ent-kaurane-3α,16β,17-triol (3), ent-kaurane(5R,8S,9R,10R,13R,16R)-2-one-16α,17-diol (4), (16R)-16β,17,19-trihydroxy-ent-kaur-3-one (5) and ent-16α,17-dihydroxy-kauran-19-oic-acid (6). These compounds showed different inhibitory effects on various human cancer cells. Compounds 3 and 6 exhibited stronger inhibitory effects on human colon cancer HCT116 cells than the other 4 compounds. Flow cytometry analysis demonstrated that both compounds 3 and 6 caused cell cycle arrest at the G0/G1 phase and induced cellular apoptosis in HCT116 cells. Compounds 3 and 6 modulated the expression levels of key signaling proteins closely related to cell proliferation and apoptosis, i.e., increasing the levels of poly(ADP-ribose) polymerase, p53, and p27, and decreasing the levels of EGFR, cyclin D1, CDK2 and CDK4. Overall, our findings provided insight into the anticancer components of Rubus corchorifolius L. f. leaves, which could facilitate their utilization as functional food ingredients. © The Royal Society of Chemistry.

We investigated the anti-inflammatory effects of 3α-hydroxy-lup-20(29)-en-23, 28-dioic acid (HLEDA) - a lupane-type triterpene isolated from leaves of Acanthopanax gracilistylus W. W. Smith (AGS), as well as the underlying molecular mechanisms in lipopolysaccharide (LPS)-induced RAW264.7 cells. Our results demonstrated that HLEDA concentration-dependently reduced the production of nitric oxide (NO), significantly suppressed LPS-induced expression of TNF-α and IL-1β at the mRNA and protein levels in RAW264.7 cells. Further analysis revealed that HLEDA could reduce the secretion of High Mobility Group Box 1 (HMGB1). Additionally, the results showed that HLEDA efficiently decreased nuclear factor-kappaB (NF-κB) activation by inhibiting the degradation and phosphorylation of IκBα. These results suggest that HLEDA exerts anti-inflammatory properties in LPS-induced macrophages, possibly through inhibition of the NF-κB signaling pathway, which mediates the expression of pro-inflammatory cytokines. These results warrant further studies that would concern candidate therapy for diseases, such as fulminant hepatitis and rheumatology of triterpenoids in AGS. © 2017 Japan Society for Bioscience, Biotechnology, and Agrochemistry.

As a widely used deoxyribonuclease, DNase I is involved in many physiological processes including tumor cell proliferation, metastasis and apoptosis. Furthermore, the level of this enzyme in serum can act as a functional biomarker for the therapeutic monitoring of systemic lupus erythematosus and other diseases. We report here a low cost and sensitive DNase I detecting system based on the single-stranded fluorogenic substrate and nanographene oxide (NGO) and use it for DNase-targeted natural product screening. The system with a detection limit of 0.005 U was then used to evaluate the effect of external factors on DNase I. The results show that Hg2+, As2+, Pb2+, Cd2+ and Cu2+ can inhibit DNase I activity in a concentration-dependent manner with IC50 values of 0.37 mM (Hg2+), 2.7 mM (As2+), 5 mM (Pb2+), 5.3 mM (Cd2+) and 7.8 mM (Cu2+), respectively. Meanwhile, 10 natural compounds isolated from Cyclocarya paliurus leaves were screened as DNase I inhibitors, while 5 compounds were identified as activators. Finally, the system was used to discriminate DNase activity of serum samples with and without HBV. The results showed that HBV infection significantly decreased the level of DNase I in serum samples. In summary, these data indicate that this method with the advantages of rapidity, low cost and high sensitivity is hopeful for DNase assay in biological samples as well as compound screening in vitro.

A rapid and accurate detection method for Mycobacterium tuberculosis (M. tuberculosis) is essential for effectively treating tuberculosis. However, current detection methods cannot meet these clinical requirements because the methods are slow or of low specificity. Consequently, a new highly specific ssDNA aptamer against M. tuberculosis reference strain H37Rv was selected by using the whole-cell systematic evolution of ligands by exponential enrichment technique. The selected aptamer was used to construct a fast and highly specific H37Rv sensor. The probe was produced by immobilizing thiol-modified aptamer on an Au interdigital electrode (Au-IDE) of a multichannel series piezoelectric quartz crystal (MSPQC) through Au-S bonding, and then single-walled carbon nanotubes (SWCNTs) were bonded on the aptamer by π-πstacking. SWCNTs were used as a signal indicator because of their considerable difference in conductivity compared with H37Rv. When H37Rv is present, it replaces the SWCNTs because it binds to the aptamer much more strongly than SWCNTs do. The replacement of SWCNTs by H37Rv resulted in a large change in the electrical properties, and this change was detected by the MSPQC. The proposed sensor is highly selective and can distinguish H37Rv from Mycobacterium smegmatis (M. smegmatis) and Bacillus Calmette-Guerin vaccine (BCG). The detection time was 70 min and the detection limit was 100 cfu/mL. Compared with conventional methods, this new SWCNT/aptamer/Au-IDE MSPQC H37Rv sensor was specific, rapid, and sensitive, and it holds great potential for the early detection of H37Rv in clinical diagnosis. ©2017 Elsevier B.V.
