目的探讨局灶性脑缺血(middle cerebral artery occlusion,MCAO)大鼠皮质小胶质细胞(microglia,MG)极化的变化规律,揭示脑泰方Ⅱ号调节MG极化的方式与特点。方法将SD大鼠随机分为正常组、假手术组、MCAO模型组、米诺环素组、脑泰方Ⅱ号组,于造模后d 1、3、5、7激光共聚焦观察MG形态;Real time-PCR法检测M1/M2型表面标记分子表达。结果 MCAO模型组MG胞体胀大,突触变粗、短,分支减少;脑泰方Ⅱ号可明显促进突触分支与MG增殖。模型组M1/M2型标志分子随时间被不同程度活化;脑泰方Ⅱ号自d 3后(米诺环素自d 1后)可逐步明显抑制M1型标记分子,促进M2型标记分子表达(P

Aim To explore the changing rule of microglia polarization in MCAO rat cortex, and to reveal the action mode and characteristics of Naotaifang II on microglia polarization. Methods SD rats were randomly divided into five groups; normal group, shamoperated group, MCAO model group, minocycline group and Naotaifang II group.
