Objective To determine the diosgenin contents in Bulbus lain from different areas with RP-HPLC. Methods The RP-HPLC were with'4.6 mm×250 mm, 5μm phenomenex ODS column, mobile phase of acetonitrile-water, 0.7 mL/min flow rate, 20 μL sample injection, 203nm wavelength and 30 ℃ temperature. Results The best linear correlation was the sample injection in the range of 0.04-1.60 μg and peak area(n=6); and the average recoveries of diosgenin was 98.2%(RSD=2.1%,n=6).
