Determination of the total polyphenol in Anhua dark-tea by Folin-Ciocalteu colorimetry

目的 建立安化黑茶中茶多酚含量的测定方法,以控制安化黑茶及其加工品质量.方法 依据GB/T8313-2008,采用Folin-Ciocaheu (FC)试剂和没食子酸对照品,运用分光光度法测定黑茶中茶多酚的含量.测定条件为:0.2 mol/L的FC试剂5 mL,7.5％Na2CO3溶液4 mL,25℃反应1h后于765 nm处测定吸光度.结果 没食子酸浓度在0.49～4.9 μg/mL范围内与吸光度的线性关系良好,平均回收率为98.0％,RSD为1.4％.测得3批安化黑茶茶多酚的平均含量为2.49％,RSD为1.3％.结论 建立的方法准确、可靠,可用于测定安化黑茶及其加工品中的茶多酚含量.

Objective To establish a method for determination of the content of total polyphenols in Anhua dark-tea.Methods According to GB/T 8313-2008,the total polyphenol content in Anhua dark-tea was analyzed by spectrophotometry with Folin-Ciocalteu reagent and Gallic acid used as standards.The optimal determination conditions could be calculated according to the absorption at 765 nm by reacting 5 mL 0.2 mol/L Folin-Ciocalteu with 4 mL 7.5% Na2CO3 for an hour at 25 ℃.Results In this method,the concentration of Gallic acid showed a good linearity with ...
