目的建立高效液相色谱-质谱联用测定人血浆中米格列醇浓度的方法,并用于健康人体内米格列醇药物动力学研究.方法采用MACHEREY-NAGELCN色谱柱(4.6mm×250mm,5μm),柱温50℃,以乙腈-甲醇-0.02%盐酸水溶液为流动相,流速0.80mL·min-1;质谱采用电喷雾电离源正源(ESI+),定量分析采用选择离子监测(SIR),质荷比为准分子离子峰;血浆样品用乙腈沉淀后,将乙腈挥干,残渣用氯仿-异丙醇提取杂质,磷酸液提取药物,外标法定量.结果米格列醇浓度在32～3 200 ng·mL-1内线性关系良好,相关系数为0.999 7,最低检测浓度5ng·mL-1(S/N=3).批内、批间RSD符合方法学要求,单次服用100mg米格列醇片后药动学参数AUC0～12,AU...

OBJECTIVE: To establish a high performance liquid chromatography- electrospray mass spectrometry method for the determination of miglitol in human plasma.
