摘要:
A liquid chromatography–electrospray ionization–mass spectrometry method with
multiple reaction monitoring was established for simultaneous quantification
of 18 bioactive constituents from the stem and root of Tripterygium
wilfordii Hook. f. collected from different places in China and
various commercial preparations. The chromatographic separations were
achieved on an Agilent Poroshell SB-C18 column (150 × 4.6 mm,
3.5 µm) with gradient elution using acetonitrile and 0.03 % formic acid
aqueous solution in 45 min. Detection was performed in the positive
ionization mode by monitoring the precursor–product combination. The
validation of the method included tests of linearity, sensitivity,
precision, repeatability, stability, and accuracy. All calibration curves
showed good linearity (r > 0.9990) within the test range. The
established method showed good precision and accuracy with intraday and
interday variations of 2–5 % and 1–4 %, respectively, and recoveries of
95.5–104.5 %.

摘要:
Two new oxanthrone C-glycosides, patientosides A (14) and B (15), together with three known ones (11-13), were isolated from Rumex patientia. Their structures were identified on the basis of spectroscopic methods. The absolute configuration for 14 and 15 were deduced by analysis of their CD spectra and comparison with those of known similar compounds. Compounds 11-15, and 14 known anthraquinones (1-4, 6-10, 16-20) previously isolated from Rumex nepalensis, Rumex hastatus, and endophytic Aspergillus fumigatus, respectively, as well as a commercially available compound rhein (5) were evaluated for their inhibitory effects on IL-6 and extracellular matrix production in mesangial cells. © 2013 Elsevier Ltd. 摘要:
A bioassay-directed phytochemical study was conducted to investigate potential allelochemicals in the roots of the invasive plant Ageratina adenophora. Eleven phenolic compounds, including seven new ones, 7-hydroxy-8,9-dehydrothymol 9-O-trans-ferulate (1), 7-hydroxythymol 9-O-trans-ferulate (2), 7,8-dihydroxythymol 9-O-trans-ferulate (3), 7,8-dihydroxythymol 9-O-cis-ferulate (4), methyl (7R)-3-deoxy-4,5-epoxy-d-manno- 2-octulosonate 8-O-trans-p-coumarate (5), methyl (7R)-3-deoxy-4,5-epoxy-d-manno- 2-octulosonate 8-O-cis-p-coumarate (6), and 3-(2-hydroxyphenyl)propyl methyl malonate (7), were isolated from a bioactive subfraction of the ethanol extract of the roots of A. adenophora. The new structures were established on the basis of detailed spectroscopic analysis. The potential phytotoxic effects of these compounds on the germination of Arabidopsis thaliana seeds were tested by a filter paper assay. Compound 7 and known compounds 3-(2-hydroxyphenyl)-1- propanol (8) and o-coumaric acid (9) remarkably showed inhibition activity against Arabidopsis seed germination at a concentration of 1.0 mM. Compounds 1, 2, 5, 6, and 10 showed slight inhibitory activity at the test concentration after treatment for 3 days, while the other compounds showed no obvious inhibitory effects. Moreover, 7-9 were further found to show obvious inhibitory activity on retarding the seedling growth of Ar. thaliana cultured in soil medium. ©2013 American Chemical Society.

摘要:
为了寻找新型的抗糖尿病分子, 从京尼平出发, 经过甲基化(苄基化)、 甲磺酰基取代、 叠氮基取代和还原等4步反应得到含有活泼胺基的2个母核化合物. 这2个母核化合物与不同的酰氯反应, 共得到13个新的京尼平衍生物, 总收率41%~63%, 其结构经核磁共振、 质谱和元素分析确认. 活性测试结果表明, 所有目标化合物在10 mmol/L 浓度下都有一定的抗糖尿病活性, 其中化合物10f对二肽基肽酶4(DPP Ⅳ)的抑制率达31.2%.

摘要:
Accurate prediction of molecular acidity with ab initio and density functional theory approaches is of great interest, but remains a challenging task. Density functional reactivity theory (DFRT) with quantum descriptors, such as molecular electrostatic potential and valence natural atomic orbital energies, has recently been developed and used for this purpose. Our previous study of substituted benzoic acids revealed a novel approach to quantitatively predict molecular acidity using the Hammett constant, which gave the same prediction accuracy as that of DFRT. In this work, we applied these two approaches to singly and doubly substituted phenol systems, a total of 83 molecules, confirming their effectiveness and robustness. High accuracy was obtained using both approaches, with the DFRT approach achieving slightly higher accuracy than the Hammett approach in general. These results shed further light on the molecular features governing physiochemical properties such as acidity and basicity, both locally on the atomic level and globally on the functional group or molecular level. Our present results confirm the validity of the sum rule of Hammett constants for doubly and multiply substituted compounds.

摘要:
In this paper, multi-walled carbon nanotubes (MWCNTs) were oxidized by 30 w% hydrogen peroxide to introduce hydroxyl groups and carboxyl groups on it. Catalyzed by p-toluenesulfonic acid, D-tartaric acid (DTA) was grafted onto MWCNTs by an esterification reaction. Fourier transform infrared spectroscopy and high resolution transmission electron microscopy were used to elucidate the introduced groups and the morphologies changes of the MWCNTs samples. The D-tartaric acid modified MWCNTs were used as chiral im pregnating reagents for thin-layer chromatography for enantioseparation of propranolol enantiomers, and the chiral separation factor was achieved over 7.23 by a mixed solvent of acetonitrile-tert-butanol-acetic acid (volume ratio is 49:49:2) as developing solvent.

This study aimed to investigate the composition of three major stilbenes (mulberroside A, oxyresveratrol, and resveratrol) in different portions of mulberries collected in different seasons and their change molds during growth by high-performance liquid chromatography. Mulberroside A levels were the highest in the bark and roots of Morus atropurpurea Roxb, Morus alba Linn, and Morus latifolia Poir. Oxyresveratrol levels were the highest in roots and stem. Both of these high levels were in September. The amount of resveratrol was very low in all samples. In the stem, Morus latifolia Poir contained more mulberroside A than the other two mulberries. Mulberroside A was not detected in the leaves of the three mulberries. In Morus atropurpurea Roxb seedlings, the root tended to contain more of the three stilbenes than leaves. The temporal peaks of resveratrol were always ahead of those for oxyresveratrol. The levels of the stilbenes varied in different portions of the varieties of mulberries collected in different season and in the seedlings of Morus atropurpurea Roxb.

摘要:
In this paper, carbon nanotubes (CNTs) were added into chitosan (CS) hydrogels in the form of chitosan modified CNTs (CS-CNTs) composites to prepare carbon nanotubes hydrogels (CNTs-GEL). The products, named CS-MWCNTs, were characterized by scanning electron microscope (SEM) and Fourier transform infrared (FTIR) spectroscopy. Swelling properties and effect of pH on controlled release performance of the two kinds of hydrogels, CNTs-GEL and pure chitosan hydrogels without CNTs (GEL), were investigated respectively. The results showed that CNTs-GEL possess better controlled release performance than GEL. The releasing equilibrium time of CNTs-GEL was longer than that of GEL in both pH = w7.4 and pH=1.2 conditions, although the release ratios of the model drug are similar in the same pH buffer solutions. It is found that release kinetics is better fitted Ritger-Peppas empirical model indicating a fick-diffusion process in pH = 1.2, while in pH = 7.4 it was non-fick diffusion involving surface diffusion and corrosion diffusion processes. © 2013 by School of Pharmacy.

目的优选乳核分散片的最佳提取工艺.方法以有效成分丹参酮ⅡA含量和浸膏得率为考察指标,采用正交试验设计,对乳核分散片的醇提工艺进行优化.结果最佳醇提工艺条件为:加入6倍、4倍量70%乙醇提取两次,分别提取1.0h、0.5h.结论优选的提取工艺合理、快速、简便可行,可为乳核分散片工业化生产提供实验依据.

目的探讨乙肝宁复方氨基酸类吸湿性及壳聚糖絮凝澄清与醇沉工艺纯化后乙肝宁中氨基酸变化规律。方法采用阳离子交换树脂分离乙肝宁水提液中的氨基酸,测定吸湿百分率;将乙肝宁水提液经絮凝澄清与醇沉工艺处理后,用732号阳离子交换树脂富集,采用异硫氰酸苯酯(PITC)柱前衍生,梯度洗脱,测定乙肝宁原浸膏及两种纯化工艺处理后游离氨基酸。结果乙肝宁复方总游离氨基酸吸湿性远远高于乙肝宁复方原浸膏;壳聚糖絮凝澄清法对乙肝宁复方水提液中精氨酸、赖氨酸的保留明显优于醇沉法,而对于脯氨酸的保留,虽不如醇沉法,但仍有较高的保留率。结论壳聚糖絮凝澄清法适合用于乙肝宁复方的纯化研究;壳聚糖絮凝澄清法相对于醇沉法可降低中药浸膏吸湿性的原因与氨基酸无关。

二角菱壳和四角菱壳不同提取物抗氧化能力比较研究

摘要:
目的:研究不同菱角壳不同溶剂的提取物的抗氧化能力。方法采用DPPH法分别测定二角菱角壳、四角菱角壳氯仿、乙酸乙酯、乙醇、水提取浸膏的抗氧化能力,同时与抗坏血酸、槲皮素、没食子酸标准品进行了对比。结果用同一溶剂提取时,二角菱壳提取浸膏的 IC50值均高于四角菱壳提取浸膏;同一种菱角壳用不同溶剂提取时,其提取浸膏与标准品相比 IC50值为:氯仿浸膏>水浸膏>乙醇浸膏>抗坏血酸>乙酸乙酯浸膏≈槲皮素>没食子酸,即其清除 DPPH自由基能力的顺序为:没食子酸>乙酸乙酯浸膏≈槲皮素>抗坏血酸>乙醇浸膏>水浸膏>氯仿浸膏。结论菱角壳具有较好的抗氧化能力。四角菱壳抗氧化能力强于二角菱壳,菱角壳乙酸乙酯提取部分清除DPPH自由基的能力最强。
