摘要:
Schisanlactone E (SE) is a major triterpene obtained from the plants of genus Kadsura. The aim of this research was to investigate the transformed metabolites of SE by fungi and evaluate the bioactivities of these products. After screening 10 strains of filamentous fungi, Cunninghamella blakesleana AS 3.970 was chosen as a potent organism to be used for the biotransformation of SE. 13 metabolites were obtained and determined to be new compounds through the use of spectroscopic data, including UV, 1D-, 2D-NMR, and HR-ESIMS. Furthermore, in an in vitro bioassay, metabolites 7 and 9 showed moderate inhibitory effects on the nitric oxide production in LPS-induced macrophages with IC50 values of 16.73, 5.91 mu M, respectively; 9 could inhibit the proliferation of acetaldehyde-induced HSC-T6 cells, with the IC50 value of 21.4 mu M. Objective: The nucleus of the solitary tract(NTS) is a primary center for both visceral afferents and somatic afferents.Previous experiments demonstrated that NTS is associated with connection of the stomach and acupoints in the Stomach Meridian of Foot-Yangming(ST Meridian).In this study, an extracellular recording and immunochemistry method was used to analyze the neuron discharge and c-fos protein expression in the NTS following acupuncture at different acupoints and non-acupoint.Methods: A total of 104 discharging neurons were detected in the NTS of 52 rats, of which, 86 were provided with complete data.Results: After acupuncture at Sibai(ST2), Zusanli(ST36), Neiting(ST44), Quanliao(SI18) and non-acupoint, the response neuron occurring rates(RNORs) in the NTS were 65.12%, 51.16%, 46.51%, 34.88% and 31.40%, respectively.For RNOR,there was a significant difference among ST2, ST36, ST44, SI18 and non-acupoint(P<0.01 or P<0.05).Accordingly, in other 48 rats, the number of c-fos immunoreactive(c-fos-IR) neurons in the NTS after electroacupuncture(EA) at ST2 was significantly higher than at other acupoints and non-acupoint(P<0.05 or P<0.01).Both EA at ST36 and at ST44 increased c-fos-IR neurons significantly as compared with EA at SI18 and non-acupoint(P<0.05 or P<0.01), while there was no difference between EA at SI18 and at non-acupoint groups(P>0.05).Conclusion: The experiment demonstrated that the afferent convergence in the NTS was different by body surface point stimulus, which suggested that NTS might be a primary center in the central nervous system receiving acupoint stimulus from ST Meridian.

以问题为基础(Problem- based learning,PBL)的教学法和案例教学法都是近年来我国医学教育改革中经常采用的两种新型教学方法。将“以问题为基础”的教学模式与案例教学法有机结合引入《针灸学》教学,符合《针灸学》的学科特点,能有效提高教学质量,在教育观念上更易于被医学生所接受,因而适应对高素质医学人才培养的需要。
