Objective: To observe the effects of Ganlu Xiaodu Fill (甘露消毒丹) and its separate prescriptions on splenic interleukin 10 (IL-10) and transforming growth factor-alpha (TGF-α) and the relative expression of nuclear factor-κB (NF-κB) in brain tissue in baby rats which were infected with Coxsackie virus 16 (CoxA16), and to investigate its mechanism underlying anti- inflammatory. Methods: The 40 mice were divided into control group, model group, the full formula group, the aromatic residual group (Ganlu Xiaodu Fill aromatic dampness pharmaceutical composition), the clear residual group (Ganlu Xiaodu Fill heat antidotes composition). The spleen level of IL-10 and TGF-αwere detected by enzyme-linked immunosorbent assay (ELISA), while the content of NF-κB protein in brain tissue was determined by Western blot. Results: Compared with the control group, spleen tissue IL-10, TGF-α content in model group increased, the brain tissue relative expression of NF-κB regulated (P〈0.05 or P〈0.01). Compared with the model group, spleen tissue IL-10, TGF-α was significantly increased in the full formula group, relative brain tissue NF-κB expression was significantly down-regulated (P〈0.05 or P〈0.01). Compared with the aromatic residual group, spleen tissue IL-10 content in full formula group obviously increased, but the relative expression of NF-κB in brain tissue down- regulated (P〈0.05). In comparison with the clear residual group, the whole spleen TGF-α content increased in the full formula group. Conclusion: Ganlu Xiaodu Fill and its separate prescriptions have the anti inflammatory effect on baby rats which were infected with Coxsackie virus 16, and the mechanism maybe related to increasing the content of IL-10 and TGF-α, and down-regulating the the relative expression of NF-κB.
