摘要:
The circadian rhythm is an endogenous clock system that coordinates and optimizes various physiological and pathophysiological processes, which accord with the master and the peripheral clock. Increasing evidence indicates that endogenous circadian rhythm disruption is involved in the lesion volume and recovery of ischemic stroke. As a critical recovery mechanism in post-stroke, angiogenesis reestablishes the regional blood supply and enhances cognitive and behavioral abilities, which is mainly composed of the following processes: endothelial cell proliferation, migration, and pericyte recruitment. The available evidence revealed that the circadian governs many aspects of angiogenesis. This study reviews the mechanism by which circadian rhythms regulate the process of angiogenesis and its contribution to functional recovery in post-stroke at the aspects of the molecular level. A comprehensive understanding of the circadian clock regulating angiogenesis in post-stroke is expected to develop new strategies for the treatment of cerebral infarction.

A convenient and highly sensitive bioassay employing graphene oxide (GO) as a nanocarrier and nicking enzyme (Nb.BbvC I)-assisted signal amplification was developed to detect Pseudomonas aeruginosa. Furthermore, the assay employed a DNA probe with a capture probe (CP) and a help probe (HP), which were designed to bind the target and signal probe (SP), respectively. In the absence of the target, the SP did not bind to the HP due to steric effects. Consequently, the fluorescence of carboxyfluorescein (FAM) was quenched by the black hole quenching group (BHQ). The CP specifically recognized the introduced target, which opened the HP that was bound to the SP. Nb.BbvC I cleaved the formed nicking site, which restored the fluorescence and simultaneously released the HP for the next round of reaction. The target densities, which ranged from 10 to 107 colony-forming units (CFU) mL−1, and the fluorescence intensity had a linear relationship. The limit of detection (LOD) under the optimal conditions was 5 CFU mL−1. Moreover, this bioassay performed well in the analysis of water samples. These results demonstrate that this method is highly specific and sensitive for determining P. aeruginosa and is promising for the clinical diagnosis of infectious diseases. © 2021 Taylor & Francis Group, LLC.

摘要:
Biofilm formation is known to promote drug resistance in methicillin-resistant Staphylococcus aureus (MRSA), which is closely related to persistent infections in hospital settings. In this study, a DNA aptamer specific to penicillin-binding protein 2a (PBP2a) with a dissociation constant (K d) of 82.97 ± 8.86 nM was obtained after 14 cycles of systematic evolution of ligands by exponential enrichment (SELEX). Next, a bifunctional complex containing the aptamer intercalated by berberine into the double-strand region was prepared and adsorbed onto the surface of graphene oxide (GO) by π-stacking interactions. The GO-loaded aptamer/berberine bifunctional complex showed significantly higher inhibition of MRSA biofilm formation than the control. Furthermore, this study shows that the complex possesses anti-biofilm activity, which can be attributed to the ability of the aptamer to reduce cell-surface attachment by blocking the function of PBP2a and berberine to attenuate the level of the accessory gene regulator (agr) system, which plays an important role in mediating MRSA biofilm formation. Therefore, the simultaneous delivery of berberine and PBP2a-targted aptamer using GO may have potential for the treatment of chronic infections caused by MRSA biofilms. <i>Objective</i>. Bioinformatics methods were used to analyze non-small-cell lung cancer gene chip data, screen differentially expressed genes (DEGs), explore biomarkers related to NSCLC prognosis, provide new targets for the treatment of NSCLC, and build immunotyping and line-map model. <i>Methods</i>. NSCLC-related gene chip data were downloaded from the GEO database, and the common DEGs of the two datasets were screened by using the GEO2R tool and FunRich 3.1.3 software. DAVID database was used for GO analysis and KEGG analysis of DEGs, and protein-protein interaction (PPI) network was constructed by STRING database and Cytoscape 3.8.0 software, and the top 20 hub genes were analyzed and screened out. The expression of pivot genes and their relationship with prognosis were verified by multiple external databases. <i>Results</i>. 159 common DEGs were screened from the two datasets. PPI network was constructed and analyzed, and the genes with the top 20 connectivity were selected as the pivotal genes of this study. The results of survival analysis and the patients’ survival curve was reflected in the line graph model of NSCLC. <i>Conclusion</i>. Through the screening and identification of the VIM-AS1 gene, as well as the analysis of immune infiltration and immune typing, the successful establishment of the rosette model has a certain guiding value for the molecular targeted therapy of patients with non-small-cell lung cancer.

摘要:
目的:采用高效液相色谱法建立防风药材和防风饮片的指纹图谱,考察防风炮制前后的量值传递信息,为建立防风饮片质量控制及质量标准提供依据.方法:液相色谱柱(月旭,Ultimate(C)XB-C18,4.6mm×250mm,5μm);以十八烷基硅烷键合硅胶为填充剂;流动相为甲醇—水;洗脱方式:线性梯度,0~70 min(20%甲醇~90%甲醇);检测波长为254 nm;流速:1.0mL· min-1;进样量:10μL;柱温:30℃,理论板数按升麻素苷峰计算应不低于2000.共测定了23批防风药材和饮片的指纹图谱,通过应用中药色谱指纹图谱相似度评价系统2012版对两者相似度进行配对t检验分析.结果:在选定的色谱条件下,确定15个色谱峰构成23批防风的指纹图谱特征峰,防风炮制前后指纹图谱基本一致(配对样本相关系数γ=0.9374,均值差异检验t=0.6841,df=22,P=0.5011),防风炮制前后其化学成分变化无统计学差异(P>0.05).结论:防风药材和饮片指纹图谱大体一致,防风炮制成饮片后,未改变防风药材的化学成分,并保留了防风原药材的质量属性,量值传递率高.

胡不群教授认为急危重症往往虚实错杂,辨析病情虚实、权衡攻补关系是处理急危重症的关键,其临证擅于以虚实为纲、以扶正驱邪为法诊疗急危重症.通过列举胡教授运用桃核承气汤治疗邪实甚急、神昏抽搐案,托里透脓散合千金苇茎汤治疗虚实并重、痰热壅肺案,独参汤合大承气汤治疗邪实正衰、热深厥甚案,将其诊疗经验总结如下:(1)邪实并存,以邪气实为急中之急者,当先驱其邪,邪去而后再议扶正;(2)邪实并存,虚实并重者,当扶正驱邪兼顾,至于其扶几分正,驱几分邪则又当据临床具体情况而定,无定法可依;(3)虚实错杂,正气欲脱,而又邪甚,必须攻伐者,则又当先托住正气,而后再议攻邪.

目的:探究氯化锂-匹罗卡品诱导癫痫大鼠海马组织差异表达共价闭合、单链环状RNA(circRNA)及功能分析.方法:通过构建氯化锂-匹罗卡品癫痫大鼠模型,取癫痫大鼠及对照大鼠海马组织通过Illumina高通量测序平台进行转录组测序,并通过EdgeR包对circRNA进行差异分析、通过R语言进行GO及KEGG信号通路富集分析,通过Targetscan与miRanda软件进行circRNA与miRNA互作分析寻找circRNA核心调控靶标.结果:对照组及癫痫组大鼠海马的circRNA进行差异分析获得的262个差异表达circRNA,信号通路富集分析发现与胞蛋白修饰过程、蛋白质变性过程、树突形态调控、神经元分化调节、离子跨膜转运的调节等生物过程相关.并进一步确定了可能与circRNA存在互作的核心调控miRNA,包括miR-322-5p、miR-322-3p、miR-323-5p、miR-323-3p、miR-301a-5p、miR-301a-3p、miR-324-5p、miR-324-3p等.结论:本研究筛选了氯化锂-匹罗卡品点燃癫痫大鼠与正常大鼠脑组织差异circRNA,系统地分析了其可能的作用功能机制,为癫痫诊断或治疗寻找到可靠的生物标志物.

摘要:
目的:通过电子舌智能感官评定与人工口尝评价相结合的方法筛选山苓荷甘粉的最佳矫味剂与辅料组合。方法:基于电子舌技术综合评分将常用的20种矫味剂与辅料和山苓荷甘干浸膏配伍,找出矫味剂最佳用量及矫味剂与辅料最优组合,再通过人工口尝评价优选出一种最佳的矫味剂与辅料组合。电子舌智能感官评定统计分析采用95%可信区间重叠法及单因素试验LSD法,进行多组间的两两比较;人工口尝评价结果采用DPS统计软件中单向有序列联表项下的秩和检验,进行处理数据。结果:电子舌智能感官评定结果表明,甜菊糖苷用量在1.05%增甜作用最好,β-环糊精抑制苦味及涩味作用最优;电子舌智能感官评定初筛出5组样品,人工口尝评价结果最终选出最优的矫味剂组合比例是:山苓荷甘粉干浸膏占48%,β-环糊精占50.95%,甜菊素占1.05%。结论:通过电子舌智能感官评定与人工口尝评价相结合的方法,可以科学正确合理地筛选出山苓荷甘粉最适合的矫味剂及辅料,显著改善口感,优化制剂处方。

肺癌(lung cancer)的高发病率与高死亡率对人民健康造成了极大的危害.中医药在恶性肿瘤的防治方面具有广泛的临床经验,在治疗肺癌方面也显示出明显的优势.实验动物模型是肺癌研究领域中必不可少的一部分.构建中医特色的病证结合动物模型是以中医辨证论治理论为基础,将中医证候纳入了评价标准,使动物模型既符合西医疾病特征又具备中医证候的特点.本文总结了当前常用的肺癌动物模型和病证结合动物模型的构建方法与特点,以期为从事肺癌科学研究的中西医科研工作者提供参考.

Objective: This study aimed to design and optimize the water extraction process for Chuantieling (喘贴灵, CTL) based on the concept of quality by design (QbD). Methods: The single-factor experiments were used to select the best experimental points of CTL water extraction. On this basis, the transfer rate of ephedrine hydrochloride and sinapine thiocyanate, and the yield of the extract were evaluated as the evaluation indicators. The liquid-solid ratio, extraction time, and pH value were selected as the main factors to carry out the Box-Behnken design (BBD). A mathematical model of the critical process parameters (CPPs) and critical quality attributes (CQAs) was established, the interaction between CQAs and CPPs was examined, and the CTL extraction process design space was established and optimized, which guaranteed the stability of the process. The high performance liquid chromatography (HPLC) was used to analyze the main active compounds in the CTL extract. Results: Through single-factor experiments, the best experimental parameters were found to be a liquid-solid ratio of 8∶1, extraction time of 90 min, pH value of 4, and extraction times of three. The experimental variance analysis results of the BBD showed that the P value of the regression model was less than 0.05, and the lack-of-fit value was greater than 0.01, indicating that the constructed model had good predictive ability. The operating space of the CPPs of the CTL water extraction process was combined with the actual production situation. In the production situation, the best extraction process was nine times of water addition, 72 min of extraction time, and 4.5 of pH value. The HPLC results showed that the peak areas of ephedrine hydrochloride, sinapine thiocyanate, tetrahydropalmatine, methyl eugenol, cinnamaldehyde, and 6-gingerol in the CTL extract accounted for 0.94%, 14.32%, 0.78%, 31.23%, 0.34%, and 0.44% of the total peak area, respectively. Conclusion: The water extraction process design space of CTL based on QbD was conducive to actual production operations, ensuring the stability of the process. © 2022 Digital Chinese Medicine

动脉粥样硬化(AS)发病机制复杂,涉及通路众多,其中NOX/ROS-NF-κB信号通路在AS形成过程中具有重要作用,但相关机制尚待进一步明确.本文主要对NOX/ROS-NF-κB信号通路进行概述,并总结该通路与AS之间的关系,以期为后续研究提供参考.

益气活血开窍组分中药对脑缺血后白质保护的网络药理学研究及药效学验证

摘要:
目的 采用网络药理学方法,以髓鞘再生的主要功能细胞少突胶质前体细胞(oligodendrocyte precursor cells,OPC s)为靶点,预测益气活血开窍组分中药有效成分对脑缺血后白质损伤的促修复作用。方法 从TCMSP、PubChem、SwissTargetPrediction数据库中预测组分中药主要有效成分促进脑卒中后OPCs修复脑白质的作用靶点;整合GEO、GeneCards和CTD数据库中获得的脑卒中疾病靶基因,通过STRING绘制“中药组分-疾病”靶点PPI网络图;并进行GO及KEGG富集分析。同时,采用大鼠脑缺血再灌注模型,从神经功能和形态学评价益气活血开窍组分中药对脑缺血再灌注后脑白质损伤的促修复作用。结果 网络药理学共获取33个疾病-药物共同潜在靶点。PPI网络及拓扑分析结果显示VEGFA、PTGS2、NTRK1、ESR2和MET等可能为核心作用靶点,富集分析主要涉及钙离子、PI3K-Akt、VEGF、Rap1、Ras、EGFR酪氨酸激酶抑制剂抗性和MAPK信号通路,KEGG富集分析GEO数据集差异基因结果显示,卒中5 d和15 d均涉及PI3K-Akt信号通路。药理学实验结果显示,益气活血开窍组分中药可改善脑缺血再灌注后大鼠神经功能障碍,减轻脑白质损伤和大鼠胼胝体、纹状体髓鞘脱失。结论 益气活血开窍组分中药可改善脑缺血再灌注后白质损伤,机制可能与各有效成分通过多靶点、多途径促进髓鞘再生和白质修复有关。

基于数据挖掘的《景岳全书·血证》用药规律分析

摘要:
目的:探讨《景岳全书·血证》组方用药规律及张景岳治疗血证的学术思想。方法:收集《景岳全书·血证》中的方剂,建立方剂数据库,采用中医传承计算平台V3.0软件分析方剂中的组方用药规律。结果:共收录方剂136首,涉及中药157味,使用频次较高药物31味(频次≥7),排名前5位分别为:甘草(65)、人参(42)、茯苓(36)、当归(32)、熟地黄(30),药物四气以温、寒为主,五味以甘、苦为主,归经以脾经最多,功效以补虚类药最高,核心药物组合26组,药物关联规则9条,K-means聚类得到新方4首。结论:通过中医传承计算平台V3.0软件揭示了《景岳全书·血证》用药规律,治法上重视调补气血,亦不忽视祛除火热外邪,擅补脾肺元气,滋养肝肾命门精血,体现了张景岳论治血证的学术思想,为现代临床治疗出血性疾病提供参考。

中医药调节肠道菌群治疗慢传输型便秘的研究进展

摘要:
本文基于肠道菌群与慢传输型便秘之间的关系,探讨了中医药调节肠道菌群治疗该病的机制,认为肠道菌群的生理功能与传统医学的整体观念及阴阳平衡理论契合.慢传输型便秘的发病与肠道菌群失调有关,中医药可以通过调节肠道菌群代谢产物、肠道神经递质、肠道免疫应答等途径促进胃肠动力提升,从而改善慢传输型便秘.本文揭示了中医药调节肠道菌群治疗慢传输型便秘的作用机制,为中医药在防治该疾病的临床与科研提供理论参考.

摘要:
背景冠心病是临床常见的心血管疾病,且心血管疾病风险存在性别差异。通脉养心丸已被广泛用于治疗冠心病,但其具体的作用机制仍不清楚。目的 采用生物信息学方法分析通脉养心丸治疗不同性别老年冠心病患者的分子机制。方法 2021年6—10月,从基因表达综合数据库(GEO)获取GSE142008数据集,将治疗前3例女性患者、3例男性患者的外周血单个核细胞样本作为对照组,治疗后3例女性患者、3例男性患者的外周血单个核细胞样本作为实验组。采用R 3.6.3软件及相关安装包进行数据预处理及差异基因的筛选,利用STRING数据库对差异基因制作蛋白质互作网络(PPI),将所得结果导入Cytoscape软件(3.8.2版本)中的MCODE、cytoHubba插件以筛选基因,再利用韦恩图在线工具将筛选出来的基因进行对比以获得关键基因。使用R 3.6.3软件及相关安装包进行GO功能富集分析和KEGG通路富集分析。结果 数据预处理结果显示:各个样本中位数基本在一个水平线上,提示样本间归一化程度好。差异基因筛选结果显示:老年女性、男性冠心病患者治疗前后比较差异基因分别有796、1 159个。PPI及关键基因选取结果显示:在老年女性冠心病患者中,最终选取了BTBD1、FBXL5、KBTBD7等10个关键基因;在老年男性冠心病患者中,最终选取了PJA2、LTN1、BTBD1等10个关键基因。在老年女性冠心病患者中,GO功能富集分析结果显示,关键基因主要富集于翻译后的蛋白质修饰等生物过程,SCF泛素连接酶复合物等细胞组成,泛素样蛋白转移酶活性等分子功能;KEGG通路富集分析结果显示,关键基因主要与泛素介导的蛋白水解、昼夜节律通路相关。在老年男性冠心病患者中,GO功能富集分析结果显示,关键基因主要富集于蛋白酶体介导的泛素依赖性蛋白质分解代谢过程等生物过程,泛素连接酶复合物等细胞组成,泛素蛋白转移酶活性等分子功能;KEGG通路富集分析结果显示,关键基因主要与泛素介导的蛋白水解通路相关。结论 通脉养心丸治疗不同性别老年冠心病患者具有多靶点和多通路协调的特点,在老年女性患者中,其作用机制主要与泛素介导的蛋白质水解、细胞自噬、炎症反应、铁代谢、生物钟、激素调节等有关;在老年男性患者中,其作用机制主要与泛素介导的蛋白质水解、细胞自噬、DNA损伤修复等有关。

面部病色行走理论来源自《内经》,依据其内容及后世医家相关阐述,将其命名为"望色行走".本文从行走方向、所向何方、表现形式、疾病逆从4个方向探讨"望色行走"的主要思想内涵,发现行走方向有六,病色所向趁虚而走,蔓延、转移为病色行走的两大方式.在病色行走的基础上结合五色查病,察其疾病浅深顺逆.以期为面部望诊提供思路.

基于Leptin/CCK-PKC通路的四神丸及其拆方治疗脾肾阳虚泄泻的药效机制研究

摘要:
目的 研究四神丸及其拆方通过调控Leptin/CCK-PKC信号通路对脾肾阳虚泄泻模型大鼠的效应机制.方法 将50只SPF级SD大鼠随机分为正常组、模型组、阳性对照组、四神丸组及四神丸拆方组,每组各10只.造模成功后灌胃14d,观察各组大鼠的一般状态、腹泻指数、胃肠运动功能检测;采用ELISA法检测大鼠血清中瘦素(Leptin)、胆囊收缩素(CCK)以及胃窦、十二指肠组织中三磷酸肌醇(IP3)的含量变化;采用RT-PCR技术检测大鼠胃窦、十二指肠组织中Leptin、瘦蛋白长型受体(Ob-Rb)、CCK、胆囊收缩素1(CCK1R)、PLC、PKC的mRNA表达.结果 与正常组相比,模型组大鼠的腹泻指数、小肠推进率、胃肠组织 IP3含量升高;胃残留率、血清中Leptin、CCK、胃窦及十二指肠组织中Leptin、OB-Rb、CCK、CCK1R、PKC的mRNA表达量均降低且差异均有统计学意义(P<0.05或P<0.01).与模型组相比,四神丸组、四神丸拆方组和阳性对照组腹泻指数、小肠推进率、胃肠组织IP3含量降低;胃残留率、血清中Leptin、CCK,胃窦及十二指肠组织中Leptin、OB-Rb、CCK、CCK1R、PKC的mRNA表达量均升高且差异均有统计学意义(P<0.05或P<0.01).结论 四神丸及其拆方组对脾肾阳虚泄泻大鼠的作用可能与调控Leptin/CCK-PKC信号通路有关.

摘要:
目的研究阿魏酸调控内皮祖细胞(endothelial progenitor cells,EPC)分泌外泌体(exosomes,Exo)及对表达血管新生微小RNA的影响。方法将EPC随机分为实验组和对照组。实验组加入8 mg·L~(-1)阿魏酸干预24 h,对照组用等容量的磷酸盐缓冲溶液干预24 h。用透射电子显微镜观察EPC - Exo形态,用蛋白质印迹法鉴定其特征性表面标志物,用二辛丁酸蛋白定量法检测总蛋白浓度,用实时荧光聚合酶链反应法检测EPC - Exo中血管新生相关miRNA分子的表达水平。结果培养24 h后,各组EPC - Exo呈圆形或椭圆形的膜囊泡结构, EPC - Exo特征性表面标志物CD9、CD63和TSG101的表达均呈阳性。干预24 h后,实验组与对照组的EPC - Exo蛋白浓度分别为(1.01 ± 0.02)和(0.85 ± 0.01) μg·μL~(-1), miR - 21表达水平分别为2.51 ± 0.34和0.99 ± 0.12, miR - 126 - 5p表达水平分别为3.97 ± 0.45和1.04 ± 0.03,miR - 146a - 5p表达水平分别为8.04 ± 0.52和1.06 ± 0.05,miR - 210表达水平分别为4.77 ± 0.37和1.12 ± 0.03,miR - 214 - 5p表达水平分别为0.74 ± 0.06和0.39 ± 0.02,差异均有统计学意义(均P < 0.05) 。结论阿魏酸能促进EPC分泌Exo,且EPC - Exo中富含有与血管新生相关的miRNAs。
