Study on the Inhibition of PLD on IAV-induced Pulmonary Macrophage Based on Autophagy and Apoptosis

摘要:
Background: Platycodin D (PLD) comes from the main triterpenoid saponins in the dry roots of Platycodon grandiflorum, which has been used in traditional Chinese medicine in the treatment of respiratory disease. This study aimed to evaluate the therapeutic effects of PLD on mouse alveolar macrophages induced by influenza A virus (IAV) in vitro and to investigate the PLD of its action. Materials and Methods: Raw264.7 mouse alveolar macrophages were grown in culture and the Cell Counting Kit-8 assay determined cell viability. Cells were treated for 24 h with increasing doses of PLD (0, 5, 10, 20, 40, 60, 80, 100 mu M). The apoptosis was detected by Annexin V-FITC/PI and Tunel assay. Levels of interleukin (IL)-1 beta, IL-6 and tumor necrosis factor (TNF)-alpha of supernatants were assessed using enzyme-linked immunosorbent assay. The numbers of phagophores, autophagosomes, and autolysosomes and the ultrastructure of cells were assessed using transmission electron microscope assay. The relative protein expression of microtubule-associated protein light chain (LC) 3I, LC3II, Beclin-1, and B cell lymphoma (Bcl2) was evaluated by Western blotting. The localizations of Beclin-1 and Bcl2 were viewed with a fluorescence microscope. Results: PLD treatment resulted in significant dose-dependent inhibition of the growth of Raw264.7 cells with the half-maximal inhibitory concentration of 115.74 mu M at 24 h. After Raw264.7 cells infected by influenza virus, PLD (2.5, 5, and 10 mu M) can improve the activity of cells, reducing apoptosis of cells, decreasing the concentrations of IL-1 beta, IL-6, and TNF-alpha in the supernatant of cells, reducing autophagic injury of cells and regulating the protein expression of Beclin-1 and Bcl2. Conclusion: PLD could inhibit excessive apoptosis and autophagy of macrophage cells induced by IAV in a dose-dependent manner and then inhibits the secretion of inflammatory factors caused by influenza virus infection caused by apoptosis and autophagy and then ultimately regulate the pathogenesis of influenza virus.

摘要:
目的 运用网络药理学及高通量分子对接技术,筛选金花清感颗粒中与人类新型冠状病毒3CL水解酶及ACE2受体蛋白结合活性较好的有效化合物,并对其干预COVID-19的机制进行预测.方法 本研究利用TCMSP数据库及高通量分子对接技术筛选金花清感颗粒中与人类新型冠状病毒3CL水解酶及ACE2受体对接活性较高的的活性化合物,并获取其对应靶标,利用STRING数据库进行靶标的蛋白网络互作,运用数据包进行京都基因与基因组百科全书(KEGG)通路富集分析,预测其作用机制.结果 金花清感颗粒共筛选有效活性化合物276种,涉及靶标基因325个.“药物-活性化合物-靶标”网络图中筛选出关键化合物11个,高通量分子对接筛选得到与人类新型冠状病毒3CL水解酶及ACE2受体结合活性较好的8个化合物,其靶基因的PPI网络主要涉及AKTI、HSP90AA1、RELA等核心靶蛋白.KEGG富集分析获得94条相关通路,主要包括卡波西肉瘤相关疱疹病毒感染、丙型肝炎、麻疹、人巨细胞病毒感染、EB病毒感染等病毒相关通路以及IL-17通路、TNF通路、AGE-RAGE等途径.结论 金花清感颗粒中3-甲氧基光甘草定、粗毛甘草素C、甘草查尔酮B等活性成分与人类新型冠状病毒3CL水解酶及ACE2受体酶结合活性较强,并且通过多种生物学途径及通路发挥干预COVID-19的作用.

摘要:
目的 研究胃溃疡脾胃虚寒型患者采用西药治疗配合温中消疡汤、艾灸治疗的临床价值,为后期临床治疗该疾病提供依据.方法 以我院于2019年1月-2019年8月期间收治的60例胃溃疡脾胃虚寒型患者作为研究对象,依据病例单双号随机分为研究组和对照组,两组患者数量均为30例,两组患者均采取西药治疗,研究组患者在此基础上采用温中消疡汤、艾灸治疗,研究比较两组患者的临床治疗效果.结果 临床治疗总有效率指标比较,研究组均优于对照组(P<0.05).结论 胃溃疡脾胃虚寒型患者采用西医治疗配合温中消疡汤、艾灸治疗临床效果显著,值得推广应用.

巨噬细胞是机体重要的免疫细胞,在不同的病理条件下表现为不同的炎症表型,即M1型巨噬细胞(经典活化的巨噬细胞)和 M2型巨噬细胞(替代活化的巨噬细胞)。在促炎表型M1型巨噬细胞中,琥珀酸大量堆积,并介导激活一系列炎症途径。本文综述了M1型巨噬细胞中,琥珀酸的来源及其病理作用,为研究琥珀酸及其介导的炎症途径提供理论支持。

摘要:
A graphene oxide (GO)–based fluorescent bioassay was developed to quantify agrA gene transcription (its mRNA) in methicillin-resistant Staphylococcus aureus (MRSA). This method is based on the use of Klenow fragment (KF)–assisted target recycling amplification and hybridization chain reaction (HCR). A triple complex was designed that contained a capture probe (CP), a trigger probe (TP), and a help probe (HP), which were partially complementary to one another. In the absence of the target, all the oligonucleotides labeled with carboxyfluorescein (FAM) are adsorbed onto the surface of GO by π-stacking interactions. This adsorption quenches the FAM signal. On the contrary, the target RNA causes the triple complex to disintegrate and initiates strand-displacement polymerization reaction (SDPR) and HCR in the presence of the appropriate raw materials, including the primer, KF, dNTPs, hairpin 1 (H1), and hairpin 2 (H2), generating double-stranded DNA (dsDNA) products. These dsDNA products are repelled by GO and produce strong fluorescence, measured at excitation/emission wavelengths of 480/514nm. The fluorescent signal is greatly amplified by SYBR Green I (SGI) due to the synergistic effect of dsDNA-SGI. The target was assayed with this method at concentrations in the range 10 fM to 100 pM, and the detection limit (LOD) was 10 fM. This method also displayed good applicability in the analysis of real samples. It provides a new way of monitoring biofilm formation and studying the mechanisms of drug actions. 目的:依托大学生创新训练中心,对独立学院学生开展课外科技活动,提升独立学院学生创新能力,有助于中医药院校独立学院学生适应当下社会对综合性高素质人才的需求。方法:依托大学生创新训练中心,结合二级学院的专业特点、分不同年级、根据兴趣双向选择,自主设计科研课题,通过参加实验设计、动手操作、记录分析和处理结果等综合评价中医药课外科技活动在提升独立学院学生创新能力的作用。结果:依托大学生创新训练中心的课外科技活动成果以论文为形式,在很大程度上提升了独立学院学生的创新能力,受益学生每学年约500人次。结论:依托一个具备较成熟的课外科技活动实施体系的大学生创新训练中心,开设一些高水平实验,在很大程度上能够提升独立学院学生创新能力。

摘要:
目的:基于TL R7/8介导的I F N-α/β蛋白表达水平探讨麻黄先煎之麻杏石甘汤抗流感病毒效应机制。方法:以TLR7/8激活剂(R848)、抑制剂(抗体TLR7和抗体TLR8)干预的小鼠巨噬细胞为对照,以酶联免疫吸附技术检测流感病毒干预小鼠巨噬细胞24、48h的IFN-α/β分泌水平。以流感病毒干预的小鼠巨噬细胞为对照,以酶联免疫吸附技术和Western Blot技术检测麻黄先煎之麻杏石甘汤含药血清干预24、48h的流感病毒感染的小鼠巨噬细胞IFN-α/β分泌水平和蛋白表达水平。结果:与正常大鼠血清组及胎牛血清组比较,TLR7/8激活剂(R848)与流感病毒干预24、48h的RAW264.7细胞IFN-α和IFN-β分泌水平和蛋白表达水平显著升高(P<0.05);与病毒干预组比较,四药同煎及麻黄先煎20、25、30min之麻杏石甘汤含药血清作用24、48h后,流感病毒感染后的巨噬细胞IFN-α/β的分泌水平和蛋白表达水平显著降低(P<0.05);与四药同煎组比较,在含药血清作用48h的时间点,麻黄先煎25min之麻杏石甘汤组IFN-α分泌水平显著降低(P<0.05)。结论:流感病毒感染对TLR7/8介导的IFN-α/β蛋白分泌水平有明显的上调作用,麻杏石甘汤能够下调流感病毒感染的巨噬细胞IFN-α/β分泌水平和蛋白表达水平,在作用48h时间点,麻黄先煎25min之麻杏石甘汤含药血清对IFN-α分泌水平的调节作用优于四药同煎。

摘要:
Objective: To systematically evaluate the cllinical efficacy of Duhuo Jisheng decoction (DHJSD) combined with non-surgical therapy on lumbar disc herniation (LDH). Methods: China Science and Technology Journal Database (VIP), Chinese National Knowledge Infrastructure (CNKI), WanFang Data, PubMed were searched and eligible randomized controlled clinical trials (RCTs) were included. Two reviewers evaluated the quality of the included RCTs and extracted data independently. Then, data analysis was performed with RevMetn 4.2 software. Results: A total of 18 RCTs were enrolled, including 1682 patients. Meta-analysis results showed that the overall response rate of DHJSD combined with non-operative therapy group (treatment group) in the treatment of LDH was better than that of the non-operative therapy group (control group) [OR = 4.29, 95% CI (3.19, 5.76), P < 0.001]. JOA score of the treatment group was significantly higher than that of control group [OR = 3.96, 95% CI (3.4, 4.52), P < 0.001]. VAS score was dramatically lower in the treatment group than the control group [OR = -1.58, 95% CI (-1.97, -1.18), P < 0.001]. Conclusion: The clinical efficacy of DHJSD combined with non-surgical therapy is better than that of non-surgical therapy alone. However, due to the low quality of the included literatures, large-scale and high-quality RCTs are still needed for further confirmation.

铁苋菜不同极性萃取物的抗氧化及抑菌活性研究

采用1,1-二苯基-2-三硝基苯肼(DPPH)法、β-胡萝卜素漂白法和清除一氧化氮自由基(NO)法来测定不同极性铁苋菜萃取物的抗氧化活性;通过微孔比浊法比较铁苋菜萃取物对3种革兰氏阴性细菌(大肠杆菌、铜绿假单胞菌和幽门螺杆菌)和4种革兰氏阳性细菌(表皮葡萄球菌、枯草芽孢杆菌、藤黄微球菌和金黄色葡萄球菌)的抑菌效果.结果表明,铁苋菜不同极性萃取物均有较好的清除DPPH、NO自由基和抑制β-胡萝卜素漂白的效果,浓度为2.0 mg/mL时,正丁醇萃取物清除DPPH自由基和抑制β-胡萝卜素漂白能力最佳,DPPH自由基清除率为94.60%,β-胡萝卜素抑制率为83.15%,乙酸乙酯萃取物清除NO自由基能力最强,清除率为82.72%.抑菌试验结果显示,铁苋菜各萃取物均有较好抑菌效果,对革兰氏阳性菌抑菌效果优于革兰氏阴性菌,其中石油醚萃取物对大肠杆菌、表皮葡萄球菌和金黄色葡萄球菌抑制作用最佳,MIC值分别为0.625,0.315,1.250 mg/mL.铁苋菜萃取物具有较好的抗氧化及抑菌效果.

目的研究从中药黄芩干燥根中提取分离药效成分黄芩苷体外对耐甲氧西林金黄色葡萄球菌活性的影响.方法以琼脂扩散法、液体稀释微孔板法检测黄芩苷对MRSA的抑菌环大小、最低抑菌浓度(MIC)和最低杀菌浓度(MBC) ;以96孔酶标板结晶紫法检测黄芩苷对MRSA的最小生物膜清除浓度(MBEC) .结果与DMSO比较,黄芩苷与氯霉素(阳性对照药物)对MRSA的抑菌环半径显著增加(P < 0. 01) ,与氯霉素比较,黄芩苷对MRSA的抑菌环半径显著缩小(P < 0. 05) ;黄芩苷对MRSA的MIC为0. 5mg /ml; MBC为1mg /ml;黄芩苷对MRSA的MBEC为0. 5mg /ml.结论黄芩苷对MRSA有一定的抗菌、抑菌作用,其效应机制可能与其抑制细菌生物膜的形成有关.

湖南省城市高血压病患者住院费用对比分析

摘要:
[目的]对比分析湖南省综合医院和中医医院城市高血压病患者住院费用及其影响因素,为控制高血压病患者住院费用提供依据.[方法]收集湖南省长沙、 岳阳、 永州、 衡阳综合医院和中医医院高血压病患者住院费用的基本构成情况,进行对比分析;采用多因素线性回归法对住院费用的影响因素进行分析.[结果]有效样本8,312例,其中中医医院样本4,695例(56%),综合医院样本3,617例(44%).住院费用的中位数分别为84,808.74元和6,968.405元.影响中医医院患者住院费用的主要因素是住院天数、 支付方式、 年龄;影响综合医院患者住院总费用的主要因素主要是支付方式和住院天数.[结论]综合医院的患者住院总费用大于中医医院的患者住院总费用;不同类型医院药品费占比均较高;住院费用随患者年龄的增加而增加;住院天数是影响住院费用增长的重要因素.

Objective: To explore the pharmacodynamic mechanism of Fu Zi Li Zhong Decoction (FZLZD) in treating gastric ulcer and its effect on gastrointestinal microecology. Methods: Rat gastric ulcer model was established by continuous vinegar gavage. The experiment included normal group, model group, ranitidine group and FZLZD group. The animals were handled after intragastricing administration of corresponding clinical isodose drugs for 7 d. The rats’ gastrointestinal histopathology was observed by HE staining. The expression of ZO-1 and Occludin mRNA in rat gastric tissue was detected by RT-PCR. The diversity of intestinal flora was analyzed by 16SrDNA sequencing. Results: FZLZD can effectively improve gastric mucosal destruction, epithelial exfoliation, inflammatory cell infiltration, inflammatory substance exudation and edema in rats with gastric ulcer. FZLZD can effectively up-regulate the expression levels of ZO-1 and Occludin mRNA in gastric tissue of rats with gastric ulcer. The results of intestinal flora diversity showed that the diversity of intestinal flora in FZLZD group was significantly higher than that of model group. Conclusions: FZLZD can regulate gastric ulcer by promoting the expression of ZO-1 and Occludin and improving the diversity of intestinal flora. © 2019 Digital Chinese Medicine

摘要:
目的 探讨基于p38 MAPK信号通路研究养阴润目丸治疗干眼兔的作用机制.方法 采用氢溴酸东莨菪碱皮下注射途径制备干眼兔模型,按随机数字表法分成模型组、p38抑制剂组、阳性药物(杞菊地黄丸)组和低、中、高剂量养阴润目丸组,同步设不造模的正常组,6只/组.各组按相应处理方式连续干预2周后,检测各组兔泪液分泌量、泪腺病理形态、泪腺组织凋亡细胞的表达及泪腺组织中p38 MAPK、Bax、Bcl-2蛋白和mRNA表达情况.结果 养阴润目丸组和阳性药物组能增加干眼兔模型泪液分泌量,抑制泪腺细胞的凋亡,下调p38 MAPK、Bax的蛋白和mRNA表达,上调Bcl-2的蛋白和mRNA表达水平,与模型组比较,差异均有统计学意义(P<0.05).结论 养阴润目丸能够通过纠正Bax/Bcl-2失衡,改善干眼泪腺细胞凋亡,进而减轻泪腺细胞损伤,恢复泪腺细胞的代谢功能,与调控p38 MAPK的关系极为密切,这可能是其治疗干眼的重要机制之一.

摘要:
随着实验教学在基础医学类教学中占据的位置越来越重要,在实验室各项制度不断完善的同时,实验室安全问题也越来越凸显。本文针对我校基础医学实验室的相关教学现状和存在的问题进行了归纳,发现人为因素和设备物品是影响实验室安全最主要的隐患,针对这些问题,我校对实验室的安全管理提出了一些措施,主要包括在废弃物的处理,试剂及设备的安全问题以及在教育上等几个方面,同时,我校积极引进了虚拟仿真实验平台,该平台的推广不仅提高了学生的学习效率,同时也减少了很多实验室安全问题,但目前其应用还不广泛,下一步希望扩大其应用,以实现效果最大化。

摘要:
中西医结合研究生培养作为医学研究生培养的重要组成部分,在医学教育研究领域占有重要地位。我校基于中西医结合研究生人才培养要求和课程体系特点,针对中西医结合学术型研究生开设医学基本实验技能训练课程。从课程内容设置、课程模式到考核方法、课程实践效果及课程评价进行了全新探索实践,以期提高研究生科研基本理论素养与科研能力,加强科研道德教育,探索出合理的中西医结合学术型研究生培养模式,切实提高中西医结合人才培养质量。

摘要:
目的获得玄参药材及饮片有机氯农药残留概貌,建立玄参有机氯农药残留的限量标准。方法采用快速气相色谱法检测全国范围内10个代表产地的玄参药材及7个产地(30批次)玄参饮片中21种有机氯农药残留量,考察99%置信区间,制定玄参有机氯农药残留的限量标准。结果玄参样品中21种有机氯农药残留量为0.000 1~0.077 7 μg/g,其中六六六、七氯、硫丹占农药残留总量的93%~96%,拟定了玄参中有机氯农药残留国家限量标准。结论玄参中有机氯农药残留主要为六六六、七氯、硫丹,制定的玄参有机氯农药残留限量标准为总有机氯≤0.700 μg/g。

Ethnopharmacological relevance: Buyang huanwu decoction (BYHWD) is a classic recipe in traditional Chinese medicine (TCM) to supplement Qi and activate blood. It has been used to recover the neural function after the injury of central nervous system for hundreds of years in China. Aim of the study: This study investigated whether Buyang huanwu decoction (BYHWD) combined with bone marrow mesenchymal stem cells (BMSCs) transplantation had synergistic effect on neuroprotection of red nucleus neurons after spinal cord injury (SCI). Materials and methods: Rubrospinal tract (RST) transection model was established and BMSCs were collected. The forelimb locomotor function was recorded using inclined plate test and spontaneous vertical exploration. cAMP level in red nucleus was detected with Enzyme-linked immunosorbent assay (ELISA). Morphology and number of red nucleus neurons was observed using Nissl's staining. Expression of cAMP-response element binding protein (CREB), ras homolog gene family member A (RhoA) and nerve growth factor (NGF) in red nucleus was detected using immunohistochemistry, qRT-PCR and Western-blotting. Results: The combination of BYHWD and BMSCs transplantation could improve the forelimb locomotor function significantly and give the red nucleus somas a better protection. Meanwhile, cAMP level, CREB and NGF increased, while RhoA decreased remarkably in the BYHWD+BMSCs group. Conclusions: BYHWD combined with BMSCs transplantation had synergistic effect on neuroprotection of red nucleus neurons after SCI; the mechanism may be related to up-regulating cAMP level, activating the cAMP/CREB/RhoA signaling pathway, and promoting expression of NGF. © 2018 The Authors
