Vine tea with strong antioxidant activity is commonly consumed as healthy tea/beverage. However, detailed information about its antioxidants is incomplete. Here, off-line hyphenation of heart-cutting high-speed countercurrent chromatography (HSCCC) with high performance liquid chromatography–diode array detector–quadrupole time-of-flight tandem mass spectrometry (HPLC–DAD–QTOF-MS/MS) were described for systematic profiling antioxidants in vine tea. At first, antioxidants were rapidly screened by 1,1-diphenyl-2-picryl-hydrazyl radical–high performance liquid chromatography (DPPH–HPLC). Subsequently, stepwise HSCCC using petroleum ether–ethyl acetate–methanol–water (4:9:4:9, v/v/v/v) and (4:9:5:8, v/v/v/v) as solvent systems was optimized to fractionate and enrich antioxidants from ethyl acetate fraction of vine tea. Finally, heart-cutting mode was used to collect five interesting HSCCC fractions for HPLC–DAD–QTOF-MS/MS analysis. Desirable orthogonality between HSCCC and HPLC led to identification of fifteen antioxidant flavonoids, while four minor flavonoids were first reported in vine tea. Results showed that the developed system is efficient to comprehensively explore antioxidants from complex natural herbs.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease that involves the degeneration and death of the nerve cells in brain and spinal cord that control voluntary muscle movement. This disease can cause patients struggling with a progressive loss of motor function while typically leaving cognitive functions intact. This paper presents a novel predication method that combines a dimension reduction (integrating partial least square into random frog algorithm) with support vector regression to predict the progression of ALS in the next 3–12 months according to the data collected from the patients over the latest three months. The experiment on the actual data from the PRO-ACT database indicates that the proposed method is effective and robust and can predict the clinical outcome by means of the slope of ALS progression, as measured using the ALS functional rating scale (ALSFRS) and the score used for monitoring ALS patients. Especially, the features selected can effectively distinguish the clinical outcome targets. It is of great benefit to aid clinical care, identify new disease predictors and potentially significantly reduce the costs of future ALS clinical trials.

α-Aryloxybenzyl analogues of morpholine are a significant class of compounds because of their influence on the central nervous system (CNS), with particular concentration on their antidepressant potency. (±)-Reboxetine, an example of such α-aryloxybenzyl analogues, is an orally active and selective noradrenaline reuptake inhibitor that is presently recognized as a prescription drug in over 60 countries for depressive sickness and has been spaciously studied for its pharmacological characteristics. (+)-(S,S)-Reboxetine is presently undergoing advanced clinical evaluation as a potential treatment for neuropathic and fibromyalgia pain. Scheming well-organized approaches to access reboxetine and its derivatives represents a significant endeavor not only for developing antidepressant drugs but also advancing medical studies by radiolabeling of reboxetine derivatives with 11C, 18F, or 123I as potential positron emission tomography radioligands for imaging the brain norepinephrine transporter system. Therefore, to fulfill the challenge of creating the reboxetine architecture by improved synthetic routes, the review combines all of the literature synthetic processes for reboxetine and its derivatives on one platform. Cons and pros of each synthetic method are discussed in this review, which will be very fruitful for the synthetic and medical communities to increase the diversity of synthetic procedures and to develop new concepts and perceptions.

Strokes are the leading cause of death and disability all over the world, however, there are few satisfactory therapies. IMM-H004 citrate (004), 7-hydroxy-5-methoxy-4-methyl-3-(4-methylpiperazin-1-yl)-coumarin, is a coumarin derivative which has potential therapeutic effects in brain ischemia with ambiguous mechanisms. We aim to study the anti-brain ischemia effect and mechanism of 004 in spontaneously hypertensive (SHR) rats. Adult male Wistar-Kyoto (WKY) rats and SHR rats were subjected to 24 h reperfusion after transient middle cerebral artery occlusion (tMCAO) for 1 h and were intravenously injected with 004 or Edaravone at the time of reperfusion. Behavioral scores, magnetic resonance imaging (MRI) and TTC staining were used to test the therapeutic effect of 004. To study the mechanism, the infiltration of leukocytes, the activation of microglia, blood viscosity, the expression of VCAM-1, MMPs and proteins involved in the MAPK/NF-κB pathway were researched. The results indicated that tMCAO/R induced serious injury, while 004 significantly alleviated the infarct volume and improved neurological deficits. 004 improved inflammatory processes, such as the enhancement of blood viscosity, expression of VCAM-1 and MMP2, release of TNF-α, IL-1β, IL-6 and IL-23, phosphorylation of JNK and p38 and translocation of NF-κB, which play crucial roles in brain I/R in SHR rats. An in vitro study also proved that 004 regulated JNK and NF-κB pathways and decreased the expression of VCAM-1, which eventually led to the suppression of neuroinflammation.

Heshouwu is a traditional non-toxic Chinese medicine commonly used in the clinical setting; however, clinical cases of Heshouwu-induced hepatotoxicity have been frequently reported. Pharmacoepidemiological studies have found that Heshouwu-induced liver injury occurs only in a small fraction of individuals taking the drug; these patients presented with the typical characteristics of idiosyncratic drug-induced liver injury(IDILI). Human IDILI can be reproduced in animals by drug coexposure to appropriate doses of lipopolysaccharide(LPS), which evokes modest inflammatory stress. Many drugs that cause human IDILI, including ranitidine and trovafloxacin, can also cause liver injury with a non-hepatotoxic dose in the LPS model.

A novel fluorescent probe, viz. PI-Cys, was developed on the basis of the excited state intramolecular proton transfer (ESIPT) mechanism for the detection of cysteine (Cys). It consists of an new ESIPT dye as the fluorophore, linked via a hydroxyl moiety to an acrylate group. The acrylate group acts as an ESIPT blocking agent as well as a functional recognition moiety. In the presence of Cys, the reaction of Cys with PI-Cys results in cleavage of the acrylate moiety from PI-Cys, thereby unleashing the ESIPT process and leading to about 20-fold enhancement of fluorescence. The probe exhibits high sensitivity and selectivity toward Cys over homocysteine and glutathione. Finally, the probe was successfully applied for fluorescence bioimaging of Cys in living cancer cells.

A novel ROI-based reversible data hiding scheme in encrypted medical images is proposed. Firstly, a content owner partitions an original medical image into the region of interest (ROI) and the region of noninterest (RONI), and then encrypts the image using an encryption key. A data-hider concatenates the least significant bits (LSB) of the encrypted ROI and Electronic Patient Record (EPR), and then embeds the concatenated data into the encrypted image by LSB substitution algorithm. With the encrypted medical image containing the embedded data, the receiver can extract the embedded data with the data-hiding key; if the receiver has the encryption key, a medical image similar to the original image can be obtained by directly decrypting the encrypted medical image; if the receiver has both the data-hiding key and the encryption key, the embedded data can be extracted without any error and ROI can be losslessly recovered after extracting the embedded data. The root of Wikstroemia indica has been widely used in China as folk medicine for the treatment for arthritis, whooping cough, cancer, and bacillosis. However, the constituents which have antibacterial activity were not clarified yet. In this study, the antibacterial effect of five extracts from W. indica on Escherichia coli was evaluated by microcalorimetry coupled with agar dilution method. The ethanol extract of W. indica was isolated with organic solvents of different polarities including petroleum (P.E.) extract, chloroform (CHCl3) extract, ethyl acetate (EtOAc) extract, n-butylalcohol (nBuOH) extract, and residue extract. The metabolic profiles of E. coli growth at 37 A degrees C were measured by microcalorimetry. According to the principal component analysis, k (1), k (2), and P (1) were obtained from heat flow power-time (HFP-time) curve. The agar dilution method was performed to verify the results of thermodynamics. The results of microcalorimetric experiment indicated that EtOAc fraction demonstrated the strongest antibacterial activity with half-inhibitory concentration of 92.4 mu g mL(-1). Meanwhile, similar results were gained from the common method of agar diffusion, which suggested that EtOAc extract could be further developed as antibacterial bioactive fraction of W. indica. Altogether, microcalorimetry is a useful technique to provide sufficient quantitative information and evaluate the antimicrobial effect with its sensitive.
