Objectives: Aim of this study is to explore whether quercetin can inhibit the enlarged fibrogenic responses of endometrial stromal cells by increasing the level of microRNA-145 (miR-145) and mediating the TGF-beta 1/Smad2/Smad3 signaling pathway, and to discuss the mechanism of signal transduction, further to provide experimental basis for revealing the pathophysiological mechanism and seeking new strategies for effective prevention and treatment of endometrial fibrosis. Methods: The expression levels of miR-145 and TGF-beta receptor 2 (TGFBR2) were detected by RT-qPCR analysis. Expressions of alpha-smooth muscle actin (alpha-SMA) and vimentin were examined by immunofluorescence staining. Cell viability was measured by MIT assay. The protein expression of collagen type 1 alpha 1 (Coll al), alpha-SMA, fibronectin (FN), TGFBR2, transforming growth factor (TGF-beta 1), Smad2/3, phospho-Smad2/3 (p-Smad2/3) were detected by western blot analysis. The interaction between miR-145 and TGFBR2 was confirmed by dual-luciferase reporter gene assay. Results: The expression level of miR-145 was decreased, whereas TGFBR2 was increased in intrauterine adhesion tissue. The expression levels of COL1A1, alpha-SMA, FN, TGFBR2, and p-Smad2/3 were increased, whereas miR-145 and cell proliferation were decreased in human endometrial stromal cells (hESCs) in response to TGF-beta 1 stimulation in a time and dose-dependent manner, which could be reversed by quercetin. Furthermore, quercetin regulates cell fibrogenic responses of endometrial stromal cells via miR-145/TGF-beta 1/Smad2/Smad3 pathway. Conclusions: These findings indicated that quercetin have a significant anti-fibrotic effect and could upregulate miR-145 and inhibit activation of TGF-beta 1/Smad2/Smad3 pathway to regulate TGF-beta 1 induced fibrogenic responses of endometrial stromal cells, which may serve as a potential therapeutic agent for endometrial fibrosis.

目的:探讨寿胎丸对复发性流产模型大鼠蜕膜细胞水通道蛋白(AQP)1、2表达的影响。方法:取复发性流产模型大鼠胚胎着床部位的蜕膜组织进行原代和传代培养,分别用寿胎丸含药血清、雌激素(E2)及孕激素(P)进行干预,另设空白对照组,4组药物对培养的蜕膜细胞作用72h后,通过q RT-PCR及Western blot方法检测蜕膜细胞中AQP1、AQP2 m RNA及蛋白的表达。结果:与空白对照组比较,寿胎丸组及P组AQP1、AQP2 m RNA及蛋白的表达均显著提高(P<0.05),E2组AQP1、AQP2 m RNA及AQP2蛋白的表达显著提高(P<0.01)。P比寿胎丸更显著上调AQP1 m RNA及蛋白的表达(P<0.01)。E2、P均较寿胎丸显著上调AQP2 m RNA的表达(P<0.01)。结论:寿胎丸可能通过上调复发性流产大鼠蜕膜组织AQP1、AQP2 m RNA及蛋白的表达水平,改善妊娠结局。寿胎丸对AQP蛋白的调节,说明"肾主水"与"肾主生殖"之间存在相关性。

Objective To visualize Professor YOU Zhaoling's academic research by means of bibliometric tools.Methods“尤昭玲”was taken as the key word in the author’s search in Chinese National Knowledge Infrastructure(CNKI).Visual analysis of the literature was carried out by CiteSpace.The author’s publication number,author’s co-operative network,author cluster analysis,keyword co-occurrence network,keyword emergent words and keyword clustering were analyzed respectively.Results Professor YOU Zhaoling's main research directions were polycystic ovary syndrome,traditional Chinese medicine(TCM)supplemented treatment of in vitro fertilization-embryo transfer,infertility and premature ovarian failure.Professor YOU original created"YOU’s Eight Methods"and"Three Phases of Type Two Assisted Pregnancy"to treat polycystic ovarian syndrome,formulated"Six Phases and Seven Steps"TCM-assisted IVF-ET technique,and created"Iceberg Theory"and"Looking at the Eye to Identify the Ovary Method"to guide the treatment of premature ovarian failure.Conclusion Professor YOU,with the help of modern physiological and pathological technology and classical theory of TCM,explains the occurrence and development of diseases,and creatively combines traditional Chinese and western medicine to guide and treat gynecological diseases,thus forming a distinctive"YOU’s"diagnosis and treatment,which provides new ideas and methods for the modernization of TCM.
