A rapid, accurate, and sensitive ultra-high performance liquid chromatography (UHPLC) method was established for simultaneously detecting naringin, hesperidin, neohesperidin, meranzin hydrate, naringenin, and hesperetin in Fructus aurantii (FA) decoction. Analysis was performed on Waters BEH (R) C18 (50 mm x 2.1 mm, 1.7 mu m) at a flow rate of 0.2 mL/min by using (A) acetonitrile and (B) 0.5% acetic acid-water as the mobile phase. The method was well validated on linearity, precision, recoveries, and stability. Then, we used the same UHPLC conditions for quantitative analysis of meranzin hydrate, naringenin, and hesperetin in rat plasma. The method proved to be linear within the concentration ranges of 3.3-3300 ng/mL for meranzin hydrate, 6.95-3555 ng/mL for naringenin, and 1.8-236 ng/mL for hesperetin. The RSD of precision ranged from 1.22% to 9.08%, and the average extraction recovery ranged from 96.49 t 1.42% to 102.01 t 3.16%. Besides, we performed a comparative pharmacokinetic study after oral administration of FA decoction at a low dose of 15 g/kg and high dose of 30 g/kg body weight for seven days to rats. The AUC((0-t)) and C-max of meranzin hydrate, naringenin, and hesperetin were multiplied significantly with the increase of FA dosage, and the t(1/2) of meranzin hydrate was faster than naringenin and hesperetin in the two groups.

Objective To assess the environment of Hunan production base of lilium lancifolium by the index of the soil,irrigation water and air quality.Methods The content distribution characteristics of cadmium(Cd),chromium(Cr),mercury(Hg),arsenic(As),plumbum(Pb)and cuprum(Cu)in Longshan region soils and irrigation water of ilium lancifolium base in Hunan province were analyzed.The six basic indexes of SO2,NO2,CO,O3,PM10 and PM2.5 in the atmospheric environment of Longshan lilium lancifolium base were determined.The environmental quality in research area was assessed by using single pollution index method,comprehensive pollution index method and potential ecological risk index.Results The content of Cd,chromium Cr,Hg,As,Pb and Cu of soil were in the range of 0.16~0.29,2.30~6.63,0.08~0.30,10.33~26.44,2.00~11.60,10.60~36.44 mg/kg;the contents of Pb,Cd,Cr,Cu,As and Hg in the irrigated area were 0.000 2~0.002 0袁0.000 2~0.000 9袁not detected,0~0.000 1,0.000 3~0.001 0 and 0.000 1~0.000 5 mg/L.The annual average values of SO2,NO2,PM10 and PM2.5 in the local atmospheric environment were 16,13,60 and 34 g/m3,respectively.The average value of CO(24 hours)was 0.9 mg/m3,and the maximum 8 hours for O3 was 94滋g/m3.According to the comprehensive pollution index and potential ecological risk index method,the average pollution level in Longshan area is in the security level,the pollution level is clean level,and the ecological risk grade is grade A(low).Conclusion Longshan has excellent production environment suitable for the growth of lilium lancifolium.However,the Pi of Cd,Hg and As in some soil samples are more than 1,or close to the critical value of 1,which has some potential ecological risk.

Acute pancreatitis (AP) is defined as an acute inflammation of pancreas that may cause damage to other tissues and organs depending upon the severity of symptoms. The diagnosis of AP is usually made by detection of raised circulating pancreatic enzyme levels, but there are occasional false positive and false negative diagnoses and such tests are often normal in delayed presentations. More accurate biomarkers would help in such situations. In this study, the global metabolites' changes of AP patients (APP) were profiled by using gas chromatography-mass spectrometry (GC-MS). Multivariate pattern recognition techniques were used to establish the classification models to distinguish APP from healthy participants (HP). Some significant metabolites including 3-hydroxybutyric acid, phosphoric acid, glycerol, citric acid, D-galactose, D-mannose, D-glucose, hexadecanoic acid and serotonin were selected as potential biomarkers for helping clinical diagnosis of Ap. Furthermore, the metabolite changes in APP with severe and mild symptoms were also analyzed. Based on the selected biomarkers, some relevant pathways were also identified. Our results suggested that GC-MS based serum metabolomics method can be used in the clinical diagnosis of AP by profiling potential biomarkers. (C) 2017 LAP and EPC. Published by Elsevier B.V. IMM-H004, a 3-piperazinylcoumarin compound derived from coumarin, has been proved effective against CA1 cell loss and spatial learning impairments resulting from transient global ischemia/reperfusion (TGCI/R), while the mechanism is still largely unknown. Here, we confirmed that treatment of rats with IMM-H004 immediately after TGCI/R ameliorated delayed neuronal death (DND) in the CA1 of hippocampus and cortex. Further study suggested that IMM-H004 contributed to the expression of antiapoptotic protein survivin through the activation of PI3K-dependent protein kinase B (PKB/Akt), which led to the phosphorylation of forkhead box O1 (FoxO1), then relieved the inhibiting effect on survivin promoter. Additionally, IMM-H004 also enhanced the expression of hepatitis B X-interacting protein (HBXIP), which formed a complex with survivin to prevent the activation of caspase death cascade, thereby halting apoptotic cell death. Finally, we injected a HBXIP siRNA into hippocampus and performed microelectroporation before ischemia/reperfusion, which abolished the protective effect of IMM-H004. Further study revealed that HBXIP maintained the high expression of Akt and survivin. Collectively, our findings demonstrated that DND after TGCI/R was alleviated by IMM-H004 through promoting the formation of survivin-HBXIP complex, which further emphasized the importance of endogenous protein involved in self-repair after stroke.
