Expression, immune infiltration and clinical significance of SPAG5 in hepatocellular carcinoma: A gene expression-based study

Background: Overexpression of sperm-associated antigen 5 (SPAG5) is a marker of poor prognosis in numerous tumors and is recognized as an index of tumor proliferation; however, its expression in liver cancer remains unclear. Methods: The Oncomine (https://www.oncomine.org) and Timer (https://cistrome.shinyapps.io/timer) databases were used to analyze the expression of SPAG5 in liver hepatocellular carcinoma (HCC) and normal liver tissues. The relationship between the expression of SPAG5 and immune infiltration of HCC was investigated using the Timer and GEPIA (http://gepia.cancer-pku.cn) databases, and the mechanism was analyzed using Gene Set Enrichment Analysis. A Kaplan–Meier Plotter (http://kmplot.com/analysis) was used to evaluate the effect of SPAG5 on the prognosis of patients with HCC. Results: The results revealed that the SPAG5 expression level was positively correlated with the infiltration levels of CD8+ T cells, macrophages, neutrophils, and especially B cells and dendritic cells. In addition, SPAG5 expression was significantly associated with T cell exhaustion. The overall survival time, progression-free survival time, recurrence-free survival time and disease-specific survival time were significantly reduced for HCC patients with high SPAG5 expression (p < 0.01) and high expression of SPAG5 was significantly associated with a poor overall survival time and progression-free survival time of grade and stage II–III HCC (p < 0.05) but not with stage I HCC (p > 0.05). Additionally, the expression of SPAG5 is related to the p53 and cell cycle signal pathways. Conclusions: In conclusion, SPAG5 is not only a marker of immune infiltration and poor prognosis, but also a potential therapeutic target for liver cancer. © 2019 John Wiley & Sons, Ltd.

Introduction and Objective: At present, there is no hematology marker with high specificity to the diagnosis and differential diagnosis of acute ischemic stroke (AIS). How to use the existing test items to improve the diagnosis efficiency worthy of discussion. D-Dimer (DD) and fibrinogen (FIB) were the common indicators in thrombotic diseases, but D-dimer to fibrinogen ratio (D/F) in AIS has not been used in clinical practice. In this work, we focus on the evaluation of D/F. Methods: 90 AIS patients were selected as the observation group and 65 other patients without coagulation function disorder as the control group. Meanwhile, a total of 33 patients with other diseases with impaired consciousness in the same period were collected. Based on the AIS patients with or without consciousness disorder divided it into consciousness disorder group and unconsciousness disorder group. Then based on the patients with or without consciousness disorder divided it into other diseases with unconsciousness disorder group and Other diseases with lacunar cerebral infarction (LCI)and disturbance of consciousness group. then compare the differences of plasma DD, FIB and D/F between groups. Results: All plasma DD, FIB and D/F ratio in AIS patients were significantly higher than in other disease group (P = 0.000, P = 0.001, P = 0.000), but DD, D/F in disorders of consciousness group was significantly higher than in unconsciousness disorders group (P = 0.007, P = 0.005). The DD of the AIS with consciousness disorder group were significantly higher than that of the other disease with consciousness disorder group (P = 0.042), and the DD, D/F ratio of Other diseases with lacunar cerebral infarction and disturbance of consciousness group were significantly higher than one(P = 0.000, P = 0.003). All others are undifferentiated. Conclusions: When DD, D/F ratio is high, other diseases caused by consciousness disorders are likely to be combined with infarcts, which can be used for the diagnosis and differential diagnosis of patients with different types of consciousness disorders, especially hospitalized patients. © 2020 Elsevier Inc.

Sperm-associated antigen 9 (SPAG9) is a biomarker and potential therapeutic target for several cancers; however, its involvement in liver cancer progression is not clear. The aim of the present study was to determine whether SPAG9 regulates proliferation of liver cancer. Immunohistochemistry and cell immunofluorescence were used to confirm the expression and the localization of SPAG9 in human liver cancer tissues and the liver cancer-derived HepG2 cells. A small interfering RNA (siRNA) designed to target SPAG9 was transiently transfected into HepG2 cells using Lipofectamine™ 2000, and proliferation, apoptosis and cell cycle progression were analyzed using CCK‑8 assay and flow cytometry; western blotting was used to detect the expression of SPAG9, JNK, p38, MKK3 and MKK6, and co-immunoprecipitation was used to assess the interaction between SPAG9 and JNK. SPAG9 was overexpressed in 16 out of 20 (80%) patients with liver cancer. The protein was localized in both the cytoplasm and nucleus of liver cancer cells obtained from patients and in HepG2 cells. Depletion of SPAG9 inhibited the proliferation of HepG2 cells, promoted apoptosis and arrested the cell cycle at the S phase. Moreover, cells deficient in SPAG9 had decreased expression of JNK, p38 and MKK3 compared to HepG2 cells not treated with an siRNA targeting SPAG9. In the present study, SPAG9 was revealed to regulate cell proliferation, apoptosis and cell cycle progression in liver cancer cells through the SPAG9/MKK3/p38 axis. There are different views of how the immune system participates in the reaction to cancer. Here, we evaluated expression of DAMP proteins HSP70 and cancer-testis antigen SPAG9 in patients with hepatocellular carcinoma(HCC) and lung cancer to explore tumor immunity. Our analysis showed that levels of HSP70 and SPAG9 antibody were significantly higher in serum of lung cancer and HCC patients than in serum from healthy subjects(P < 0.001), but there were no differences in levels of HSP70 antibody in patients and controls. Levels of serum SPAG9 antibody in newly diagnosed lung cancer patients were significantly higher than in treated lung cancer patients(P < 0.05), but there were no differences in levels of HSP70 or HSP70 antibody. Levels of serum HSP70 and SPAG9 antibody, but not HSP70 antibody, were also higher in hepatitis/cirrhosis patients than in healthy subjects(P = 0.005, P < 0.001). Levels of serum SPAG9 antibody were significantly higher in HCC patients than in hepatitis/cirrhosis patients, but there were no differences in HSP70 or HSP70 antibody levels. Finally, levels of serum HSP70 and SPAG9 antibody were significantly higher in HCC patients than in lung cancer patients(P < 0.05, P < 0.001). These results indicate that cancer-testis antigen SPAG9 induces a strong humoral immune response in cancer patients but HSP70 does not. These results show that SPAG9 has potential as a tumor-specific biomarker.

摘要:
目的 探讨可溶性模式识别受体血清淀粉样蛋白A(SAA)及血清C-反应蛋白(CRP)水平在非感染性疾病中的诊断价值,并绘制受试者工作特征(R O C曲线)找寻最佳临界值来辅助鉴别感染性疾病与非感染性疾病,为临床诊断提供依据.方法 选择在湖南省第二人民医院住院部的91例非感染性疾病患者为非感染性疾病组.其中急性心肌梗死30例,急性胰腺炎30例,急性创伤31例.同时纳入感染组60例,健康对照组30例.通过检测以上3组的血清SAA、CRP水平,进行统计学分析.结果 (1)非感染性疾病组、感染组与对照组血清CRP水平之间差异有统计学意义(χ2=52.448,P<0.001),血清SAA水平差异有统计学意义(χ2=43.993,P<0.001);(2)在非感染性疾病组中,各组与健康对照组间SAA、CRP水平差异有统计学意义(χ2=53.534,P<0.001;χ2=66.504,P<0.001).S A A组间两两比较发现,急性心肌梗死、急性胰腺炎、急性创伤组与对照组比较差异有统计学意义(Z=-4.547、-6.409、-6.708,P=0.000),其中急性胰腺炎组与急性创伤组之间比较差异无统计学意义(Z=-0.15,P=0.88);CRP组间两两比较发现,急性心肌梗死组、急性胰腺炎组、急性创伤组与对照组比较差异有统计学意义(Z=-5.005、-5.287、-5.914,P=0.000),其中急性胰腺炎组与急性创伤组比较差异无统计学意义(Z=-0.15,P=0.286);(3)spearman相关分析可得出,血清SAA与CRP有一定相关性(r=0.6494,P<0.0001);(4)SAA、CRP单项检测诊断感染与非感染性疾病组的曲线下面积(AUC)分别为0.759、0.747;临界值分别为97.3 mg/L、42.15 mg/L;SAA、CRP联合检测诊断感染与非感染性疾病的AUC为0.765.结论 血清中可溶性识别受体SAA、CRP在非感染性疾病中有较高表达,对于鉴别感染与非感染性疾病组有一定诊断效能,可以作为疾病分类的标志物.

siRNA靶向抑制SPAG9对HepG2细胞增殖、凋亡及JNK信号通路的影响

目的探讨精子相关抗原9(SPAG 9)在肝癌细胞HepG 2中表达对HepG 2增殖分化及凋亡的影响,并初步揭示SPAG 9与JNK信号通路的关系。方法采用细胞免疫荧光检测SPAG 9在HepG 2中的表达及定位,siRNA技术敲低SPAG 9的表达,Western blot法检测转染后SPAG 9及JNK蛋白的表达,CCK8(cell counting kit-8)实验检测肿瘤细胞的增殖能力,流式细胞术检测肿瘤细胞的周期及凋亡。结果SPAG 9在HepG 2中表达,定位于细胞质和细胞核。与对照组细胞比较,转染SPAG 9 siRNA使JNK蛋白的表达下调,差异有统计学意义(P<0.05)。与对照组比较,转染SPAG 9 siRNA的HepG 2增殖能力显著降低(P<0.01),细胞凋亡显著增加(P<0.01),细胞周期阻滞于S期。结论HepG 2细胞表达SPAG 9蛋白,定位于细胞质和细胞核;SPAG 9促进HepG 2细胞增殖分化、抑制凋亡与JNK信号通路有关;敲低SPAG 9的表达使HepG 2细胞增殖阻滞于S期。

目的 初步探讨姜黄素及姜黄提取物对血栓模型大鼠凝血功能的影响,为进一步研究其作用机制做前期准备.方法 分别用姜黄素及姜黄提取物灌胃14 d后,采用物理方法建立大鼠血栓模型,获取各组大鼠血浆,全自动凝血仪检测活化部分凝血活酶时间(APTT)、纤维蛋白原(FIB)、凝血酶时间(TT)及抗凝血酶3(AT3).结果 血栓模型鼠血栓形成后,血浆APTT显著延长(P=0.011);姜黄素低、高剂量灌胃处理的血栓模型鼠血浆APTT缩短(P=0.02,P<0.001),而姜黄提取物处理的血栓模型鼠血浆APTT进一步延长(P<0.001,P=0.019);血栓模型鼠血栓形成后血浆FIB显著升高(P<0.001),姜黄素及姜黄提取物低、高剂量对血浆FIB没有显著影响;姜黄素灌胃缩短T T时间(P=0.03,P=0.001),而姜黄提取物没有此作用;姜黄素有降低A T3的作用,而姜黄提取物有升高A T3的作用.结论 姜黄素没有抗凝作用,而姜黄提取物则表现出较强的抗凝作用,提示姜黄提取物的活血化瘀机制与抗凝作用有关,而姜黄素的作用机制有待进一步探讨.

Inhibin B (INHBB), a heterodimer of a common α-subunit and a βB-subunit, is a glycoprotein belonging to the transforming growth factor-β (TGF-β) family. In this study, we observed INHBB expression was reduced in nasopharyngeal carcinoma (NPC) tissues compared to non-tumor nasopharyngeal epithelium tissues, and INHBB was associated with lymph node metastasis, stage of disease, and clinical progress. Positive expression of INHBB in NPC predicted a better prognosis (overall survival, P=0.038). However, the molecular mechanisms of INHBB have not been addressed in NPC. We induced anoikis-resistant cells in NPC cell lines under anchorage-independent conditions, then found epithelial-mesenchymal transition markers changed, cell apoptosis decreased, cell cycle was modified, and invasion strengthened in anoikis-resistant NPC cells. Theseanoikis-resistant NPCcellsshowed decreased expression of INHBB compared with adhesioncells. Furthermore, INHBB was found to influence the above-mentioned changes. In the anoikis-resistant NPC cells with INHBB overexpression, apoptotic cells increased, S phase cells weakened, vimentin, matrix metallopeptidase-9, and vascular endothelial growth factor A expression were downregulated, and E-cadherin expression was upregulated, and vice versa in knockdown of INHBB (INHBB shRNA) anoikis-resistant NPC cells. Diminished INHBB expression could activate the TGF-β pathway to phosphorylate Smad2/3 and form complexes in the nucleus, which resulted in the above changes. Thus, our results revealed for the first time that INHBB could suppress anoikis resistance and migration of NPC cells by the TGF-β signaling pathway, decrease p53 overexpression, and could serve as a potential biomarker for NPC metastasis and prognosis as well as a therapeutic application. © 2018 The Authors. 摘要:
目的 以姜黄提取物为参照研究姜黄素对大鼠血栓模型的影响及其与损伤相关模式分子(DAM P)热休克蛋白70(HSP70)的关系.方法 分别对大鼠进行姜黄素及姜黄提取物灌胃14 d后用物理方法建立大鼠血栓模型,以假手术组、蒸馏水灌胃血栓模型组(血栓模型组)大鼠为对照,获取各组大鼠血液标本和血管组织,采用ELISA法检测各组大鼠血浆HSP70水平,采用免疫组化法检测各组大鼠血管组织中HSP70的表达.结果 血栓模型组大鼠血浆H S P70水平显著高于假手术组大鼠(P=0.003),姜黄素低、高剂量灌胃血栓模型组大鼠血浆HSP70均显著降低(P=0.005、0.002),而姜黄提取物灌胃血栓模型组大鼠血浆HSP70水平无明显变化;与假手术组大鼠相比,血栓模型组大鼠血管组织中H S P70表达增强,姜黄素灌胃使血栓模型大鼠血管组织中H S P70表达增强,而姜黄提取物没有此作用.结论 姜黄素抗炎作用与增加细胞内H S P70表达,降低细胞外H S P70有关,姜黄提取物的抗炎作用可能是通过其他机制.

目的 评价国产乳胶增强免疫比浊法降钙素原(PCT)诊断试剂在西门子ADVIA 2400系统上应用的性能.方法 参考美国临床实验室标准化委员会(CLSI)指南对乳胶增强免疫比浊法PCT诊断试剂在西门子ADVIA 2400系统检测的精密度、准确度及线性范围进行性能验证,同时与参考方法(电化学发光法)进行一致性比较.结果 乳胶增强免疫比浊法在西门子ADVIA 2400系统上测定PCT低值和高值质控品的批内精密度分别为5.5% 、2.3%,批间精密度分别为6.6% 、3.1%,总精密度分别为6.6% 、3.9%.PCT浓度在0.19~49 ng/mL范围的线性良好(r=0.9957).乳胶增强比浊法与电化学发光法的Passing-Bablok回归方程为Y=0.846 X+0.0772(r=0.9894).两种方法在cut-off值分别为0.5、2.0、10.0 ng/mL时的Kappa值分别为0.742、0.893、0.921,西格玛(σ)值为3.5,两方法在低、高PCT浓度处的质量目标指数(QGI)分别为0.68、2.41.结论 国产乳胶增强免疫比浊PCT试剂在ADVIA-2400系统上应用符合性能验证标准,但采用六西格玛标准,该方法在精密度和准确度方面尚需进一步改进.

Cancer testis antigen sperm-associated antigen 9 (SPAG9) is highly expressed in many types of cancers. In the present study, to obtain a better understanding of the relevance of SPAG9 in cancer diagnosis and treatment, the expression of SPAG9 mRNA and protein in lung cancer specimens was evaluated by RT-PCR, western blotting and immunohistochemistry. ELISA was used to quantify the SPAG9 autoantibody in the peripheral blood of lung cancer patients. The results showed that the expression of SPAG9 mRNA and protein in the lung cancer tissues was significantly higher than that in the adjacent non-cancerous tissues (P<0.01). The level of the SPAG9 autoantibody in the serum of lung cancer patients was significantly higher than the level in the healthy controls (P<0.001), and the level of the SPAG9 autoantibody in the serum of untreated patients was significantly higher than that in treated patients (P=0.002). SPAG9 IgG antibody levels were significantly lower in treated adenocarcinoma and small cell lung cancer patients than these levels in the untreated patients (P=0.006, P=0.026, respectively), while no statistical difference was found between treated and untreated squamous cell carcinoma patients. Our results suggest that the SPAG9 antibody in serum is a promising marker for the diagnosis of lung cancer, and the level of the humoral immune response to this antigen appears to be related to the type of lung cancer.

Objective: To study changes in blood lipid metabolism in sepsis patients, especially high-density lipoprotein cholesterol (HDL-C) changes in the diagnosis of sepsis and the type of bacteria involved. Methods: Two-hundred-twenty cases of patients with febrile infections were divided into local infection, systemic inflammatory response syndrome or sepsis (sepsis) group. For controls, 81 cases of patients with a healthy check-up were used. Lipid levels and inflammatory state were supervised, and a comparative analysis of patients admitted to the hospital after 1, 5, 10 days was performed. Results: In patients with sepsis, total cholesterol, HDL-C, and apolipoprotein A 1 (apoA 1) were significantly decreased in this group. Particularly HDL-C was decreased 1 day after admission. Compared with the patients with gram-positive sepsis, HDL-C and apoA1 were significantly reduced in the patients with gram-negative sepsis at admission. The 24-h change ratio of HDL-C was different between the gram-negative and gram-positive sepsis patients with a 70.5 % specificity and 76.5 % sensitivity. The area under the curve was 0.744, and the critical value was −21.1 %. Conclusions: The sepsis patients had lower HDL-C than the other groups. The 24-h change ratio of HDL-C can be used as a sepsis diagnosis maker and to distinguish between the bacteria involved in sepsis. © 2016, The Author(s).

目的:监测脓毒症患者炎症参数的变化,尤其是髓过氧化物指数(M PXI)的改变对脓毒症的诊断及其细菌类型的区分。方法183例细菌感染患者分为局部感染组、系统性炎性反应综合征组(SIRS )、脓毒症组,54例健康体检者为对照组。测定患者入院时、第1、5、10天炎症情况并作比较分析。结果入院时患者炎症状态明显不同,局部感染和脓毒症患者,M PXI均降低;其他患者炎症指标均升高,差异具有统计学意义(P<0.05);脓毒症患者及SIRS患者MPXI从第1天开始就发生变化,炎症患者降钙素原(PCT )从第5天开始升高;入院时M PXI预示革兰阴性脓毒症感染特异度为62.1%,灵敏度为70.3%,-2.1为最佳临界点。结论脓毒症患者M PXI水平降低,其改变可作为脓毒症诊断及感染细菌类型的区分。

Introduction: Plasma cell-free DNA (cfDNA) of trauma patients has been widely investigated, but it has not been resolved whether cfDNA can be used as a non-invasive, rapid and sensitive marker of injury. Here we evaluated serum cfDNA in patients after injury and assessed the relationship between cfDNA levels and clinical prognosis. Methods: Fifty-six trauma patients formed three groups (minor, moderate and severe) according to Injury Severity Score (ISS), 33 of these cases were also divided according to microbiological and clinical evidence of infection. Plasma cfDNA and other indices were measured 1-6 h, 24-36 h and 60-90 h after injury. Results: The severe and moderate injury groups showed significantly higher positive percentage cfDNA than in the control (p < 0.001, p = 0.004). Positive plasma cfDNA samples were higher 1-6 h after injury than 24-48 h and 60-90 h, and the mean plasma cfDNA concentrations at 60-90 h were higher than that at the earlier two time points but not significantly. The ISS of the infected group was significantly higher than in the non-infected group (p = 0.02). cfDNA was found in 48.5% penetrating trauma samples, significantly higher than that in blunt injury (17.4%) (p = 0.024). Conclusions: Plasma cfDNA is a potential marker for trauma prognosis especially death, after severe injury, but its sensitivity presents limitations for clinical use. No evidence was found to relate plasma cfDNA to infection. Circulating cfDNA seems to be connected with injury type and mode, open wounds and surgical operations, which may be the primary reasons for plasma cfDNA increase. (c) 2013 Elsevier B.V. 摘要:
目的研究急性冠状动脉综合征(ACS)患者血液中性粒细胞髓过氧化物酶活性指数(MPXI)的变化及临床应用。方法选择2011年5~8月该院收治的ACS患者57例。其中,不稳定型心绞痛(UAP)20例为UAP组。非ST段抬高心肌梗死(NSTEMI)20例为NSTEMI组。ST段抬高型心肌梗死(STEMI)17例为STEMI组。选取同期该院体检中心健康体检者20例为对照组。比较4组MPXI、心肌肌钙蛋白I(cTnI)等指标的结果。结果对照组、UAP组、NSTEMI组血液MPXI、白细胞(WBC)、中性粒细胞(NEUT)、中性粒细胞比值(NEUT%)低于STEMI组(P<0.05)。结论常规血细胞分析能快捷方便检测出血液MPXI结果,可作为ACS疾病粥样斑块不稳定性的标志物之一,有利于ACS疾病的早期诊断与监测。
