Purpose: Ulcerative colitis and colitis-associated colorectal cancer (CAC) is a serious health issue, but etiopathological factors remain unclear. Aldo-keto reductase 1B10 (AKR1B10) is specifically expressed in the colonic epithelium, but downregulated in colorectal cancer. This study was aimed to investigate the etiopathogenic role of AKR1B10 in ulcerative colitis and CAC. Experimental Design: Ulcerative colitis and CAC biopsies (paraffin-embedded sections) and frozen tissues were collected to examine AKR1B10 expression. Aldo-keto reductase 1B8 (the ortholog of human AKR1B10) knockout (AKR1B8-/-) mice were produced to estimate its role in the susceptibility and severity of chronic colitis and associated dysplastic lesions, induced by dextran sulfate sodium (DSS) at a low dose (2%). Genome-wide exome sequencing was used to profile DNA damage in DSS-induced colitis and tumors. Results: AKR1B10 expression was markedly diminished in over 90% of ulcerative colitis and CAC tissues. AKR1B8 deficiency led to reduced lipid synthesis from butyrate and diminished proliferation of colonic epithelial cells. The DSS-treated AKR1B8-/- mice demonstrated impaired injury repair of colonic epithelium and more severe bleeding, in flammation, and ulceration. These AKR1B8-/- mice had more severe oxidative stress and DNA damage, and dysplasias were more frequent and at a higher grade in the AKR1B8-/- mice than in wild-type mice. Palpable masses were seen in the AKR1B8-/- mice only, not in wild-type. Conclusions: AKR1B8 is a critical protein in the proliferation and injury repair of the colonic epithelium and in the pathogenesis of ulcerative colitis and CAC, being a new etiopathogenic factor of these diseases. ©2014 AACR.

摘要:
目的研究加味二陈汤对Apo E-/-小鼠血清、主动脉壁及瓣膜的IL-6、TNF-α表达的影响。方法40只Apo E-/-小鼠中10只为空白对照组,30只以高脂饲喂养4周构建动脉粥样硬化(As)小鼠模型,均分成模型组、辛伐他汀组、加味二陈汤组,分别用生理盐水、辛伐他汀及加味二陈汤对小鼠灌胃给药,8周后处死小鼠,Elisa法检测血清中IL-6、TNF-α的含量;免疫组化法检测IL-6、TNF-α在主动脉壁及瓣膜的表达。结果辛伐他汀组血清IL-6和TNF-α分别为392.06±37.70 pg/m L、189.1±50.4 pg/m L,与模型组相比表达下降(P<0.05);加味二陈汤组血清IL-6、TNF-α分别为442.41±172.26 pg/m L、47.2±13.3 pg/m L,与模型组相比表达下降(P<0.05),与辛伐他汀组相比,表达下降(P<0.05);免疫组化结果显示加味二陈汤可降低IL-6、TNF-α在主动脉壁内的表达。结论加味二陈汤可降低血清和主动脉壁及瓣膜的IL-6、TNF-α的表达。

摘要:
目的探讨乙酰水杨酸姜黄素酯对动脉粥样硬化病变的作用及机制。方法 40只7周龄Apo E-/-小鼠,随机分为对照组、高脂组、辛伐他汀组[5 mg/(kg·d)]、乙酰水杨酸姜黄素酯低剂量组[23.8 mg/(kg·d)]和乙酰水杨酸姜黄素酯高剂量组[71.5 mg/(kg·d)],其中对照组给予普通饲料,其他组给予高脂饲料喂养。连续干预4周之后处死动物,检测血清中血脂水平;油红O染色法观察小鼠主动脉斑块;ELISA法或免疫组化法检测血清和肝脏炎症因子TNF-α、IL-6水平;Western blot检测小鼠肝脏中小凹蛋白1和NFκB p65的表达。结果与对照组相比,高脂组血清中TC和LDL-C升高,HDL-C降低,主动脉粥样斑块增多。与高脂组相比,乙酰水杨酸姜黄素酯组小鼠血清中的TC和LDL-C降低,肝脏脂质减少,血清和肝脏炎症因子水平下降,而主动脉粥样硬化斑块减轻;肝脏小凹蛋白1水平升高而NFκB p65水平降低。结论乙酰水杨酸姜黄素酯能抑制高脂喂养所致的Apo E-/-小鼠动脉硬化斑块的形成;机制可能与增加肝脏小凹蛋白1、减少NFκB p65蛋白水平,调节肝脏脂质代谢及炎症反应有关。

Background/Objectives:Epidemics of obesity and diabetes are escalating. High-calorie/high-fat food is a major cause for these global health issues, but molecular mechanisms underlying high-fat, diet-induced obesity are still not well understood. The aryl hydrocarbon receptor (AhR), a transcription factor that acts as a xenobiotic sensor, mediates environmental toxicant-induced obesity, insulin resistance and development of diabetes. AhR also influences lipid metabolism and diet-induced obesity. The effects of AhR deficiency on diet-induced obesity, hepatic steatosis and insulin resistance were examined.Methods:Male wild-type (WT), AhR null (AhR -/-) and AhR heterozygote (AhR +/-) mice were fed a normal chow diet (NCD, 10% kcal from fat) or a high-fat diet (HFD, 60% kcal from fat) for up to 14 weeks. Adiposity, adipose and liver morphology, insulin signaling, metabolic parameters and gene profiles were assessed.Results:AhR deficiency protected against HFD-induced obesity, hepatic steatosis, insulin resistance and inflammation. Moreover, AhR deficiency preserved insulin signaling in major metabolic tissues. These protective effects result from a higher energy expenditure in AhR-deficient mice compared with WT. Levels of transcript for both the thermogenic gene, uncoupling protein 1 (Ucp1), in brown adipose tissue and mitochondrial β-oxidation genes in muscle were significantly higher in AhR -/- and AhR +/- mice compared with WT.Conclusions:This work documents a physiologically relevant function for AhR in regulation of body weight, hepatic fat deposition, insulin sensitivity and energy expenditure under HFD exposure, suggesting that AhR signaling may be developed as a potential therapeutic target for treatment of obesity and metabolic disorders. © 2015 Macmillan Publishers Limited. 摘要:
本发明涉及中草药的提取领域,特别涉及一种从夏枯草中同时提取四种化合物的方法及应用。本发明利用粗分和细分相结合的方法,能够从夏枯草中同时提取出异迷迭香酸苷、迷迭香酸、异迷迭香酸苷甲酯、迷迭香酸甲酯四种化合物。本发明提供的方法样品在粗分时,分离快,所用时间短,成分交叉少且又不拖尾;在细分时,能减少分离步骤,同时能得到高产量和高纯度的目标化合物。本发明还提供了利用上述四种化合物混合后得到的夏枯草提取物混合物及其在抗肿瘤药物中的应用。

用高保真DNA聚合酶介导的PCR检测β地中海贫血热点突变

摘要:
目的 用高保真DNA聚合酶介导的PCR检测β地中海贫血基因热点突变.方法 选取已知中国人群β珠蛋白基因CD41-42、IVS-2nt654、TATAbox-28、CD17、CD71-72及CD26热点突变位点为检测靶点,分别设计3'末端与野生型基因位点或突变型基因位点完全配对的引物,并用硫化磷酸修饰,进行高保真DNA聚合酶介导的引物延伸反应.用多重PCR反应体系检测临床已知分别含CD26和TATAbox-28突变热点患者的DNA标本.结果 对野生模板而言,仅有野生型等位基因相关引物有产物产生,而突变型等位基因位点特异性引物无产物产生.反之,使用突变模板,仅突变型等位基因位点相关引物有产物产生,而野生型等位基因位点特异性引物无产物产生.高保真DNA聚合酶介导的多重引物延伸反应筛查含有CD26或TATAbox-28突变的患者DNA标本时显示,突变引物混合物只有相应突变位点的产物产生,而野生引物混合物则有除突变位点以外的其他5个位点的产物产生.结论 建立了基于高保真DNA聚合酶的筛查β地中海贫血基因热点突变的PCR技术.

摘要:
Epigallocatechin gallate (EGCG) is a major polyphenol in green tea with beneficial effects on the impairment in learning and memory. Autophagy is a cellular process that protects neurons from stressful conditions. The present study was designed to investigate whether EGCG can rescue chronic unpredictable mild stress (CUMS)-induced cognitive impairment in rats and whether its protective effect involves improvement of autophagic flux. As expected, our results showed that CUMS significantly impaired memory performance and inhibited autophagic flux as indicated by elevated LC3-II and p62 protein levels. At the same time, we observed an increased neuronal loss and activated mammalian target of rapamycin (mTOR)/p70 ribosomal protein S6 kinase (p70S6k) signaling in the CA1 regions. Interestingly, chronic treatment with EGCG (25 mg/kg, i.p.) significantly improved those behavioral alterations, attenuated histopathological abnormalities in hippocampal CA1 regions, reduced amyloid beta1-42 (Aβ1-42) levels, and restored autophagic flux. However, blocking autophagic flux with chloroquine, an inhibitor of autophagic flux, reversed these effects of EGCG. Taken together, these findings suggest that the impaired autophagy in CA1 regions of CUMS rats may contribute to learning and memory impairment. Therefore, we conclude that EGCG attenuation of CUMS-induced learning and memory impairment may be through rescuing autophagic flux. © 2014 Gu et al.

近年来,针对肿瘤生长的关键调控因子,尤其是针对肿瘤信号通路中起关键调控作用的表皮生长因子受体酪氨酸激酶进行的小分子激酶抑制剂的研发为人们提供了有效对抗肿瘤的新策略。综述了以4-芳氨基喹唑啉为母核具有烷基化功能基的表皮生长因子受体酪氨酸激酶抑制剂的研究进展,介绍了产生烷基化作用的功能基团以及抑制剂对受体蛋白的作用机理和结合方式。

摘要:
Cancer stem cells (CSCs) play crucial roles in tumor progression, chemo- and radiotherapy resistance, and recurrence. Recent studies on CSCs have advanced understanding of molecular oncology and development of novel therapeutic strategies. This review article updates the hypothesis and paradigm of CSCs with a focus on major signaling pathways and effectors that regulate CSC apoptosis. Selective CSC apoptotic inducers are introduced and their therapeutic potentials are discussed. These include synthetic and natural compounds, antibodies and recombinant proteins, and oligonucleotides. © 2014 by the authors; licensee MDPI, Basel, Switzerland.

摘要:
目的:建立不同批次夏桑菊颗粒的指纹图谱。方法:采用UPLC,建立12批夏桑菊颗粒的指纹图谱。ACQUITY UPLC BEH C_(18)色谱柱(2.1 mm×50 mm,1.7 μm),流动相为乙腈-0.5%醋酸溶液,梯度洗脱,柱温30℃,流速0.4 mL/min,检测波长320 nm,标定了16个色谱峰,分别采用相似度软件、聚类分析和主成分分析等方法,对12批样品进行系统的比较与归类。结果:建立了夏桑菊颗粒专属性的UPLC指纹图谱,将不同批次的样品分为6大类。结论:该方法重复性好,简便可靠,可用于夏桑菊颗粒的快速鉴别,可为夏桑菊颗粒的质量控制提供依据。

摘要:
目的:探讨烟酸联合姜黄素对高脂血症大鼠的降脂作用及肝脏TNF-α与IL-6表达的影响。方法50只SD大鼠随机分为溶剂对照组、高脂模型组、烟酸组〔100mg/(kg· d)〕、姜黄素组〔200mg/(kg· d)〕、联合用药组〔姜黄素200mg/(kg· d)+烟酸100mg/(kg· d)〕。8周后检测大鼠血脂,处死,取其肝脏,HE染色,免疫组化检测肝脏TNF-α与IL-6的表达情况。结果与对照组相比,高脂模型组的TC、LDL均明显升高(P<0.05),肝脏TNF-α与IL-6表达明显增高。与高脂模型组相比,烟酸组能降低TG、LDL含量(P<0.05),姜黄素组与联合用药组均能显著降低TC、TG、LDL含量(P<0.05),肝脏TNF-α与IL-6表达明显降低。结论烟酸与姜黄素联合用药可增强降脂作用,且较两者单独用药效果更佳;联合用药能够明显降低肝脏TNF-α与IL-6的表达。

柴胡龙牡汤对小鼠Lewis肺癌的实验研究

摘要:
目的:观察和评价柴胡龙牡汤对Lewis肺癌小鼠的治疗作用。方法:将40只C57BL/6J小鼠常规接种Lewis肺癌,造模成功后按随机数字表法分为柴胡龙牡汤组、模型组、顺铂对照组和恩度联合NP对照组4组。治疗时间均为2周。称瘤重计算抑瘤率,统计肺部结节数量并计算抗肺转移率,用MTT法检测肿瘤细胞的增殖能力、血液中红细胞、血小板、白细胞的比值和对脾脏中自然杀伤细胞活性、IFN-γ和IL-10含量及T淋巴细胞比值的影响。结果:柴胡龙牡汤组肿瘤质量和肺部转移结节数量明显减少(P < 0. 05);柴胡龙牡汤组在第2~3天的肿瘤细胞数目明显低于模型组(P < 0. 05);模型组、顺铂对照组和恩度联合NP治疗组自然杀伤细胞活性明显降低,IFN-γ含量减少,IL-10含量升高(P < 0. 05)。柴胡龙牡汤组的白细胞数量高于其他组(P <0. 05),脾脏T淋巴细胞的CD4~+、CD4~+/ CD8~+比值明显高于模型组、顺铂对照组和恩度联合NP治疗组,差异有统计学意义(P <0. 05)。结论:柴胡龙牡汤对Lewis肺癌肿瘤细胞有抑制作用。
