实现大型仪器设备开放共享的网络化管理是提高仪器设备管理效率、节约运行成本的有效途径。文章以湖南中医药大学大型仪器共享管理平台为例,介绍并总结了该平台的建设发展、功能应用和实施效果,以期为高校大型仪器设备共享平台的建设提供参考和借鉴。

摘要:
The asparagus of Triarrhena lutarioriparia (TL) is a popular vegetable with abundant chemical compounds in China. This study aims to optimize the ultrasound-assisted extraction (UAE) method for its content of total flavonoid and antioxidant activities by response surface methodology (RSM). Box-Behnken design was adopted to evaluate the influences of ethanol concentration, extraction time and solvent-to-sample ratio on the extraction yield of total flavonoid and the antioxidant activity. Considering the maximum content of extracted total flavonoids and antioxidant activity, the optimal extraction conditions were acquired with 70% (v/v) ethanol by UAE for 60min at a solvent-to-sample ratio of 40mL/g. The proportion of the extraction of total flavonoid was 15.88mg/g and antioxidant activity reached 79.53%. The RSM would be recommended as an appropriate model for simultaneous optimization of the UAE conditions for the content of total flavonoid and the antioxidant activity of asparagus of TL. © 2021, The Korean Society of Food Science and Technology.

摘要:
目的 观察白背叶总黄酮体外对肝星状细胞(HSC-T6)增殖的抑制作用和相关胶原蛋白、细胞因子的影响.方法 体外培养HSC-T6细胞,将白背叶总黄酮(终浓度为200、100、50、25、12.5μg/mL)作用于细胞48 h后,以MTT法检测细胞存活率,计算IC50;利用流式细胞仪检测细胞凋亡情况;收集细胞上清液,用ELISA法分别检测胶原蛋白COL-Ⅰ、COL-Ⅲ的表达水平和细胞因子TGF-β1、PDGF-BB的含量.结果 白背叶总黄酮能通过诱导HSC-T6凋亡而抑制HSC-T6细胞的增殖,其抑制作用与药物终浓度呈量效关系;并显著降低HSC-T6细胞生成的胶原蛋白COL-Ⅰ、COL-Ⅲ和细胞因子TGF-β1、PDGF-BB含量,与空白对照组比较差异有统计学意义(P<0.05或0.01).结论白背叶总黄酮体外对HSC-T6细胞增殖有显著抑制作用,可治疗肝纤维化.

摘要:
The present study aimed to develop a strategy involving quantitative analysis of multicomponents by single marker in combination with high-performance liquid chromatography fingerprint qualitative analysis for performing the quality control of Aurantii Fructus. The content of 12 components (eriocitrin, neoeriocitrin, narirutin, naringin, hesperidin, neohesperidin, meranzin, poncirin, naringenin, nobiletin, tangeretin, and auraptene) in samples was determined using reliable relative correction factors that were obtained using naringin as an internal reference standard. The new method demonstrated good applicability, and no significant differences were observed between the external standard method and the new method as determined by calculating standard method difference. Qualitative evaluation of samples was conducted using similarity analysis, hierarchical cluster analysis, and quality fluctuation analysis. Chromatographic fingerprint data were divided into three groups by similarity and hierarchical cluster analyses, and seven components may have a more significant impact on the quality of Aurantii Fructus in quality fluctuation analysis. Overall, the study suggests that the qualitative and quantitative analyses of multicomponents using quantitative analysis of multicomponents by single marker combined with chromatographic fingerprinting can be considered good quality criteria for performing quality control and providing technical support for the further pharmacological and pharmaceutical research of Aurantii Fructus. © 2020 WILEY-VCH Verlag GmbH & Co. 摘要:
The broad-spectrum DNA alkylating therapeutic, chlorambucil (CBL), has limited safety and shows lower therapy effect because of a short half-life while used in the clinic. Therefore, it is very necessary to develop a more efficient and safer type of CBL derivate against tumors with selective targeting of cancer cells. In addition, the natural product of honokiol (HN), the novel potent chemo-preventive or therapeutic entity/carrier, can target the mitochondria of cancer cells through STAT3 to prevent cancer from spreading and metastasizing. In this study, we designed and synthesized the honokiol-chlorambucil (HN-CBL) co-prodrugs through carbonate ester linkage conjugating with the targeted delivery help of the HN skeleton in cancer cells. Biological evaluation indicated that HN-CBL can remarkably enhance the antiproliferation of human leukemic cell lines CCRF-CEM, Jurkat, U937, MV4-11, and K562. Furthermore, HN-CBL can also selectively inhibit the lymphocytic leukemia (LL) cell survival compared to those mononuclear cells derived from healthy donors (PBMCs), enhance mitochondrial activity in leukemia cells, and induce LL cell apoptosis. Molecular docking and western blot study showed that HN-CBL can also bind with the STAT3 protein at some hydrophobic residues and downregulate the phosphorylation level of STAT3-like HN. Significantly, HN-CBL could dramatically delay leukemia growth in vivo with no observable physiological toxicity. Thus, HN-CBL may provide a novel and effective targeting therapeutic against LL with fewer side effects. Copyright © 2020 American Chemical Society.

摘要:
目的:将HPD100与D101大孔吸附树脂纯化禾本科芦笋总黄酮与总酚酸,优选总黄酮与总酚酸纯化工艺。方法:测定静态吸附率、解吸率、静态吸附曲线来评价HPD100大孔吸附树脂与D101大孔树脂对芦笋中总黄酮与总酚酸的静态吸附性能;然后对径高比、洗脱溶剂、水洗体积、洗脱剂量等考察获取动态纯化工艺。结果:总黄酮与总酚酸在HPD100大孔树脂上静态吸附量为分别为38.23 mg·g-1与22.40 mg·g-1,解吸率大小分别为68.95%与79.05%。动态优选的结果为浓度1∶0.25,上样量4 BV,径高比1∶4,静置4 h,9 BV水洗,50 ml 90%乙醇洗脱,总黄酮与总酚酸纯度分别达34.90%与20.07%。总黄酮与总酚酸在D101大孔树脂上静态吸附量为分别为40.05 mg·g-1与32 mg·g-1,解吸率大小分别为62.78%与71.74%。动态优选的结果为浓度1∶0.25,上样量3 BV,径高比1∶5,静置4 h,7 BV水洗,60 ml 90%乙醇洗脱,总黄酮与总酚酸纯度分别达34.10%与17.27%。结论:HPD100与D101大孔吸附树脂可以用于禾本科芦笋中总黄酮与总酚酸有效部位的纯化。

<jats:title>Abstract</jats:title><jats:p>In this study, a series of new flavones (2‐phenyl‐chromone), 2‐naphthyl chromone, 2‐anthryl‐chromone, or 2‐biphenyl‐chromone derivatives containing 6 or 7‐substituted tertiary amine side chain were designed, synthesized, and evaluated in acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibition. The results indicated that the alteration of aromatic ring connecting to chromone scaffold brings about a significant impact on biological activity. Compared with flavones, the inhibitory activity of 2‐naphthyl chromone, 2‐anthryl‐chromone derivatives against AChE significantly decreased, while that of 2‐biphenyl chromone derivatives with 7‐substituted tertiary amine side chain is better than relative flavones derivatives. For all new synthesized compounds, the position of tertiary amine side chain obviously influenced the activity of inhibiting AChE. The results above provide great worthy information for the further development of new AChE inhibitors. Among the newly synthesized compounds, compound <jats:bold>5a</jats:bold> is potent in AChE inhibition (IC<jats:sub>50</jats:sub> = 1.29 ± 0.10 μmol/L) with high selectivity for AChE over BChE (selectivity ratio: 27.96). An enzyme kinetic study of compound <jats:bold>5a</jats:bold> suggests that it produces a mixed‐type inhibitory effect against AChE.</jats:p>

目的考察禾本科芦笋中总黄酮在D101大孔吸附树脂上静态与动态吸附性能,优选总黄酮纯化工艺。方法以静态吸附率、解吸率、静态吸附曲线为依据比较D101、AB-8和HPD100的静态吸附性能,优选最优树脂,然后对其动态吸附工艺进行考察。结果静态吸附量大小关系为AB-8> D101> HPD100,采用95%乙醇解吸附时解吸率大小顺序为HPD100>AB-8>D101。以D101大孔树脂进行动态吸附观察,动态优选的结果为5床容积(BV)的样品(水∶样品的容积比为1∶0.25)上样于径高比1∶5的D101大孔树脂柱,静置4 h,7 BV水洗,60 ml 90%乙醇洗脱,纯度提高了6.54倍。结论 D101大孔吸附树脂可以用于禾本科芦笋中总黄酮的纯化。

Objective: This study was conducted to develop an method for identification of Trachemys scripta elegans (Ba Xi Gui, 巴西龟) shell glue from Chinemys reevesii (Zhong Hua Cao Gui, 中华草龟) shell glue. Methods: In this research, an ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS) method coupled with Binary Compare tool of UNIFI software was validated to differentiate Trachemys scripta elegans (Ba Xi Gui, 巴西龟) shell glue from Chinemys reevesii (Zhong Hua Cao Gui, 中华草龟) shell glue. The gelatins were digested enzymatically into peptides using trypsin. The UPLC-QTOF/MS data of the trypsin digested samples was subjected to Binary Compare of UNIFI software in order to screen out the unique peptides. PEAKS® Studio software was utilized to identify the sequence of the marker peptides. Results: It was found that m/z 641.3 was the marker peptide in samples of Trachemys scripta elegans (Ba Xi Gui, 巴西龟) shell glue, and m/z 649.3 was the marker peptide in samples of Chinemys reevesii (Zhong Hua Cao Gui, 中华草龟) shell glue, with possible amino acid sequence of GEAGPSGPAGPTGAR and GESGPSGPAGPTGAR respectively. Conclusions: The results will be helpful for the differentiating between these 2 gelatins, and it can be also used for quality control of Tortoise shell glue (Gui Jia Jiao, 龟甲胶). © 2019 Digital Chinese Medicine

摘要:
五味子是一种常见的补益中药,为木兰科植物五味子干燥成熟果实,最早收载于《神农本草经》上品中药。五味子的药用部位主要是果实、藤茎、枝叶,经过多年的研究发现其化学成分主要为木脂素、多糖、芳香油及三萜类化合物,因其药理作用丰富,有较大的开发和利用价值。随着民间广泛应用和研发的重视,五味子未知的药理作用、药用部位及有效成分不断被挖掘出来。本文将对近几年报道的五味子通过降低转氨酶、炎性因子发挥肝肾保护作用,诱导肿瘤细胞凋亡,抑制肿瘤细胞生长及肿瘤转移等发挥抗肿瘤、镇静催眠、改善记忆力、免疫调节作用等方面的研究进行综述,进一步明确了五味子的有效成分、药效作用及其作用机制。

目的:建立高效液相色谱(HPLC)法同时测定湘产枳壳中芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷和橙皮内酯的含量。方法:采用Venusil XBP C18(4. 6mm×250mm,5μm)色谱柱,以乙腈-0. 1%磷酸水为流动相(80∶20),流速1. 0ml/min,柱温35℃,变波长检测。结果:芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、橙皮内酯分别在100~2000ng、420~8400ng、32. 12~642. 4ng、260~5200ng、42. 75~855ng内呈良好线性关系,平均回收率分别为98. 7%、101. 0%、100. 7%、100. 6%、98. 6%,且精密度、重复性、稳定性良好;各批次样品中芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、橙皮内酯的含量范围分别为0. 48%~0. 84%、4. 44%~8. 50%、0. 19%~0. 95%、3. 03%~10. 19%、0. 16%~0. 30%;聚类分析分为3类,即11号样品为一类,2号与10号样品为一类,其余样品为一类;橙皮苷含量在各批次样本中波动较大。结论:本研究建立了湘产枳壳中5种成分的HPLC含量测定方法,该方法简便,准确度高,可为湘产枳壳质量控制提供科学依据。

摘要:
目的建立并识别不同产地白三七药材HPLC指纹图谱,为其质量控制及标准制定提供参考。方法采用HPLC法建立白三七指纹图谱,同时应用相似度评价、聚类分析和主成分分析(PCA)对实验数据进行识别,以分析不同产地白三七药材间及同科同属5种药材的相似性及差异性,并通过总量统计矩法对白三七药材共有峰进行定性定量分析。结果白三七指纹图谱共标定16个共有峰,并指认了人参皂苷Rg 1、Re、Rb 1及竹节参皂苷V、IV、IVa色谱峰;其中竹节参皂苷V和IV在不同批次白三七样品中的质量波动较小;各产地白三七样品除产自四川大凉山样品(S7)外相似度在0.90以上;聚类分析及主成分分析均将白三七样品分为3类;五加科人参属5种药材成分总量统计矩参数差异较大。结论建立的白三七HPLC指纹图谱和化学识别模式可为其质量控制和区分同科同属5种药材提供科学依据。

摘要:
目的 建立竹节参Panax japonicusC.A.Mey HPLC指纹图谱,并测定7种核苷类成分的含有量.方法 竹节参0.1%氨水提取液的分析采用Venusil MP C18色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-0.01%甲酸水,梯度洗脱;体积流量0.8 mL/min;柱温30℃;检测波长254 nm.对结果 进行聚类分析和主成分分析.结果 10批样品指纹图谱中有16个共有峰,除S2外相似度均在0.90以上.7种成分在各自范围内线性关系良好(R2≥0.999 6),平均加样回收率92.74%~98.66%,RSD 2.32%~2.97%.10批样品分为3类.结论 该方法 简便可靠,重复性好,可用于竹节参药材的质量控制.

摘要:
目的 采用超高效液相色谱-四级杆-飞行时间串联质谱法(UPLC-Q-TOF-MS)对西洋参的主要化学成分及入血成分进行分析鉴定.方法 采用超高效液相色谱梯度洗脱分离各主要成分;电喷雾电离源负离子模式检测,四级杆飞行时间串联质谱法对各主要色谱峰进行归属.灌胃给予大鼠3 g·kg-西洋参粉,收集血清样品.采用UPLC-Q-TOF-MS分析并鉴定西洋参粉提取物及入血成分.结果 通过质谱信息并结合对照品数据及相关文献,提取物中共鉴定了22个化合物,大鼠含药血清中共鉴定了8个入血成分.结论 UPLC-Q-TOF-MS可较全面地对西洋参成分进行定性分析,为进一步阐明药效物质基础和建立全面的质量控制提供参考依据.

摘要:
目的:基于抗氧化活性评价优选禾本科芦笋多糖酶法提取工艺。方法:以活性部位总多糖浸出量与DPPH自由基清除率为评价指标进行评价,在单因素试验的基础上进行正交试验,优选禾本科芦笋多糖的提取工艺。结果:最佳酶法提取工艺为芦笋粉末加3. 0%纤维素酶、p H 6. 5的柠檬酸缓冲溶液,在45℃温浸提取1. 5h,多糖提取量与DPPH自由基清除率分别达到66. 58mg/g与65. 24%。结论:正交试验法优选禾本科芦笋多糖的酶法提取工艺可行,该工艺为禾本科芦笋多糖的酶法提取优化提供了试验依据。

摘要:
目的:探讨麸炒对枳壳挥发油中柠檬烯含量的影响,并分析枳壳挥发油的抑菌活性与柠檬烯含量的关系。方法:采用气相色谱结合外标法测定不同产地枳壳炮制前后挥发油中柠檬烯含量区别;采用96孔板法测定枳壳挥发油对金黄色葡萄球菌、表皮葡萄球菌、大肠杆菌和沙门氏菌的最小抑菌浓度(MIC),以1/MIC值代表抑菌活性,运用双变量相关分析计算挥发油的抑菌活性与柠檬.烯含量的关联性。结果:麸炒会降低枳壳柠檬烯含量;关联分析结果表明,枳壳挥发油中柠檬烯含量越高.其挥发油对金黄色葡萄球菌和大肠杆菌的抑制作用越强。结论:麸炒炮制可影响枳壳挥发油的化学组成;柠檬烯为枳壳挥发油的抑菌物质基础之一。

摘要:
Background: Polygonum cuspidatum Sieb. et Zucc. (named Huzhang in china) is a traditional and popular chinese medicinal herb used in removing jaundice, clearing heat-toxin, improving blood circulation, expelling stasis, dispelling wind and dampness, repelling phlegm, and suppressing cough. it is widely used in drug and functional food fields and distributed throughout the world, including in china, Japan, and north america. Objective: to control the quality of Polygonum cuspidatum, an effective, reliable, and simple method for simultaneous determination of two stilbenes (polydatin, resveratrol) and four anthraquinones (emodin, physcion, rhein, and anthraglycoside B) was developed and validated for the first time in this study by reversed-phase Hplc (rp-Hplc). Methods: Separation was carried out on agilent c 18 column (250 × 4.6 mm I.D., 5 μm) with acetonitrile and 0.10% aqueous phosphoric acid as mobile phase and gradient elution at a flow rate of 0.8 mL/min. Detection was conducted with mobile wavelength at 30°c. Results: Good validation of the method including linearity, precision, repeatability, and recovery was performed. the contents of the studied analytes are significantly different, and resveratrol and rhein in particular existed in greater fluctuation among the samples. Conclusion: a simple, reliable, and sensitive method has been successfully established and applied to the analysis for simultaneous determination of the target compounds in 11 batches of samples. Highlights: Separation and quantitative analysis of two stilbenes and four anthraquinones from P. cuspidatum were developed by rp-Hplc. this method is convenient, sensitive, and accurate and can provide a reliable basis for further applications of P. cuspidatum in drug or food fields. © 2019 AOAC International. 目的建立一测多评法同时测定竹节参中7种皂苷类成分的含量,并验证此方法在竹节参中应用的可行性与适用性。方法采用HPLC法,以竹节参皂苷Ⅴ为内参物,建立人参皂苷Rg_1、Re、Rb_1,假人参皂苷RT_1,竹节参皂苷Ⅳ、Ⅳa的相对校正因子(f_(s/i)),并利用f_(s/i)计算竹节参样品中成分的含量,同时用外标法测定各成分的含量,比较2种方法的差异。结果人参皂苷Rg_1、Re、Rb_1,假人参皂苷RT_1,竹节参皂苷Ⅳ和Ⅳa的f_(s/i)分别为1.286 7、1.432 7、0.966 6、0.962 4、1.207 2、 0.938 4。采用一测多评法与外标法得到的10批竹节参样品各成分含量无显著性差异。结论建立的一测多评法准确性高,可用于竹节参皂苷类成分的定量分析和质量评价。

摘要:
Chalcones containing tertiary amine side-chains have potent activity as acetylcholinesterase (AChE) inhibitors. However, the effects of the location of the tertiary amine groups as well as of other groups on AChE and butyrylcholinesterase (BChE) activity have not been reported. Here, we report the synthesis and testing of 36 new coumarin–chalcone hybrids (5d–7j, 9d–11f, 12k–13m) against AChE and BChE. The nature and position of the chalcone substituents had major effects on inhibitory activity as well as selectivity for AChE over BChE. Compounds with para-substituted chalcone fragments in which the substituents were choline-like had potent activity against AChE and poor activity against BChE, while ortho-substituted analogs exhibited an opposite effect. Replacement of the terminal amine groups by amide, alkyl or alkenyl groups abrogated activity. Compound 5e showed potent inhibitory activity (IC50=0.15±0.01μmol/L) and good selectivity for AChE over BChE (ratio 27.4), and a kinetic study showed that 5e exhibited mixed-type inhibition against AChE. Computational docking results indicate that 5e binds to Trp 279, Tyr334 and Trp 84 in AChE, but only to Trp 82 in BChE. Overall, the results show that coumarin–chalcone hybrids with choline-like side-chains have promising activity and selectivity against AChE and be promising therapeutic leads for Alzheimer’s disease. © 2018, Springer International Publishing AG, part of Springer Nature.

目的:测定肉桂酸与桂皮醛的平衡溶解度及表观油水分配系数(P(app))。方法:采用HPLC法测定肉桂酸与桂皮醛在不同pH的缓冲溶液中的平衡溶解度;结合摇瓶法测定其在不同pH缓冲盐溶液及正辛醇-水溶液中的表观油水分配系数。结果:在pH 7.8的缓冲溶液中肉桂酸的平衡溶解度最大,pH 6.8的缓冲溶液中桂皮醛的平衡溶解度最大;肉桂酸与桂皮醛在正辛醇饱和的水溶液中lg P(app)值分别为0.85与1.26,在不同磷酸盐缓冲液中(pH 1.2~7.8)的lg P(app)范围分别在-1.04~2.27,0.29~1.67之间。结论:此法简便、准确、快捷,可预测化学成分在体内的吸收;在胃肠道环境中,桂皮醛的吸收较好,肉桂酸在胃中的吸收较差,在肠道中吸收较好。
