Ethnopharmacological relevance: Chaihu-Longgu-Muli Decoction (CLMD) is a classic prescription created by Zhong-jing Zhang, a famous ancient Chinese medical scientist, to harmonize uncontrollable body activities and calm the minds. Now Traditional Chinese Medicine (TCM) physicians often apply it to treat psychiatric diseases such as epilepsy. Aim of the study: This study investigated the mechanism of the effect of Chaihu-Longgu-Muli Decoction (CLMD) on hippocampal neurons pyroptosis in rats with Temporal Lobe Epilepsy (TLE). Materials and methods: The lithium chloride-pilocarpine-induced TLE rat model was established. The behavioral testing was performed and, the expression of IL-1β and TNF-α in serum was detected by ELISA, qRT-PCR was used to detect the mRNA expression of NLRP3, Caspase-1, IL-1β and TNF-α in hippocampus. The expression of NLRP3 and Caspase-1 in hippocampal dentate gyrus was detected by immunofluorescence assay. Results: CLMD could significantly suppress the frequency and duration time of epileptic seizures, reduce the expression of NLRP3, Caspase-1 TNF-α and IL-1β. Conclusions: CLMD exerted an obvious antiepileptic effect by improving pyroptosis in hippocampal neurons of TLE rats. © 2021

目的 探究陈士铎治疗儿科疾病的处方用药特色以及规律.方法 从《陈士铎医学全书》中手工检索并且阅读关于儿科的医案,通过全文阅读提取方药信息,用Microsoft Excel建立“陈士铎儿科用药数据库”,同时整理归纳数据,应用Excel 2013频数分析、以及结合专业知识分析归纳四气、五味和归经;应用IBM SPSS Statistics 19系统聚类分析;应用IBM SPSS Modeler 18.0药物间网络化关联展示以及关联规则分析.结果 纳入29个方剂,总计82味药,用药频率为281次,使用频数≥10次的药物8味,人参、当归、白术、荆芥、茯苓、甘草、陈皮、麦冬,累计使用110频次,贡献百分比为39.15%,分析发现其中补气药、健脾药使用频率较高;四气以寒33频次(40.24%)、温29频次(35.37%)、平为主13频次(15.85%);五味以甘40频次(30.77)、辛38频次(29.23%)、苦30频次(23.08%)为主;归经药物主要入脾(胃)52频次(30.23%)、肺(大肠)38频次(22.09%).系统聚类分析结果显示,陈氏多用健脾益气之药,多围绕四君子汤加减变化,同时消补兼施、消散并用;关联规则聚类分析结果显示,陈氏儿科疾病用药以四君子配伍消导之法最为明显.结论 陈氏治疗儿科疾病,注重小儿易虚之体,用药多以脾胃入手;药物组方精当,补中有散,补泻兼施,注重五行生克制化关系用药,强调治法与选药.

Ethnopharmacological relevance: Chaihu-Longgu-Muli decoction (CLMD) is a well-known ancient formula in traditional Chinese medicine (TCM) to relieve disorder, clear away heat, tranquilize the mind and allay excitement. It has been used for the therapy of neuropsychiatric disorders such as epilepsy, dementia, insomnia, anxiety, and depression for several centuries in China. Aim of the study: This paper is based on the assumption that the mechanism by which CLMD relieves epileptic symptoms in rats is associated with improving autophagy. Several experimental methods are designed to testify the hypothesis. Materials and methods: The lithium-pilocarpine-induced epilepsy model was established in rats. The seizure frequency was recorded. Morphology and number of autophagosomes in hippocampal dentate gyrus was detected with a transmission electron microscope (TEM). Expression of Beclin-1, microtubule-associated proteins 1A/1B light chain 3 (LC3), and mammalian target of rapamycin (mTOR) in dentate gyrus was measured by immunofluorescence assay, quantitative reverse transcription polymerase chain reaction (qRT-PCR) and Western-blotting. Results: CLMD could significantly relieve the seizure frequency and improve autophagy in hippocampal dentate gyrus. Meanwhile, the level of Beclin-1 and LC3B decreased significantly, while mTOR increased remarkably after medical intervention. Conclusions: CLMD could improve autophagy in hippocampal dentate gyrus due to epilepsy, especially at high dose. The mechanism may be related to upregulated expression of mTOR and downregulated expression of Beclin-1 and LC3B. © 2020 Elsevier B.V.

Polygonatum sibiricum polysaccharide (PSP) has been shown to alleviate hyperglycemia and reduce oxidative stress to delay the progression of diabetic retinopathy and cataracts. However, its role and underlying mechanisms in regulating type 2 diabetes mellitus (T2DM) remain unclear. Nuclear factor erythroid 2-related factor 2 (Nrf2) activation plays a protective role in T2DM. The present study focused on the effect of PSP on inflammatory cytokine secretion and Nrf2 expression in the adipocytes of T2DM patients. In this study, high-glucose- and high-insulin-induced 3T3-L 1 adipocytes were used to mimic insulin-resistant (IR)-3T3-L 1 adipocytes. Furthermore, the effect and underlying mechanisms of PSP on inflammation and glucose uptake in IR-3T3-L1 adipocytes were investigated. The present study found that proliferation after 50, 100 and 250 µg/ml PSP treatment had no significant change in normal 3T3-L 1 adipocytes. A total of 50, 100 and250 µg/ml of PSP also alleviated IL- 1ß, IL- 6, and TNF-a levels and promoted proliferation, glucose uptake, and glucose transporter 4 expression in IR- 3T3-L 1 adipocytes. Furthermore, 50, 100 and 250 µg/ml PSP promoted Nrf2 and HO- 1 expression. However, silencing Nrf2 expression reversed the effect of 100 µg/ml PSP in IR- 3T3-L 1 adipocytes. In conclusion, these results suggest that PSP alleviates inflammatory cytokines and promotes glucose uptake in IR-3T3-L1 adipocytes by promoting Nrf2 expression. PSP may be a potential therapeutic agent for T2DM treatment by promoting Nrf2 expression. © 2019 Spandidos Publications. All rights reserved.

Long non-coding RNA (lncRNA) plays an important role in malignant tumor occurrence, development, and chemoresistance, but the mechanism of how they affect nasopharyngeal cancer (NPC) paclitaxel chemosensitivity is unclear. In this study, lncRNA array of CNE-1 and HNE-2 paclitaxel-resistant cells and their parental strains revealed that the paclitaxel-resistant strains had significantly lower MRVI1-AS1 (murine retrovirus integration site 1 homolog antisense RNA 1) expression than the parental strains, and that MRVI1-AS1 overexpression in vitro and in vivo increased paclitaxel chemosensitivity. Further, MRVI1-AS1 upregulated ATF3 (activating transcription factor 3) by simultaneously inhibiting miR-513a-5p (microRNA-513a-5p) and miR-27b-3p expression levels to increase NPC paclitaxel chemosensitivity. Chromatin immunoprecipitation and quantitative real-time PCR showed that ATF3 could feed-back MRVI1-AS1 regulation positively. Furthermore, MRVI1-AS1 and ATF3 could form a positive feedback loop, which promoted the expression of RASSF1 (Ras association domain family member 1), a Hippo–TAZ (tafazzin) signaling pathway regulatory factor, thereby inhibiting TAZ expression. The MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyltetrazolium bromide) assay and flow cytometry showed that the decreased TAZ increased NPC cell paclitaxel chemosensitivity. Overall, the results indicate that the MRVI1-AS1/ATF3 signaling pathway can increase NPC paclitaxel chemosensitivity by modulating the Hippo–TAZ signaling pathway. Therefore, targeting the loop may be a new NPC treatment strategy. © 2019, The Author(s), under exclusive licence to Springer Nature Limited.

MiR-130b and SAM and SH3 domain containing 1 (SASH1) play an important role in many types of human cancers. The aim of our research was to study their interactions in the process of the proliferation and aggressiveness of oesophageal squamous cell carcinoma (ESCC) cells. Microarray analysis was done to screen the differentially expressed genes in the ESCC tissues. miR-130b and SASH1 mRNA levels in the ESCC tissues and cells were detected by qRT-PCR. Dual luciferase reporter system was used to verify the target relationship between miR-130b and SASH1. The effects of miR-130b on SASH1 expression were explored by western blot in KYSE30 and TE1 cell lines. CCK-8 assay, flow cytometry, Transwell, and wound healing assays were conducted to explore the effects of miR-130b and SASH1 invitro. In addition, invivo experiments were conducted to study the roles of miR-130b and SASH1. miR-130b was highly expressed, while SASH1 was the opposite in both the ESCC tissues and cells. The expression of SASH1 was inhibited by the direct binding of miR-130b. The inhibition of miR-130b reduced the proliferation and aggressiveness of ESCC cells, while it also induced apoptosis and cell cycle arrest in the ESCC cells by suppressing SASH1. The invivo assay suggested that the overexpression of miR-130b promoted the growth of ESCC tumours. MiR-130b was up-regulated in the ESCC tumour tissues and cells, acting as a tumour promoter. A stimulating effect was demonstrated on ESCC cell growth and aggressiveness by suppressing SASH1, which is an anti-oncogene. © 2018 The Authors. 摘要:
采用分子动力学模拟的方法,用末端距分布、扭转角分布和全局取向有序参数对正二十六烷烃体系相变过程的微观结构变化进行了研究。研究表明:体系的有序性均随着温度的升高而逐渐降低,正二十六烷烃分子在熔化过程中倾向于采取伸展和交叉两种构象,且逐渐以交叉构象为主,这一倾向与熔化过程中体系有序度降低的热力学理论一致。根据体系自扩散系数的突变,可以计算出正二十六烷烃的熔化与凝固温度分别为329.4K和335.1K,与实验值基本吻合。

目的:基于文献研究小儿反复呼吸道感染非急性期证素的分布及组合规律,对其基础证的主次症进行筛选.方法:收集近20年小儿反复呼吸道感染非急性期的相关文献并整理,运用频率描述及两分类Logistic回归分析的方法进行统计分析.结果:共纳入合格文献158篇,共计339条记录.小儿反复呼吸道感染非急性期的病位证素以肺、脾为主,病性证素以气虚、阴虚、痰及血瘀为主;其组合形式以三、四证素组合最为常见,组合类型则以肺+脾+气虚、肺+气虚、肺+气虚+血瘀及肺+脾+气虚+血瘀最为常见;其基础证主要有肺脾气虚证及肺气虚证.肺脾气虚证的主症为纳差、自汗、大便溏及久咳,次症为乏力、面色萎黄及面色少华.肺气虚证的主症为恶风、恶寒、面白,次症为语声低弱、久咳、面色少华、自汗.结论:小儿反复呼吸道感染非急性期证素分布及组合存在一定的规律,可为进一步研究提供参考.

温中运脾散敷脐治疗小儿迁延性腹泻42例总结

摘要:
目的:观察温中运脾散敷脐治疗小儿迁延性腹泻的临床疗效。方法:将84例迁延性腹泻患儿随机分为治疗组和对照组各42例,治疗组予温中运脾散敷脐,对照组口服培菲康治疗。两组疗程为14d。结果:总有效率治疗组为85.71%,对照组为61.90%,两组比较,差异有统计学意义(P〈0.05)。治疗组主要症状消失时间、病程明显短于对照组(P〈0.05),血红蛋白含量及微量元素锌、铁含量明显高于对照组,差异有统计学意义(P〈0.05)。结论:温中运脾散敷脐治疗小儿迁延性腹泻有较好的临床疗效。

摘要:
目的:观察运脾消食散配合针刺四缝穴治疗小儿厌食的临床疗效。方法:将132例小儿厌食患儿随机分为治疗组和对照组各66例。治疗组口服运脾消食散配合针刺四缝穴治疗,对照组服用健脾消食口服液治疗,疗程均为4周。结果:总有效率治疗组为92.42%,对照组为65.15%,两组比较,差异有统计学意义(P<0.05);治疗组血红蛋白及微量无素锌、铁含量明显高于对照组,差异有统计学意义(P<0.01)。结论:运脾消食散配合针刺四缝穴治疗小儿厌食疗效确切,值得临床推广运用。

目的 观察五虎汤对哮喘婴幼儿DC刺激T淋巴细胞分泌IL-4、IL-5的影响.方法 将60例哮喘婴幼儿随机分为五虎汤治疗组和西药对照组(分别简称治疗组,对照组)各30例,另选健康婴幼儿10例.以Thomas法分离、培养并诱导治疗组及对照组哮喘婴幼儿和健康婴幼儿外周血DC细胞,并与健康儿童的T淋巴细胞共同培养后,采用酶联免疫双抗体夹心法(ELISA)检测各组患儿血清中IL-4、IL-5浓度.结果 婴幼儿哮喘患儿DC刺激T淋巴细胞分泌IL-4、IL-5较健康组显著升高,差异具有统计学意义(P<0.01);五虎汤治疗后IL-4、IL-5均较治疗前显著降低,差异具有统计学意义(P<0.05或P<0.01),且治疗组下降幅度均较对照组显著,差异具有统计学意义(P<0.01).结论 五虎汤具有抑制哮喘患儿Th2类细胞因子IL-4、IL-5的产生,从而调节Th1/Th2的失衡.

目的 观察五虎汤对婴幼儿哮喘患儿外周血树突状细胞(DC)共刺激分子CD80、CD83和CD86表达的影响,为探讨五虎汤防治本病的机制提供依据.方法 将60例痰热壅肺证哮喘患儿随机分为治疗组及对照组,并设健康组(10名健康体检儿童)作对照.以Thomas法分离、培养并诱导治疗组(采用五虎汤口服)、对照组(采用西药辅舒酮等吸入或口服)和健康儿的外周血树突状细胞(DC),流式细胞仪检测外周血DC表面共刺激分子(CD80、CD83和CD86)的表达,并分析其治疗前后的变化.结果 治疗前哮喘患儿外周血DC表面CD80、CD86的表达较健康组明显升高(P<0.01);治疗后治疗组、对照组CD80(分别为13.18±3.02、15.34±3.90)、CD86(分别为29.65±8.55、35.88±13.85)表达较治疗前(CD80分别为18.06±4.53、18.40±3.86,CD86分别为38.61±10.54、38.29±11.67)均降低(P<0.01,P<0.05);治疗组较对照组降低更明显(P<0.05),而CD83两组治疗前后、组间及与健康组比较,差异均无统计学意义(P>0.05).结论 五虎汤可通过降低CD80、CD86表达而对哮喘惠儿DC共刺激分子产生调节作用.

目的:观察五虎汤对哮喘婴幼儿树突状细胞(DC)刺激T淋巴细胞分泌IL-2、IL-4、IL-5、IFN-γ的影响。方法:将60例哮喘婴幼儿随机分为2组各30例,对照组予西药常规治疗;治疗组予五虎汤治疗。另选健康组婴幼儿10例。以Thomas法分离、培养并诱导治疗组及时照组哮喘婴幼儿和健康婴幼儿外周血DC细胞,并与健康儿童的T淋巴细胞共同培养后,采用酶联免疫双抗体夹心法(ELISA)检测各组患儿血清中IL-2、IL-4、IL-5、IFN-γ浓度。结果:哮喘婴幼儿DC刺激T淋巴细胞分泌IL-2较健康组显著降低(P<0.05),而IL-4、IL-5较健康组显著升高(P<0.01),IFN-γ与健康组比较,差异无显著性意义(P>0.05);五虎汤治疗后IL-2较治疗前显著增高(P<0.01),而IL-4、IL-5均较治疗前显著降低(P<0.05,P<0.01),且治疗组IL-4、IL-5下降幅度均较对照组显著(P<0.01)。结论:五虎汤具有抑制哮喘患儿Th2类细胞因子IL- 4、IL-5的产生,提高IL-2水平,从而调节Th1/Th2的作用。

摘要:
目的 现察咳喘宁对哮喘豚鼠嗜酸性粒细胞(EOS)的调节作用,阐明其防治哮喘的作用机制.方法 用卵清蛋白复制豚鼠哮喘模型,以地塞米松为阳性对照药,现察咳喘宁干预后血中和肺泡灌洗液中EOS的改变情况.结果 咳喘宁能有效降低哮喘琢鼠血和肺泡灌洗液中EOS的数量,与模型组相比羔异有统计学意义(P<0.05).地塞米松降低血中EOS的效果优于咳喘宁.但差异无统计学意义(P>0.05).结论 咳喘宁能降低哮喘豚鼠血中和肺泡灌洗液中EOS水平.

摘要:
目的 观察益气补肾活血法佐治小儿原发性肾病综合征的临床疗效.方法 将60例肾病综合征惠儿随机分为两组.对照组28例予激素治疗,治疗组32例予激素加中药治疗,进行临床疗效对比观察.结果 治疗组疾病疗效总有效率为93.75%,完全缓解率为75%,与对照组比较,差异有统计学意义(P<0.05),且治疗组在消除蛋白尿、升高血浆白蛋白、改善血脂代谢及血液高凝状态、增强免疫功能、减少病情复发等方面均明显优于对照组(P<0.05,P<0.01).结论 益气补肾活血法佐治小儿肾病综合征能明显提高临床疗效,减少复发.

摘要:
目的:观察咳喘宁对病毒诱发哮喘患儿T h亚群细胞因子表达的影响。方法:120例病毒诱发哮喘患儿随机分为咳喘宁治疗组60例和西药对照组60例,分别采用咳喘宁、利巴韦林治疗,观察两组患儿治疗后血清中IL-12、IL-4的含量变化。结果:咳喘宁能显著提高患儿血中IL-12,降低IL-4,其作用明显优于对照组。结论:咳喘宁治疗病毒诱发小儿哮喘的机制可能与提高患儿IL-12水平、降低IL4水平有关。

目的:观察咳喘宁防治病毒诱发小儿哮喘的临床疗效及对患儿Th亚群细胞因子表达的影响。方法:将120例病毒诱发哮喘患儿随机分为咳喘宁治疗组60例和西药对照组60例,观察两组临床疗效及患)Lb-清中IL-12、IL-4的含量变化。结果:治疗组总有效率91.67%,临床控制率35%,均明显优于对照组(分别为P〈0.01、P〈0.05);咳喘宁能显著提高患儿血中IL-12、降低IL-4,其作用明显优于对照组(P〈0.01)。结论:咳喘宁防治病毒诱发小儿哮喘具有较好疗效,其作用机制可能与提高患儿IL-12水平、降低IL-4水平有关。

目的:观察咳喘宁治疗病毒诱发小儿哮喘的临床疗效.方法:将120例病毒诱发哮喘患儿随机分为2组,治疗组60例以咳喘宁(主要由炙麻黄、苦杏仁、桃仁、大青叶、黄芪、生石膏、细茶、甘草等组成)治疗,对照组60例采用利巴韦林治疗.结果:治疗组总有效率为91.67%,临床控制率为35.00%,均明显优于对照组(分别为76.67%,16.67%),2组比较,差异有显著性或非常显著性意义(P<0.05,P<0.01).结论:咳喘宁治疗病毒诱发小儿哮喘具有较好疗效,且无明显毒副作用.

清解化瘀汤治疗儿童过敏性紫癜和预防肾损害的临床观察

摘要:
Objective:To observe the clinical effect of Qingjie Huayu Tang (QHT,清解化瘀汤)for children allergic purpura and prevention of renal damage. Mothods:68 cases were allocated to 2 groups randomly.The control group was treat- ed by routine western medicine,and the treatment group added with QHT.2 weeks constituted the course of treatment.Results:The total effective rate was 92.10% and 70.00% in treatment group and control group respectively(P<0.05),the time of disappearance of symptoms and signs,and the hospitalized time of treatment group were shorter than control group(P<0.05).After treatment,the urineβ_2-MG and serum IgA of both groups were decreased in different degrees(P<0.05,P<0.01),but was more marked in treatment group as compared with control group(P<0.05).A follow-up of 3 mouths showed that the recurrence rate of purpura was 23.68% and 46.66% in treatment group and control group respectively(P<0.0.5); the abnormal rate of routine urine examination was 7.9% and 30.0% in treatment group and control group respectively(P<0.05),Conclusion:QHT can markedly increase the effective rate,decrease the recurrence rate and prevent the renal damage of children allergic puroura.

清肺化痰通络法治疗小儿支原体肺炎42例临床观察

摘要:
目的:观察中医清肺化痰通络法联合抗生素治疗小儿支原体肺炎的临床疗效.方法:将82例患儿随机分为两组,治疗组42例予清肺化痰通络法联合阿奇霉素治疗,对照组40例用阿奇霉素治疗,进行临床对比观察.结果:治疗组与对照组总有效率分别为92.9%和75.0%,差异有统计学意义(P<0.05).治疗组的发热、咳嗽、肺部体征消失的时间、胸部X片恢复正常的时间明显少于对照组,差异有统计学意义(P<0.05).结论:清肺化痰通络法联合阿奇霉素治疗儿童支原体肺炎有较好的疗效.
