Tetrahydroxy stilbene glycoside attenuates acetaminophen-induced hepatotoxicity by UHPLC-Q-TOF/MS-based metabolomics and multivariate data analysis

摘要:
Tetrahydroxy stilbene glycoside (TSG) is a main active compound in Polygonum multiflorum. Acetaminophen (APAP) is a well-known analgesic and antipyretic drug. It is considered to be safe within a therapeutic range, in case of acute intoxication hepatotoxicity occurs. This present study aims to observe TSG-provided alleviation on APAP-induced hepatoxicity in C57BL/6 mice. APAP performs extensive necrosis and dissolves nucleus suggesting liver damage from hepatic histopathology. Serum alanine aminotransferase, aspartate aminotransferase, lactate dehydrogenase, and alkaline phosphatase analysis and liver histological evaluation showed that TSG reduced the hepatotoxicity induced by a toxic dose of APAP. Moreover, TSG alone had no hepatotoxicity. TSG eliminated hepatic glutathione depletion and cysteine adducts formation. It also reduced the expression of interleukin-10 and lowered the production of reactive oxygen species in liver tissues. Luminex was used to detect cytokine production in different groups. Herein, we used an untargeted metabolomics approach by performing ultrahigh-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry on treated mice to identify metabolic disruptions under APAP and TSG. Major alterations were observed for purine metabolism, amino acid metabolism, and fatty acid metabolism. These data provide metabolic evidence and biomarkers in the liver that the ABC transporters, Glycine serine and threonine metabolism, and Choline metabolism in cancer changed the most. These targets of metabolites have the potential to improve our understanding of homeostatic. Meanwhile, these metabolites revealed that TSG can alleviate inflammation caused by APAP and promote the activity of intrinsic antioxidants. In summary, TSG can regulate lipid metabolism, promote the production of antioxidant enzymes, and decrease the inflammatory response.

摘要:
目的 运用网络药理学及高通量分子对接技术,筛选金花清感颗粒中与人类新型冠状病毒3CL水解酶及ACE2受体蛋白结合活性较好的有效化合物,并对其干预COVID-19的机制进行预测.方法 本研究利用TCMSP数据库及高通量分子对接技术筛选金花清感颗粒中与人类新型冠状病毒3CL水解酶及ACE2受体对接活性较高的的活性化合物,并获取其对应靶标,利用STRING数据库进行靶标的蛋白网络互作,运用数据包进行京都基因与基因组百科全书(KEGG)通路富集分析,预测其作用机制.结果 金花清感颗粒共筛选有效活性化合物276种,涉及靶标基因325个.“药物-活性化合物-靶标”网络图中筛选出关键化合物11个,高通量分子对接筛选得到与人类新型冠状病毒3CL水解酶及ACE2受体结合活性较好的8个化合物,其靶基因的PPI网络主要涉及AKTI、HSP90AA1、RELA等核心靶蛋白.KEGG富集分析获得94条相关通路,主要包括卡波西肉瘤相关疱疹病毒感染、丙型肝炎、麻疹、人巨细胞病毒感染、EB病毒感染等病毒相关通路以及IL-17通路、TNF通路、AGE-RAGE等途径.结论 金花清感颗粒中3-甲氧基光甘草定、粗毛甘草素C、甘草查尔酮B等活性成分与人类新型冠状病毒3CL水解酶及ACE2受体酶结合活性较强,并且通过多种生物学途径及通路发挥干预COVID-19的作用.

摘要:
目的:基于线粒体途径探讨知柏地黄汤含药血清干预解脲脲原体(UU)感染大鼠生精细胞活力的影响及其作用机制。方法:采用在体UU感染建模,随后原代提取大鼠生精细胞,以知柏地黄汤含药血清(低、中、高剂量组)为实验施加因素,同时设空白组、强力霉素组和模型组。给予相应药物干预后进行指标检测。结果:知柏地黄汤含药血清能改善UU感染的生精细胞活性,提高存活率(P<0.01, P<0.05);知柏地黄汤含药血清能有效提高UU感染的生精细胞线粒体ANT4转运活性(P<0.01,P<0.05);知柏地黄汤含药血清能有效上调UU感染的生精细胞中VDAC2、Cyp-D蛋白和mRNA表达水平(P<0.01, P<0.05),下调VDAC1蛋白和mRNA表达水平(P<0.01, P<0.05)。结论:知柏地黄汤可能通过抑制UU感染引起的VDAC1的过度激活,上调VDAC2、Cyp-D mRNA和蛋白表达,上调ANT4的转运活性,进而明显改善线粒体结构的破坏,最终改善生精细胞的活力。

目的:基于TL R7/8介导的I F N-α/β蛋白表达水平探讨麻黄先煎之麻杏石甘汤抗流感病毒效应机制。方法:以TLR7/8激活剂(R848)、抑制剂(抗体TLR7和抗体TLR8)干预的小鼠巨噬细胞为对照,以酶联免疫吸附技术检测流感病毒干预小鼠巨噬细胞24、48h的IFN-α/β分泌水平。以流感病毒干预的小鼠巨噬细胞为对照,以酶联免疫吸附技术和Western Blot技术检测麻黄先煎之麻杏石甘汤含药血清干预24、48h的流感病毒感染的小鼠巨噬细胞IFN-α/β分泌水平和蛋白表达水平。结果:与正常大鼠血清组及胎牛血清组比较,TLR7/8激活剂(R848)与流感病毒干预24、48h的RAW264.7细胞IFN-α和IFN-β分泌水平和蛋白表达水平显著升高(P<0.05);与病毒干预组比较,四药同煎及麻黄先煎20、25、30min之麻杏石甘汤含药血清作用24、48h后,流感病毒感染后的巨噬细胞IFN-α/β的分泌水平和蛋白表达水平显著降低(P<0.05);与四药同煎组比较,在含药血清作用48h的时间点,麻黄先煎25min之麻杏石甘汤组IFN-α分泌水平显著降低(P<0.05)。结论:流感病毒感染对TLR7/8介导的IFN-α/β蛋白分泌水平有明显的上调作用,麻杏石甘汤能够下调流感病毒感染的巨噬细胞IFN-α/β分泌水平和蛋白表达水平,在作用48h时间点,麻黄先煎25min之麻杏石甘汤含药血清对IFN-α分泌水平的调节作用优于四药同煎。

目的:依托大学生创新训练中心,对独立学院学生开展课外科技活动,提升独立学院学生创新能力,有助于中医药院校独立学院学生适应当下社会对综合性高素质人才的需求。方法:依托大学生创新训练中心,结合二级学院的专业特点、分不同年级、根据兴趣双向选择,自主设计科研课题,通过参加实验设计、动手操作、记录分析和处理结果等综合评价中医药课外科技活动在提升独立学院学生创新能力的作用。结果:依托大学生创新训练中心的课外科技活动成果以论文为形式,在很大程度上提升了独立学院学生的创新能力,受益学生每学年约500人次。结论:依托一个具备较成熟的课外科技活动实施体系的大学生创新训练中心,开设一些高水平实验,在很大程度上能够提升独立学院学生创新能力。

摘要:
In intervertebral disc degeneration (IDD), increased proinflammatory molecules secreted by human nucleus pulposus cells (HNPCs) could promote the expression of extracellular matrix (ECM)-degrading enzymes. IDD could be affected by both genetic and environmental factors, including microRNAs (miRNAs). Aucubin, the active ingredient of a traditional Chinese medicine herb Du Zhong, has been reported to promote osteogenic differentiation; however, the role of aucubin in IDD and the underlying mechanism remain unclear. Herein, we evaluated the effect of aucubin on TNF-alpha- or IL-1beta-induced ECM degradation in HNPCs. By using online tools, miR-140 was selected as a candidate miRNA that is related to TNF-alpha or IL-1beta signaling. Overexpression of miR-140 enhanced the effect of aucubin on ECM degradation. Moreover, cAMP responsive element binding protein 1 (CREB1), a major transcriptional factor in immune-related signaling, was a direct downstream target of miR-140. CREB1 knockdown mimicked the function of miR-140 overexpression on ECM degradation. In summary, aucubin might ameliorate IL-1beta- or TNF-alpha-induced ECM degradation in HNPCs through regulating miR-140/CREB1.

摘要:
Froth flotation has been proved to be a promising approach for commercial scale harvesting of microalgae. However, all the surfactants used in the microalgae flotation harvesting process are conventional monomeric surfactants contain a single similar hydrophobic group in the molecule, which results in a low harvesting efficiency. In this work, a novel Gemini surfactant, N,N′-bis(cetyldimethyl)-1,4-butane diammonium dibromide (BCBD) was prepared, and originally recommended as a collector for froth flotation harvesting of Chlorella vulgaris from culture medium. The performance of BCBD was compared with the results acquired using its conventional monomeric surfactant cetyl trimethyl ammonium bromide (CTAB). The bench-scale flotation results showed that BCBD had excellent collecting power for Chlorella vulgaris. Achieving the obviously superior flotation harvesting performance (flotation recovery increased by 21.4% and enrichment ratio increased by 22.9), the dosage of Gemini type BCBD collector is five times less than that of monomeric CTAB collector. © 2018 Elsevier Ltd

摘要:
Deletions or some mutations in the gene encoding the multifunctional protein, DJ-1, have been considered to be linked with autosomal recessive early onset Parkinson's disease (PD). Current emerging evidence suggests that DJ-1 is involved in the protection against oxidative stress-induced mitochondrial damage. However, the exact molecular mechanisms underlying this are not completely clear. The aim of this study was to investigate the effects of DJ-1 on the Akt pathway, nuclear factor erythroid 2-related factor (Nrf2), and c-Jun N-terminal kinase (JNK) with regard to modulating mitochondrial function. Here we showed that knockdown of DJ-1 resulted in mitochondrial dysfunction, including a decrease in active mitochondrial mass, complex I deficits, and inhibition of cellular adenosine 5′-triphosphate (ATP) content in the dopaminergic neuron-like cells PC12 and SH-SY5Y. Additionally, loss of DJ-1 impaired Akt signaling, and reduced nuclear translocation of Nrf2, thereby inhibiting activity of Nrf2-regulated downstream antioxidant enzymes such as heme oxygenase-1 and NAD(P)H quinone oxidoreductase 1. Moreover, DJ-1 knockdown also led to a significant increase in the mitochondrial reactive oxygen species, and then promoted the activation of JNK pathways. Furthermore, oxidative stress and mitochondrial dysfunction induced by knockdown of DJ-1 were blocked by a JNK inhibitor, which confirmed the important role of JNK activation in mitochondrial dysfunction. In conclusion, the present study indicates that DJ-1 knockdown leads to mitochondrial dysfunction in dopaminergic neuron-like cells, at least in part, through suppressing the Akt/GSK3β pathway and impairing the oxidative stress response, as well as through the subsequent increased JNK activation in dopaminergic neuron-like cells. © 2019 Elsevier Inc.

目的研究从中药黄芩干燥根中提取分离药效成分黄芩苷体外对耐甲氧西林金黄色葡萄球菌活性的影响.方法以琼脂扩散法、液体稀释微孔板法检测黄芩苷对MRSA的抑菌环大小、最低抑菌浓度(MIC)和最低杀菌浓度(MBC) ;以96孔酶标板结晶紫法检测黄芩苷对MRSA的最小生物膜清除浓度(MBEC) .结果与DMSO比较,黄芩苷与氯霉素(阳性对照药物)对MRSA的抑菌环半径显著增加(P < 0. 01) ,与氯霉素比较,黄芩苷对MRSA的抑菌环半径显著缩小(P < 0. 05) ;黄芩苷对MRSA的MIC为0. 5mg /ml; MBC为1mg /ml;黄芩苷对MRSA的MBEC为0. 5mg /ml.结论黄芩苷对MRSA有一定的抗菌、抑菌作用,其效应机制可能与其抑制细菌生物膜的形成有关.

湖南省城市高血压病患者住院费用对比分析

摘要:
[目的]对比分析湖南省综合医院和中医医院城市高血压病患者住院费用及其影响因素,为控制高血压病患者住院费用提供依据.[方法]收集湖南省长沙、 岳阳、 永州、 衡阳综合医院和中医医院高血压病患者住院费用的基本构成情况,进行对比分析;采用多因素线性回归法对住院费用的影响因素进行分析.[结果]有效样本8,312例,其中中医医院样本4,695例(56%),综合医院样本3,617例(44%).住院费用的中位数分别为84,808.74元和6,968.405元.影响中医医院患者住院费用的主要因素是住院天数、 支付方式、 年龄;影响综合医院患者住院总费用的主要因素主要是支付方式和住院天数.[结论]综合医院的患者住院总费用大于中医医院的患者住院总费用;不同类型医院药品费占比均较高;住院费用随患者年龄的增加而增加;住院天数是影响住院费用增长的重要因素.

采用1,1-二苯基-2-三硝基苯肼(DPPH)法、β-胡萝卜素漂白法和清除一氧化氮自由基(NO)法来测定不同极性铁苋菜萃取物的抗氧化活性;通过微孔比浊法比较铁苋菜萃取物对3种革兰氏阴性细菌(大肠杆菌、铜绿假单胞菌和幽门螺杆菌)和4种革兰氏阳性细菌(表皮葡萄球菌、枯草芽孢杆菌、藤黄微球菌和金黄色葡萄球菌)的抑菌效果.结果表明,铁苋菜不同极性萃取物均有较好的清除DPPH、NO自由基和抑制β-胡萝卜素漂白的效果,浓度为2.0 mg/mL时,正丁醇萃取物清除DPPH自由基和抑制β-胡萝卜素漂白能力最佳,DPPH自由基清除率为94.60%,β-胡萝卜素抑制率为83.15%,乙酸乙酯萃取物清除NO自由基能力最强,清除率为82.72%.抑菌试验结果显示,铁苋菜各萃取物均有较好抑菌效果,对革兰氏阳性菌抑菌效果优于革兰氏阴性菌,其中石油醚萃取物对大肠杆菌、表皮葡萄球菌和金黄色葡萄球菌抑制作用最佳,MIC值分别为0.625,0.315,1.250 mg/mL.铁苋菜萃取物具有较好的抗氧化及抑菌效果.

摘要:
Objective: To explore the pharmacodynamic mechanism of Fu Zi Li Zhong Decoction (FZLZD) in treating gastric ulcer and its effect on gastrointestinal microecology. Methods: Rat gastric ulcer model was established by continuous vinegar gavage. The experiment included normal group, model group, ranitidine group and FZLZD group. The animals were handled after intragastricing administration of corresponding clinical isodose drugs for 7 d. The rats’ gastrointestinal histopathology was observed by HE staining. The expression of ZO-1 and Occludin mRNA in rat gastric tissue was detected by RT-PCR. The diversity of intestinal flora was analyzed by 16SrDNA sequencing. Results: FZLZD can effectively improve gastric mucosal destruction, epithelial exfoliation, inflammatory cell infiltration, inflammatory substance exudation and edema in rats with gastric ulcer. FZLZD can effectively up-regulate the expression levels of ZO-1 and Occludin mRNA in gastric tissue of rats with gastric ulcer. The results of intestinal flora diversity showed that the diversity of intestinal flora in FZLZD group was significantly higher than that of model group. Conclusions: FZLZD can regulate gastric ulcer by promoting the expression of ZO-1 and Occludin and improving the diversity of intestinal flora. © 2019 Digital Chinese Medicine

摘要:
Potash fertilizer ensures the necessary agricultural yields to feed an increasing world population. Over 80% of the world's potash fertilizer is produced by the selective froth flotation of KCl (sylvite) from NaCl (halite). However, all the surfactants used in the potash froth flotation process are traditional monomeric surfactants contain a single similar hydrophobic group in the molecule, which leads to a low production efficiency. In this work, a novel Gemini surfactant, N,N’-bis (octadecyldimethyl)-1,4-butane diammonium dibromide (BOBD) was prepared, and originally recommended as a collector for froth flotation separation of sylvite from halite for potash ore. The performance of BOBD was compared with the results acquired using its traditional monomeric surfactant octadecylamine hydrochloride (OAH). The bench-scale flotation results showed that BOBD had excellent collecting power for sylvite and significant selectivity against halite at natural pH. Moreover, BOBD presented stronger desalination efficiency than OAH. The results gained with the Gemini collector were better in terms of KCl recovery and NaCl content than those gained with four times higher dosages of the conventional monomeric surfactant. © 2018 Elsevier Ltd

摘要:
随着实验教学在基础医学类教学中占据的位置越来越重要,在实验室各项制度不断完善的同时,实验室安全问题也越来越凸显。本文针对我校基础医学实验室的相关教学现状和存在的问题进行了归纳,发现人为因素和设备物品是影响实验室安全最主要的隐患,针对这些问题,我校对实验室的安全管理提出了一些措施,主要包括在废弃物的处理,试剂及设备的安全问题以及在教育上等几个方面,同时,我校积极引进了虚拟仿真实验平台,该平台的推广不仅提高了学生的学习效率,同时也减少了很多实验室安全问题,但目前其应用还不广泛,下一步希望扩大其应用,以实现效果最大化。

摘要:
中西医结合研究生培养作为医学研究生培养的重要组成部分,在医学教育研究领域占有重要地位。我校基于中西医结合研究生人才培养要求和课程体系特点,针对中西医结合学术型研究生开设医学基本实验技能训练课程。从课程内容设置、课程模式到考核方法、课程实践效果及课程评价进行了全新探索实践,以期提高研究生科研基本理论素养与科研能力,加强科研道德教育,探索出合理的中西医结合学术型研究生培养模式,切实提高中西医结合人才培养质量。

目的 探讨基于p38 MAPK信号通路研究养阴润目丸治疗干眼兔的作用机制.方法 采用氢溴酸东莨菪碱皮下注射途径制备干眼兔模型,按随机数字表法分成模型组、p38抑制剂组、阳性药物(杞菊地黄丸)组和低、中、高剂量养阴润目丸组,同步设不造模的正常组,6只/组.各组按相应处理方式连续干预2周后,检测各组兔泪液分泌量、泪腺病理形态、泪腺组织凋亡细胞的表达及泪腺组织中p38 MAPK、Bax、Bcl-2蛋白和mRNA表达情况.结果 养阴润目丸组和阳性药物组能增加干眼兔模型泪液分泌量,抑制泪腺细胞的凋亡,下调p38 MAPK、Bax的蛋白和mRNA表达,上调Bcl-2的蛋白和mRNA表达水平,与模型组比较,差异均有统计学意义(P<0.05).结论 养阴润目丸能够通过纠正Bax/Bcl-2失衡,改善干眼泪腺细胞凋亡,进而减轻泪腺细胞损伤,恢复泪腺细胞的代谢功能,与调控p38 MAPK的关系极为密切,这可能是其治疗干眼的重要机制之一.

摘要:
外泌体是细胞膜衍生出的一类通过在细胞间传递RNA和蛋白质来调控细胞间通讯的小囊泡。研究发现,当细胞受到压力释放外泌体后,可加重各种病理变化的进程,如病毒的感染过程。而外泌体microRNA(miRNA)在病毒感染及产生炎症时又起着调控作用。流感病毒感染是引起流行性感冒及病毒性肺炎的重要原因,感染机制复杂且尚未得到全面阐述。流感病毒可入侵人体呼吸道,多引发肺部炎症加重肺部损伤,甚至导致肺衰竭。本文总结目前研究进展,重点阐述了外泌体在流感病毒感染中的作用机制及对肺部炎症的影响,为后续研究提供了相关思路及理论指导,以期为流感病毒引起的疾病带来新的有效诊疗方法。

目的 探索如何依托“互联网+”平台实现在中医院校内《临床分子生物学检验》实验课程的优化.方法 以互联网为基础,依托名师空间课堂和中医药虚拟仿真实验平台,运用多种教学方式和资源对《临床分子生物学检验》实验课内容进行整合,提高《临床分子生物学检验》实验课程的教学质量.结果 通过互联网媒介平台,形成微课、微视频、PPT、数字化教材等多样化教学手段的开发和应用,有效提高了中医院校医学检验技术专业医学生的科研思维能力,为中医药创新发展培养优秀人才提供了方案.结论 此种探索模式培养了学生独立思考、自主设计实验项目的创新能力,但后期更为专业的教学模式构建仍需不断努力与探索.
