Curcumin inhibits the growth of liver cancer by impairing myeloid-derived suppressor cells in murine tumor tissues

Curcumin, one of the active ingredients of Curcuma longa (Jianghuang), has been reported to exert multiple bioactivities, including pro-apoptotic and anti-inflammatory activities. In recent years, curcumin has been extensively studied, and it has been revealed that curcumin inhibits the growth of numerous types of cancer. However, to the best of our knowledge, the inhibitory effects of curcumin on the activation or expansion of myeloid-derived suppressor cells (MDSCs) in liver cancer and the underlying mechanism have not yet been determined. Therefore, the present study aimed to investigate the inhibitory effect of curcumin on MDSC activity and the associated anti-neoplastic mechanism in a HepG2 xenograft mouse model. The effect of curcumin on the viability of Huh-7, MHCC-97H and HepG2 cells in vitro was analyzed using a Cell Counting Kit-8 assay. The effects of curcumin on tumor growth, numbers of MDSCs, expres- sion levels of proteins involved in the toll-like receptor 4 (TLR4)/NF-κB signaling pathway, levels of related inflam- matory factors and angiogenesis were determined in HepG2 xenograft model mice, which were given different doses of curcumin via intragastrical administration. The results of the present study revealed that curcumin inhibited the viability of Huh-7, MHCC-97H and HepG2 cells and the growth of HepG2 xenograft tumors in mice. Flow cytometric analysis indicated that curcumin reduced the number of MDSCs in mouse xenograft tumors. In addition, the results demonstrated that curcumin inhibited the TLR4/NF-κB signaling pathway and the expression of inflammatory factors, including IL-6, IL-1β, prostaglandin E2 and cyclooxygenase-2, in mouse xenograft tumors. Furthermore, curcumin suppressed the secretion of granulocyte-macrophage colony-stimulating factor (GM-CSF) and granulocyte-colony stimulating factor (G-CSF), which are essential factors for MDSCs modulation, in tumor tissues. Additionally, curcumin was revealed to inhibit angiogenesis, which was demonstrated by the downregulation of the expres- sion levels of vascular endothelial growth factor, CD31 and α-smooth muscle actin in western blotting, immunohistochem- istry and immunofluorescence experiments. In conclusion, the findings of the present study identified a novel mechanism via which curcumin may suppress the growth of liver cancer by reducing the numbers of MDSCs and subsequently disrupting the process of angiogenesis. These conclusions were supported by the observed inactivation of the TLR4/NF-κB signaling pathway-mediated inflammatory response and the downregu- lation of GM-CSF and G-CSF secretion in xenograft tissues. © 2021 Spandidos Publications. 慢性前列腺炎是男性泌尿生殖系统的常见病、多发病,其病因及发病机制复杂,尚未完全明确,临床表现多样,尚无彻底根治的方法,易于迁延反复,缠绵难愈,中医药对慢性前列腺炎有较好疗效。中医学认为"通则不痛",现代研究证明芳香透窍药麝香、乳香配伍能调控炎症因子,抑制炎症反应,且具有调节前列腺上皮屏障的促通透、促新生作用,从而缓解慢性前列腺炎疼痛症状。基于"通则不痛"理论对芳香透窍药在慢性前列腺炎治疗中缓解疼痛作用进行探讨,为芳香透窍法在慢性前列腺炎中的运用提供一定的临床思路。

摘要:
目的 系统评价西黄丸/西黄胶囊辅助治疗恶性肿瘤的临床有效性和安全性.方法 通过计算机对中国知网(CNKI)、中国医院数字图书馆(CHKD)、中国生物医学文献数据库(CBM)、PubMed外文数据库、EMBASE外文数据库进行检索,搜集关于西黄丸辅助治疗恶性肿瘤的随机对照试验,检索时间从2010年1月至2019年10月.提取资料后采用Cochrane及Jadad评分标准对文献进行质量评价,并采用Review Manager 5.3软件进行Meta分析,应用GRADE系统对结局指标进行质量分级.结果 共纳入17个研究,共1482例患者.Meta分析显示,采用西黄丸/西黄胶囊辅助治疗恶性肿瘤患者,实体瘤疗效评价[RR=1.26,95%CI(1.17,1.36),P<0.05];生活质量评分[MD=10.28,95%CI(7.84,12.72),P<0.05];骨髓抑制反应[RR=0.55,95%CI(0.37,0.84),P<0.05];胃肠道反应[RR=0.6,95%CI(0.49,0.72),P<0.05];肝肾功能异常[RR=0.81,95%CI(0.62,1.06),P=0.12>0.05].GRADE系统显示实体瘤疗效评价、生活质量评分及骨髓抑制反应质量为低等级,胃肠道反应及肝肾功能异常质量为中等级.结论 采用西黄丸/西黄胶囊辅助治疗恶性肿瘤可提高实体瘤疗效及肿瘤患者生活质量,且在产生骨髓抑制反应及胃肠道反应方面低于对照组,但在产生肝肾功能异常方面与对照组相比无差异.

男性迟发性性腺功能减退症(LOH)是中老年男性常见病、多发病,中、西医对其病因病机、诊治等认识存在差异与统一,以睾酮水平低下为研究关键切入点,睾酮补充治疗(TST)是LOH治疗的重要方法之一,但其剂量、疗程、安全性等问题尚未解决。中医药基于辨证论治理论体系,可以整体改善LOH患者症状,具有一定优势。文章同时从Leydig细胞睾酮合成酶、活性氧簇(ROS)、自噬、旁分泌、雄激素受体(AR)、下丘脑-垂体-睾丸轴等多靶点、多途径对中医药干预LOH的可能机制提供研究思路。

摘要:
目的 探讨雄蚕益肾方对迟发性性腺功能减退症(lateonset hypogonadism,LOH)大鼠睾丸组织形态学改变及抗氧化作用.方法 将18月龄SD雄性大鼠随机分为模型组,丙酸睾酮组,雄蚕益肾方低、中、高剂量组,2月龄SD雄性大鼠设为正常组.雄蚕益肾方低、中、高剂量组分别给予10.4、20.8、41.6 g/kg的雄蚕益肾方配方颗粒剂灌胃;丙酸睾酮组肌注5.21 mg/kg丙酸睾酮,隔日1次;正常组和模型组给予相同体积的蒸馏水灌胃,均连续28 d.实验结束后,记录大鼠悬尾实验不动时间,称体质量、采血,ELSIA法检测睾酮(T),睾丸组织进行黄嘌呤氧化酶法(SOD)、紫外吸收法(CAT)、硫代巴比妥酸法(MDA)、病理及电镜检测.结果 与正常组比较,模型组T水平降低(P<0.05),悬尾不动时间延长(P<0.05),睾丸组织抗氧化酶SOD、CAT降低(P<0.05)、MDA升高(P<0.05),HE染色睾丸间质减少,电镜示Leydig细胞线粒体数量减少、水肿明显,均符合LOH改变.与模型组比较,雄蚕益肾方各剂量组和丙酸睾酮组均能提高T水平(P<0.05)、缩短悬尾不动时间(P<0.05),雄蚕益肾方中、高剂量组和丙酸睾酮组均能升高SOD、CAT(P<0.05),降低MDA(P<0.05).雄蚕益肾方各剂量组大鼠睾丸组织生精小管内精子数量增多,睾丸间质增加;电镜示Leydig细胞线粒体数量明显增多,部分可见线粒体鞘,部分线粒体嵴清晰,线粒体水肿不明显.结论 雄蚕益肾方能提升LOH大鼠T水平、缩短悬尾不动时间、一定程度改变睾丸组织形态和超微结构,可能与提高睾丸组织抗氧化能力有关.

Objectives: To develop a dual-modal neural network model to characterize ultrasound (US) images of breast masses. Materials and methods: A combined US B-mode and color Doppler neural network model was developed to classify US images of the breast. Three datasets with breast masses were originally detected and interpreted by 20 experienced radiologists according to Breast Imaging-Reporting and Data System (BI-RADS) lexicon ((1) training set, 103212 masses from 45,433 + 12,519 patients. (2) held-out validation set, 2748 masses from 1197 + 395 patients. (3) test set, 605 masses from 337 + 78 patients). The neural network was first trained on training set. Then, the trained model was tested on a held-out validation set to evaluate agreement on BI-RADS category between the model and the radiologists. In addition, the model and a reader study of 10 radiologists were applied to the test set with biopsy-proven results. To evaluate the performance of the model in benign or malignant classifications, the receiver operating characteristic curve, sensitivities, and specificities were compared. Results: The trained dual-modal model showed favorable agreement with the assessment performed by the radiologists (κ= 0.73; 95% confidence interval, 0.71–0.75) in classifying breast masses into four BI-RADS categories in the validation set. For the binary categorization of benign or malignant breast masses in the test set, the dual-modal model achieved the area under the ROC curve (AUC) of 0.982, while the readers scored an AUC of 0.948 in terms of the ROC convex hull. Conclusion: The dual-modal model can be used to assess breast masses at a level comparable to that of an experienced radiologist. Key Points: • A neural network model based on ultrasonic imaging can classify breast masses into different Breast Imaging-Reporting and Data System categories according to the probability of malignancy. • A combined ultrasonic B-mode and color Doppler neural network model achieved a high level of agreement with the readings of an experienced radiologist and has the potential to automate the routine characterization of breast masses. © 2020, European Society of Radiology.

目的:探讨麝香配伍乳香对慢性前列腺炎(CP)小鼠前列腺组织中P物质(SP)的影响。方法:将6周龄雄性NOD/SCID小鼠随机分为4组:空白组、模型组、麝香-乳香组、麝香-乳香高剂量组。除空白组以外,均建立CP模型。造模20d后根据组别分别予以相应0.9%氯化钠溶液及不同剂量的麝香-乳香灌胃。连续灌胃14d后采用免疫组化法及ELISA法分别检测小鼠前列腺组织及血清中SP的含量。结果:模型组SP染色阳性细胞数较空白组增多(P<0.05),经麝香-乳香干预后,前列腺组织及血清中SP水平较模型组显著下降(P<0.05),与麝香-乳香高剂量组比较,麝香-乳香组SP水平差异无统计学意义。结论:具有神经炎性因子及疼痛神经递质双重属性的SP参与了CP的病理过程;麝香-乳香"祛瘀以通",其"瘀"包含SP,并且其"祛瘀以通"的靶点之一即为SP;麝香-乳香对CP小鼠前列腺组织中SP含量影响无剂量依赖性。

基于线粒体途径研究知柏地黄汤对解脲脲原体感染大鼠生精细胞活力的影响

摘要:
目的:基于线粒体途径探讨知柏地黄汤含药血清干预解脲脲原体(UU)感染大鼠生精细胞活力的影响及其作用机制。方法:采用在体UU感染建模,随后原代提取大鼠生精细胞,以知柏地黄汤含药血清(低、中、高剂量组)为实验施加因素,同时设空白组、强力霉素组和模型组。给予相应药物干预后进行指标检测。结果:知柏地黄汤含药血清能改善UU感染的生精细胞活性,提高存活率(P<0.01, P<0.05);知柏地黄汤含药血清能有效提高UU感染的生精细胞线粒体ANT4转运活性(P<0.01,P<0.05);知柏地黄汤含药血清能有效上调UU感染的生精细胞中VDAC2、Cyp-D蛋白和mRNA表达水平(P<0.01, P<0.05),下调VDAC1蛋白和mRNA表达水平(P<0.01, P<0.05)。结论:知柏地黄汤可能通过抑制UU感染引起的VDAC1的过度激活,上调VDAC2、Cyp-D mRNA和蛋白表达,上调ANT4的转运活性,进而明显改善线粒体结构的破坏,最终改善生精细胞的活力。

Stromal-epithelial interaction serves a pivotal role in normal prostate growth, as well as the onset of benign prostatic hyperplasia (BPH). The present study aimed to explore the role of cyclopamine in the proliferation and apoptosis of epithelial and stromal cells in rats with BPH by blocking the Hedgehog signaling pathway. Cyclopamine (an inhibitor of the Hedgehog signaling pathway) was administered in a rat model of BPH, and the expression of Ki67 (proliferation factor) was determined by immunohistochemistry. In addition, epithelial and stromal cells were separated and cultured in order to investigate the role of cyclopamine in the progression of BPH. The expression of Hedgehog signaling pathway- and apoptosis-related genes, including basic fibroblastic growth factor (b-FGF) and transforming growth factor β (TGF-β), was evaluated using reverse transcription-quantitative polymerase chain reaction and western blot analysis. Cell proliferation, cell cycle and apoptosis were analyzed using an MTT assay and flow cytometry. We identified upregulated Ki67 expression and activated Hedgehog signaling pathway in rats with BPH. Cyclopamine inhibited the activation of the Hedgehog signaling pathway. In response to cyclopamine treatment, epithelial and stromal cell proliferation was inhibited; this was concomitant with decreased Ki67, TGF-β, and b-FGF expression. On the other hand, epithelial cell apoptosis was enhanced, which was associated with increased Bax and reduced Bcl-2 expression. Based on these findings, we proposed that cyclopamine may serve as a potential therapeutic agent in the treatment of BPH. Cyclopamine could inhibit epithelial and stromal cell proliferation, and induce epithelial cell apoptosis by suppressing the Hedgehog signaling pathway. © 2020 Spandidos Publications. 摘要:
目的:探讨雄蚕益肾方对LOH大鼠Leydig细胞胆固醇转运蛋白、睾酮合成酶和SF-1表达的影响.方法:25只18月龄雄性SD大鼠随机均分为模型组、丙酸睾酮组、雄蚕益肾方-低、中、高剂量组,2月龄雄性SD大鼠5只设为正常组.雄蚕益肾方-低、中、高剂量组分别给予10.4、20.8、41.6 g/(kg·d)体重剂量的雄蚕益肾方配方颗粒剂灌胃;丙酸睾酮组大鼠肌注5.21 mg/(kg·d).体重剂量的丙酸睾酮,每周3次;正常组和模型组给予相同体积的蒸馏水灌胃,均连续28 d.实验结束后取睾丸组织进行Western印迹检测StAR、TSPO、CYP11 A1、HSD3B7、HSD17B4和SF-1蛋白表达. 结果:模型组大鼠睾丸组织StAR、TSPO、CYP11A1、HSD3B7、HSD17B4蛋白和SF-1表达均显著低于正常组(P<0.05);与模型组相比,雄蚕益肾方-低、中、高各剂量组大鼠睾丸组织StAR、TSPO、CYP11A1、HSD3B7、HSD17B4蛋白和SF-1显著增高(P<0.05). 结论:雄蚕益肾方能上调LOH大鼠睾丸组织胆固醇转运蛋白(StAR、TSPO)、睾酮合成酶(CYP11 A1、HSD3B7、HSD17B4)以及上游转录因子SF-1表达水平,可能是其干预LOH的机制之一.

摘要:
目的:分析湖南部分地区前列腺癌患者流行病学特征,为防控策略的制定提供参考。方法:收集2010年1月1日~2019年12月31日就诊中南大学湘雅医院、湖南中医药大学第一附属医院、湖南中医药研究院附属医院的2 877例前列腺癌患者年龄、病理类型和中医证型资料,分析探讨前列腺癌发病现状及流行趋势。结果: 2010年291例,2011年315例,2012年213例,2013年220例,2014年159例,2015年226例,2016年199例,2017年180例,2018年577例,2019年497例。患病人群年龄资料显示:40岁以下罕见,65~79岁呈高发病率。年龄组变化趋势为:65~79岁年龄组发病比例逐年增加,10年间增加至63.9%;80岁及以上组发病上升更明显,10年间增长率达到97.9%。病理分型中主要为腺癌。腺癌1441例(50.1%),腺泡细胞腺癌例201例(7.00% );滤泡状腺癌37例(1.29% ),前列腺导管癌27例(0.94%),病例未特指273例(9.49%),其他腺癌166例(5.77%)。其中157例患者进行了中医辨证分型,主要为肾虚证型,其中肾阴虚者40例(25.5%),肾阳虚69例(43.9%)。结论: 2010~2019年湖南地区前列腺癌发病呈老龄化趋势,65~69岁为发病峰值,65~79老年男性为高危人群,需加强健康宣教和早诊早治工作,降低其发病率。

摘要:
BACKGROUND Liver cancer is the sixth most frequently occurring cancer in the world and the fourth most common cause of cancer mortality. The pathogenesis of liver cancer is closely associated with inflammation and immune response in the tumor microenvironment. New therapeutic agents for liver cancer, which can control inflammation and restore cellular immunity, are required. Curcumin (Cur) is a natural anti-inflammatory drug, and total ginsenosides (TG) are a commonly used immunoregulatory drug. Of note, both Cur and TG have been shown to exert anti-liver cancer effects. AIM To determine the synergistic immunomodulatory and anti-inflammatory effects of Cur combined with TG in a mouse model of subcutaneous liver cancer. METHODS A subcutaneous liver cancer model was established in BALB/c mice by a subcutaneous injection of hepatoma cell line. Animals were treated with Cur (200 mg/kg per day), TG (104 mg/kg per day or 520 mg/kg per day), the combination of Cur (200 mg/kg per day) and TG (104 mg/kg per day or 520 mg/kg per day), or 5-fluorouracil combined with cisplatin as a positive control for 21 d. Tumor volume was measured and the protein expression of programmed cell death 1 and programmed cell death 1 ligand 1 (PD-L1), inflammatory indicators Toll like receptor 4 (TLR4) and nuclear factor-kappa B (NF-kappa B), and vascular growth-related factors nitric oxide synthases (iNOS) and matrix metalloproteinase 9 were analyzed by Western blot analysis. CD4(+)CD25(+)Foxp3(+) regulatory T cells (Tregs) were counted by flow cytometry. RESULTS The combination therapy of Cur and TG significantly inhibited the growth of liver cancer, as compared to vehicle-treated animals, and TG showed dose dependence. Cur combined with TG-520 markedly decreased the protein expression of PD-L1 (P < 0.0001), while CD4(+)CD25(+)Foxp3(+) Tregs regulated by the PD-L1 signaling pathway exhibited a positive correlation with PD-L1. Cur combined with TG-520 also inhibited the cascade action mediated by NF-kappa B (P < 0.0001), thus inhibiting the TLR4/NF-kappa B signalling pathway (P = 0.0088, P < 0.0001), which is associated with inflammation and acts on PD-L1. It also inhibited the NF-kappa B-MMP9 signalling pathway (P < 0.0001), which is associated with tumor angiogenesis. CONCLUSION Cur combined with TG regulates immune escape through the PD-L1 pathway and inhibits liver cancer growth through NF-kappa B-mediated inflammation and angiogenesis.

目的:挖掘中国知网数据库(CNKI)中治疗前列腺癌的中药复方,并尝试探讨前列腺癌的用药规律及病机治法。方法:收集近10年中国知网数据库收录的治疗前列腺癌文献,按照纳入及排除标准对文献进行筛选,将纳入文献中的中药复方导入Excel 2007并建立数据库,运用中医传承辅助平台(V2.5)统计中药复方的药物及四气五味归经频次、药物组合频次并分析用药规律和提炼本病的药物核心组合及聚类新处方,最后总结并探讨前列腺癌的病机治法。结果:共收集到80个治疗前列腺癌的中药复方共有177味中药纳入研究,以补虚药、利水渗湿药、清热药与活血化瘀药为主,出现频次最高的中药是黄芪。药物四气以温寒平为主,五味以辛苦甘为主,归经以肝脾肾经为主。频次最高的药对是“白术-茯苓”频次为22次,关联规则分析显示置信度为1的有6条,并聚类获得6对核心药物组合、3首新处方。结论:本虚标实是前列腺癌发病的重要病机,扶正法贯穿始终,以“扶正-活血-解毒”为主轴,注重调理肝脾肾三经,辨证采用活血化瘀、清热利湿、通窍解毒为主线的治法,可为中医临床治疗前列腺癌提供用药参考,以通窍活血逐瘀法为主治疗前列腺癌的聚类新处方是日后值得研究的方向。

目的:在"以学生为中心"的基础上,将基于问题学习(problem-based learning,PBL)教学法结合形成性评价应用于中医外科临床教学中,以提升中医外科教学效果及学生的临床学习能力.方法:将80名中医外科临床实习学生分为试验组、对照组各40名,两组均进行PBL教学,对照组实施一次性终结考核评价;试验组实施形成性评价,包括闭卷测试、临床操作能力、教师评价、学生自评与互评等,并进行满意度评估.结果:对照组自评量表总分为(45.00±2.12)分,对照组学生认为PBL教学结合一次性终结评价主要培养中医外科思维能力,约有83%的学生认同;试验组自评量表总分为(78.00±3.11)分,且在提高学习积极性、兴趣、团队协作、临床操作能力、查阅文献能力及分析解决问题能力方面效果明显(P<0.05).两组学生均对PBL教学方式及增强理论知识记忆力评价较高,两组比较差异无统计学意义(P>0.05).结论:PBL教学结合形成性评价体系应用于中医外科临床教学,不仅能促进教师的教学效果,且能提高学生的临床学习效果.

Objective: The high mortality rate associated with acute respiratory distress syndrome (ARDS) is a major challenge for intensive care units. In the present study, we applied bioinformatics and animal models to identify core genes and potential corresponding pathways in ARDS. Results: Using bioinformatics analysis, IL-1 beta was identified as the core gene of ARDS. Cell experiments showed that up-regulation of IL-1 beta downregulates claudin18 to promote lung barrier function damage by regulating the IL-1 beta-HER2/HER3 axis, further promoting the development of ARDS. This was validated in the animal models. Conclusion: IL-1 beta promotes the development of ARDS by regulating the IL-1 beta-HER2/HER3 axis. These findings deepen the understanding of the pathological mechanisms of ARDS. Methods: Transcription data sets related to ARDS were subjected to differential expression gene analysis, functional enrichment analysis, and receiver operating characteristic curve analysis and, so as to identify core genes in ARDS. Cell experiments were used to further explore the effects of core genes on lung barrier function damage. Animal models were applied to validate the effects of core gene in mediating biological signal pathways in ARDS.
