摘要:
Biofilms are the main reason for a large number deaths and high health costs. Their better protection compared to planktonic form against conventional antibiotics leads to poor treatment efficiency. Nanoagent-targeted delivery is a promising avenue for disease therapeutic, but its application targeting biofilms has not been reported currently. The roles, if any, of aptamers acting as delivery carrier and targeting factor, the graphene oxide (GO), and GO modified with aptamers against biofilms were then systematically evaluated. Here, we successfully developed an aptamer-targeted GO strategy against biofilms. We investigated the efficacy of aptamer–GO conjugates by UV spectrophotometer, inverted microscopy, and atomic force microscopy; 93.5 ± 3.4% Salmonella typhimurium biofilms were inhibited and 84.6 ± 5.1% of biofilms were dispersed by a ST-3-GO conjugate. More importantly, this conjugate represented distinctively toxicity to S. typhimurium. Thus, this strategy significantly displays excellent antibiofilm properties and may serve as a long-term solution for biofilm control. © 2018 International Union of Biochemistry and Molecular Biology, Inc.

摘要:
探索适应高等医学人才培养要求的形成性评价考核体系是一个亟待解决的问题。为客观、合理地评价医学生对生物化学的学习能力和掌握程度,湖南中医药大学生物化学教研室结合微信公众平台改革现有形成性评价体系,构建了由分章随堂测试、阶段性测试、实验成绩考核和终结性测试为主体的生物化学形成性评价体系。实践证明,该评价体系可以及时、有效地对学生学习过程进行合理评价,有利于促进学生主动学习,提高学生学习效果和综合素质,提升生物化学教学质量。

摘要:
Biofilm formation involves the development of extracellular matrix and initially depends on adherence and tropism by flagellar movement. With the widespread development of antibiotic resistance and tolerance of biofilms, there is a growing need for novel anti-infective strategies. No currently approved medications specifically target biofilms. Aptamers are single-stranded nucleic acid molecules that may bind to their targets with high affinity and affect the target functions. We developed a bifunctional conjugate by linking an aptamer targeting bacterial flagella with ampicillin. We investigated its influence on biofilm prevention and dissolution by ultraviolet–visible spectrophotometry, inverted microscopy, and atomic force microscopy. This conjugate had distinctive antibacterial activity. Notably, the conjugate was more active than either component, and thus had a synergistic effect against biofilms. © 2017 Elsevier Inc.

摘要:
The authors describe an aptamer based fluorometric assay for the determination of ATP. It is based on deoxyribonuclease I-aided target recycling and signal amplification. The DNA probe consists of two regions (sequences) that represent the capture probe and the signal probe, respectively. In the absence of ATP, the probe is adsorbed by the surface of graphene oxide (GO) via π-stacking interactions. This results in quenching of the fluorescent label (carboxyfluorescein) and protects it from being cleaved by DNase. Upon adding ATP, the probe will be repelled by GO because ATP binds to the aptamer. This triggers an increase in fluorescence as measured at excitation/emission wavelengths of 480/514 nm. The detection limit is as low as 0.2 nM, and the calibration plot is linear in the 10 to 400 nM ATP concentration range. The assay is specific and sensitive, and in our perception has a large potential in terms of detecting other species including pathogenic microorganisms, small molecules, metal ions, and proteins. 摘要:
Excessive autophagic activity of alveolar type II epithelial (AT-II) cells is one of the main causes of acute lung injury (ALI); however, the underlying molecular mechanism remains to be determined. The microRNAs (miRNAs) are involved with autophagy in many diseases. The objective of this study was therefore to investigate the relationship between the miRNA expression and the autophagic activity of the AT-II cells in the pathogenesis of ALI and its molecular mechanism. A mouse model of ALI and AT-II cell injury was induced using lipopolysaccharide (LPS) in vivo and in vitro, and the expression of miR-34a and the autophagy-related proteins LC3 II/I and p62 were determined. Moreover, the autophagic activity was investigated after miR-34a overexpression and inhibition. The effects of miR-34a on its target gene, FoxO3, in regulating autophagic activity in AT-II cells were also determined. LPS induced autophagic activity and increased the expression of miR-34a in lung tissues and in AT-II cells. The in vitro results showed that the upregulation of miR-34a suppressed, whereas the inhibition of miR-34a promoted, autophagy in AT-II cells. Moreover, miR-34a could directly bind to the 3′-untranslated region of the autophagy-related gene, FoxO3, to decrease its expression. In addition, the knockdown of FoxO3 expression inhibited the autophagic activity in AT-II cells. Together, this study suggested that miR-34a might suppress the excessive autophagic activity in AT-II cells via targeting FoxO3 to reduce the damage of LPS-induced ALI. © 2017, Springer Science+Business Media New York.

目的:将翻转课堂应用于中医院校分子生物学教学并评价其效果。方法实验组和对照组分别采用翻转课堂与传统教学模式,通过问卷调查、闭卷考试评价其教学效果。结果采用翻转课堂教学模式,学生学习效果与解决问题、自主学习能力等均显著提高。结论翻转课堂教学模式加强了教师在教学过程中对学生的引导,极大地激发了学生学习主动性。将翻转课堂引入分子生物学教学是提高学生学习效果和综合素质的有效途径,是深化分子生物学教学改革的重要手段之一。

摘要:
为在有限的课时中围绕教学目标提高医学分子生物学教学质量,将形成性评价考核应用于医学分子生物学课程中,并采用期末考试、实验技能考核、撰写小综述及课堂提问等多种考核形式考查学生成绩。结果显示形成性评价考核较传统考核明显提高学生平时成绩,学生的综合成绩中优秀率、良好率和中等率均高于传统考核;而其及格率和不及格率均低于传统考核。因此,形成性评价考核能够增强教学效果,促进医学分子生物学教学质量提高。

Rosuvastatin, a 3-hydroxy-3-methylglutaryl-coenzyme A reductase inhibitor used to lower blood low-density lipoprotein cholesterol, is a substrate of the membrane ABCG2 exporter. ABCG2 variants have been shown to alter rosuvastatin disposition. The objective of this study is to determine the impact of ABCG2 34/421 compound haplotypes on rosuvastatin pharmacokinetics in healthy Chinese volunteer subjects. Eight hundred healthy Chinese males were genotyped by polymerase chain reaction-pyrosequencing for ABCG2 34G > A, ABCG2 421C > A, SLCO1B1 521T > C, andCYP2C9∗3 variants. Sixty-twomale subjects with wild-type SLCO1B1 c.521TT and CYP2C9∗3 were recruited for this pharmacokinetic study of rosuvastatin. A single oral dose of 10 mg rosuvastatin was administrated to each subject, and blood samples were collected before and at various time points after drug administration. Plasma concentration of rosuvastatin was determined by high-performance liquid chromatography-tandem mass spectrometry, and pharmacokinetic analysis was carried out using the WinNonlin program. In Chinese males, high allele frequency of ABCG2 c.34G > A (0.275) and c.421C > A (0.282) was observed, resulting in a considerable portion (23.3%) of subjects being ABCG2 34/421 compound heterozygotes. Compared with subjects with ABCG2 wild-type (c.34GG/421CC), plasma rosuvastatin Cmax and area under the curve, AUC0-∞, were significantly higher, while the apparent oral clearance, CL/F, was significantly lower in subjects with c.34AA, c.421AA, and c.34GA/ 421CA genotypes. Both t1/2 and Tmax were similar among subjects with different genotypes. A high frequency of ABCG2 c.34G > A and c.421C > A variants was present in Chinese males, and the disposition of rosuvastatin was significantly affected by both variants. These data suggest that it is advisable to genotype these variants when prescribing rosuvastatin to Chinese subjects, leading to a precise dose for each individual. 内质网应激相关基因在2型糖尿病合并脂肪肝小鼠肝脏中的表达变化

目的:探讨内质网应激(ERS)在高脂饮食2型糖尿病转基因MKR小鼠脂肪肝发生中的作用。方法高脂饲料诱发MKR鼠形成脂肪肝,观察肝脏形态变化,检测肝脏甘油三酯和脂肪酸水平,RT-PCR检测小鼠肝脏内质网应激相关基因mRNA表达变化。结果与正常小鼠比较, MKR组小鼠肝脏GRP78、XBP1、FAS、ACC-α、GSK3βmRNA表达水平升高(P<0.05);与MKR小鼠比较,高脂饮食MKR小鼠肝脏甘油三酯(TG)和脂肪酸(FFA)水平显著升高(P<0.01),肝脏GRP78、XBP1、CHOP、SREBP1c 、FAS、ACC-α、GSK3β和EDEM1基因mRNA表达水平均增加(P<0.01),而ApoB100 mRNA表达水平降低(P<0.01)。结论 ERS参与调节肝脏脂质及 ApoB100的代谢过程,在转基因2型糖尿病 MKR鼠脂肪肝形成中发挥重要作用。

骨性关节炎是一种以关节软骨退行性病变和关节周围骨质增生为病理特征的慢性退行性疾病。研究表明,软骨细胞凋亡在骨性关节炎发生发展中起到关键性作用,有望成为骨关节炎治疗的潜在靶点。该文就骨关节炎中软骨细胞凋亡相关的三条信号通路及针对上述信号通路中关键分子的中医药治疗作简要综述,为骨关节炎的中医药防治提供理论依据。

Biofilms are microbial communities that are attached to a solid surface using extracellular polymeric substances. Motility and initial attachment mediated by flagella are required for biofilm formation. Therefore, blocking the motility of flagella is a potential strategy to inhibit biofilm formation. In this study, single-stranded DNA aptamers specific to the Salmonella choleraesuis were selected after 14 cycles of the systematic evolution of ligands by exponential enrichment (SELEX). Among the selected aptamers, the aptamer 3 showed the highest affinity for S. choleraesuis with a dissociation constant (Kd) of 41 ± 2 nM. Aptamer 3, conjugated with magnetic beads, was then used to capture its binding target on the bacteria. After mass spectrometry and specific binding analysis, the flagellin was identified as the target captured by aptamer 3. Furthermore, inhibition experiments, inverted microscopy and atomic force microscopy demonstrated that aptamer 3 was able to control the biofilm formation and promote the inhibitory effect of an antibiotic on bacterial biofilms.
