Since the fluctuation of cellular hydrogen sulfide (H2S) is a very important third endogenously generated gaseous signaling molecule and plays a key role in the development of numerous human disorders, the real-time fluorescence detection of H2S in living systems has attracted plenty of interest during past decade. Although a lot of H2S fluorescent probes have been reported, the relationship between the physiology and pathology of H2S in organelles remains unclear, especially for inflammatory tissue. In this work, by adopting a weakly basic morpholine group as the lysosome-targeting site, a naphthalimide derivative as the signal reporter group and a 4-dinitrobenzene-ether (DNB) as fluorescence signal quencher and H2S-selective recognition moiety, we reported a new lysosome-targeting TP fluorescent probe LyNP-H2S for H2S detection and imaging in living cells and inflamed tissues. The probe LyNP-H2S exhibits very low fluorescence signal in the absence of H2S, and displays a significant 262-fold fluorescence intensity enhancement in the presence of H2S at 540 nm. Moreover, LyNP-H2S has the capability of quantitative detection of H2S at concentrations ranging from 0 to 12.0 mu M (limit of detection = 9.8 nM), rapid response, as well as high sensitivity and selectivity toward H2S. Impressively, the results of living cell and inflamed tissues imaging test demonstrate that LyNP-H2S has the potentiality of being an ideal probe for real-time H2S detection in biosystems. Background. Continuous wavelet transform (CWT) based scalogram can be used for photoplethysmography (PPG) signal transformation to classify blood pressure (BP) with deep learning. We aimed to investigate the determinants that can improve the accuracy of BP classification based on PPG and deep learning and establish a better algorithm for the prediction. Methods. The dataset from PhysioNet was accessed to extract raw PPG signals for testing and its corresponding BPs as category labels. The BP category of normal or abnormal followed the criteria of the 2017 American College of Cardiology/American Heart Association (ACC/AHA) Hypertension Guidelines. The PPG signals were transformed into 224 * 224 * 3-pixel scalogram via different CWTs and segment units. All of them are fed into different convolutional neural networks (CNN) for training and validation. The receiver-operating characteristic and loss and accuracy curves were used to evaluate and compare the performance of different methods. Results. Both wavelet type and segment length could affect the accuracy, and Cgau1 wavelet and segment-300 revealed the best performance (accuracy 90%) without obvious overfitting. This method performed better than previously reported MATLAB Morse wavelet transformed scalogram on both of our proposed CNN and CNN-GoogLeNet. Conclusions. We have established a new algorithm with high accuracy to predict BP classification from PPG via matching of CWT type and segment length, which is a promising solution for rapid prediction of BP classification from real-time processing of PPG signal on a wearable device.

The quality of honeysuckle is typically degraded seriously by unscrupulous mixing with adulterating substances, especially salt. This study proposes a model based on fourier-transform mid-infrared (Mid-IR) spectroscopy and diode-array near-infrared (NIR) spectroscopy in combination with chemometric techniques for the identification and quantification of salt-adulterated honeysuckle. For model training, Mid-IR and NIR spectra of 72 sample batches (including 32 batches of salt-free samples and 40 batches of salt-adulterated samples) were collected. After that, honeysuckle sample authentication was performed using the partial least squares discriminant analysis (PLS-DA) model based on NIR spectroscopy data. This model was optimized after processing by 28 types of pretreatment schemes, possessed of 100% correct authentication of salt-free and salt-adulterated sample at 0.5%-20% (W/W) levels. Subsequently, PLS regression models were built for quantify salt adulteration levels. These models were optimized by the synergy interval PLS (SiPLS) and backward interval PLS (BiPLS) algorithms. The SiPLS-optimized mid-IR-based PLS model demonstrated the lowest values of two error metrics, namely, a root-mean-square error of estimation (RMSEE) of 2.38%, and a root-mean-square error of prediction (RMSEP) of 3.14%. Then, optimal PLS models were obtained through applying several variable selection methods, including variable importance for projection (VIP), competitive adaptive reweighted sampling (CARS), the successive projection algorithm (SPA), and uninformative variable elimination (UVE). The results indicated that the SiPLS-VIP multi-dimensional analysis methods showed a maximum RMSEP reduction of 1.02% in comparison with models constructed using the BiPLS or SiPLS algorithms alone. In general, the proposed multi-dimensional analysis methods show a great potential to complement existing methodologies for rapid and effective salt adulteration detection in honeysuckle samples.

Ethnopharmacological relevance: Chaihu-Longgu-Muli Decoction (CLMD) is a classic prescription created by Zhong-jing Zhang, a famous ancient Chinese medical scientist, to harmonize uncontrollable body activities and calm the minds. Now Traditional Chinese Medicine (TCM) physicians often apply it to treat psychiatric diseases such as epilepsy. Aim of the study: This study investigated the mechanism of the effect of Chaihu-Longgu-Muli Decoction (CLMD) on hippocampal neurons pyroptosis in rats with Temporal Lobe Epilepsy (TLE). Materials and methods: The lithium chloride-pilocarpine-induced TLE rat model was established. The behavioral testing was performed and, the expression of IL-1 beta and TNF-alpha in serum was detected by ELISA, qRT-PCR was used to detect the mRNA expression of NLRP3, Caspase-1, IL-1 beta and TNF-alpha in hippocampus. The expression of NLRP3 and Caspase-1 in hippocampal dentate gyrus was detected by immunofluorescence assay. Results: CLMD could significantly suppress the frequency and duration time of epileptic seizures, reduce the expression of NLRP3, Caspase-1 TNF-alpha and IL-1 beta. Conclusions: CLMD exerted an obvious antiepileptic effect by improving pyroptosis in hippocampal neurons of TLE rats.

BACKGROUND: Acupuncture combined with traditional Chinese medicine (TCM) bonesetting is an effective and more acceptable treatment method for distal radius fractures; this study aims to evaluate the clinical efficacy and other relevant factors of it compared with western medicine therapy such as operation. METHODS: Databases CNKI, Wanfang, VIP, SinoMed, and PubMed were searched for the current study. The retrieval time was from the establishment to November 14, 2021. Literature quality was evaluated according to the bias risk assessment criteria of Cochrane Collaboration network. RevMan 5.3 and Stata 12.0 were used to perform this research. RESULTS: This study will appraise the effectiveness and advantages of acupuncture combined with TCM bonesetting for distal radius fractures in terms of excellent and good rate, length of stay, hospitalization expenses, complication, and other factors. CONCLUSION: This study provides reliable evidence-based support for the clinical efficacy and advantages of acupuncture combined with TCM bonesetting for distal radius fractures. The upregulation of proprotein convertase subtilisin/kexin type 9 (PCSK9) was reported to be involved in regulating the levels of inflammatory markers and apoptosis in macrophages. This study aims to investigate the function and regulation of PCSK9 in myocardial ischaemia. The results of our study showed dramatically increased expression of PCSK9 induced by hypoxia/reoxygenation (H/R) stress rather than by apoptosis in primary murine cardiomyocytes and HL-1 cells. Moreover, PCSK9 promoted H/R-induced pro-inflammatory cytokine release from macrophages, while silencing of PCSK9 inhibited the expression of the pro-inflammatory cytokines TNF-alpha, IL-6 and IL-1 beta. Additionally, PCSK9 facilitated the release of pro-inflammatory cytokines from macrophages under H/R conditions, which decreased cardiomyocyte viability and promoted apoptosis of cardiomyocytes. For the underlying mechanisms, we identified PCSK9-induced NF-kappa B activation as being involved in the cardiomyocyte apoptosis, which was blocked by the NF-kappa B inhibitor BAY 11-7082. Collectively, this study provides new insights into the therapeutic possibility of regulating PCSK9 in cardiomyocytes for the treatment of ischaemic cardiomyopathy.

Background: Platycodin D (PLD) comes from the main triterpenoid saponins in the dry roots of Platycodon grandiflorum, which has been used in traditional Chinese medicine in the treatment of respiratory disease. This study aimed to evaluate the therapeutic effects of PLD on mouse alveolar macrophages induced by influenza A virus (IAV) in vitro and to investigate the PLD of its action. Materials and Methods: Raw264.7 mouse alveolar macrophages were grown in culture and the Cell Counting Kit-8 assay determined cell viability. Cells were treated for 24 h with increasing doses of PLD (0, 5, 10, 20, 40, 60, 80, 100 mu M). The apoptosis was detected by Annexin V-FITC/PI and Tunel assay. Levels of interleukin (IL)-1 beta, IL-6 and tumor necrosis factor (TNF)-alpha of supernatants were assessed using enzyme-linked immunosorbent assay. The numbers of phagophores, autophagosomes, and autolysosomes and the ultrastructure of cells were assessed using transmission electron microscope assay. The relative protein expression of microtubule-associated protein light chain (LC) 3I, LC3II, Beclin-1, and B cell lymphoma (Bcl2) was evaluated by Western blotting. The localizations of Beclin-1 and Bcl2 were viewed with a fluorescence microscope. Results: PLD treatment resulted in significant dose-dependent inhibition of the growth of Raw264.7 cells with the half-maximal inhibitory concentration of 115.74 mu M at 24 h. After Raw264.7 cells infected by influenza virus, PLD (2.5, 5, and 10 mu M) can improve the activity of cells, reducing apoptosis of cells, decreasing the concentrations of IL-1 beta, IL-6, and TNF-alpha in the supernatant of cells, reducing autophagic injury of cells and regulating the protein expression of Beclin-1 and Bcl2. Conclusion: PLD could inhibit excessive apoptosis and autophagy of macrophage cells induced by IAV in a dose-dependent manner and then inhibits the secretion of inflammatory factors caused by influenza virus infection caused by apoptosis and autophagy and then ultimately regulate the pathogenesis of influenza virus.

A meta-analysis was performed to explore the association of coronary artery disease (CAD) with bone mineral density (BMD). Low BMD was not found to be associated with prevalence of CAD. Though the BMD of CADs is significantly lower than that of non-CADs, the confounding effect of aging could not be excluded as CADs might be more prevalent in older patients. PURPOSE/INTRODUCTION: The clinical association of coronary artery disease (CAD) with bone mineral density (BMD) has been increasingly reported, but findings on the relationship between the two age-related processes are conflicting. The aim of our study was to conduct a meta-analysis to evaluate the associations between CAD and BMD. METHODS: We searched PubMed, Embase, and Cochrane Library. Odds ratio (OR) and 95% confidence interval (CI) were pooled to assess the association between low BMD and the prevalence of CAD. For continuous data, standardized mean difference (SMD) with its 95% CI was pooled. Correlation coefficients of BMD and Gensini score were pooled after being transformed by Fischer z-transformation. Subgroup and meta-regression analyses were performed to explore the sources of heterogeneity. RESULTS: The meta-analysis involved 4170 participants from 11 studies. Pooled ORs for the incidence of CAD in patients with low BMD versus patients with normal BMD was 1.58 (95% CI 0.99-2.52, P = 0.06), and no statistical difference was found in men and women subgroups. After confounding age, the combined OR was 1.60 (95% OR 0.69-3.72, P = 0.27). Pooling data for comparing BMD of CADs and non-CADs were - 0.28 (95% CI - 0.47 to - 0.09, P = 0.004) in femoral neck and calcaneus, - 0.42 (95% CI - 0.89-0.05, P = 0.08) in lumbar spine, and - 0.25 (95% CI - 0.40 to - 0.11, P = 0.000) in the overall. A significance was detected in pooled correlation analysis between CAG Gensini score and BMD (COR = - 0.4435 [- 0.6647; - 0.1508], P = 0.004). No sources of heterogeneity were acquired, and no publication bias was identified. CONCLUSIONS: Low BMD was not associated with the prevalence of CAD. Without age adjustment, the BMD of CADs is significantly lower than that of non-CADs, and the patients with lower BMD are inclined to more severe coronary artery lesions.

Traumatic brain injury (TBI) is a major source of mortality and long-term disability worldwide. The mechanisms associated with TBI development are poorly understood, and little progress has been made in the treatment of TBI. Tanshinone IIA is an effective agent to treat a variety of disorders; however, the mechanisms of Tanshinone IIA on TBI remain unclear. The aim of the present study was to investigate the therapeutic potential of Tanshinone IIA on TBI and its underlying molecular mechanisms. Changes in microvascular permeability were examined to determine the extent of TBI with Evans blue dye. Brain edema was assessed by measuring the wet weight to dry weight ratio. The expression levels of CD11, interleukin- (IL-) 1 beta, and tumor necrosis factor- (TNF-) alpha mRNA were determined by reverse transcription-quantitative PCR. Aquaporin-4 (AQP4), glial fibrillary acidic protein (GFAP), and p47phox protein expression levels were detected by western blotting. Superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-PX) activities, and malondialdehyde (MDA) content were determined using commercial kits. Cell apoptosis was detected by western blotting and TUNEL staining. Tanshinone IIA (10 mg/kg/day, intraperitoneal administration) significantly reduced brain water content and vascular permeability at 12, 24, 48, and 72 h after TBI. Tanshinone IIA downregulated the mRNA expression levels of various factors induced by TBI, including CD11, IL-1 beta, and TNF-alpha. Notably, CD11 mRNA downregulation suggested that Tanshinone IIA inhibited microglia activation. Further results showed that Tanshinone IIA treatment significantly downregulated AQP4 and GFAP expression. TBI-induced oxidative stress and apoptosis were markedly reversed by Tanshinone IIA, with an increase in SOD and GSH-PX activities and a decrease in the MDA content. Moreover, Tanshinone IIA decreased TBI-induced NADPH oxidase activation via the inhibition of p47phox. Tanshinone IIA attenuated TBI, and its mechanism of action may involve the inhibition of oxidative stress and apoptosis.

Background. Administration of nonsteroidal anti-inflammatory drugs (NSAIDs) often causes small intestinal ulcers in patients, but few effective drugs are currently available to manage such serious adverse events of NSAIDs. Li-Zhong decoction (LZD), a well-known traditional Chinese medicine (TCM) formula, is commonly prescribed for treatment of gastrointestinal diseases. The present study aimed to investigate the anti-ulcerogenic activity of LZD on indomethacin- (IND-) induced duodenal ulcer in rats. Mechanistic studies of action of LZD were focused on involvement of TLR-2/MyD88 signaling pathway. Methods. Fifty male Sprague-Dawley (SD) rats were randomly and evenly divided into five groups: normal control, ulcer control (IND, 25 mg/kg), IND + esomeprazole (ESO, 4.17 mg/kg), and IND + low and high doses of LZD (3.75 and 7.50 g/kg). Macroscopic and histopathological examinations were performed for evaluation of ulcer index (UI), curative index (CI), and microscopic score (MS). Levels of duodenal inflammatory biomarkers and cytoprotective mediators including interleukin-4 (IL-4), IL-10, tumor necrosis factor-alpha (TNF-alpha), and prostaglandin E-2 (PGE(2)) were measured by ELISA. Expression levels of TLR-2 and MyD88 mRNA were assessed by qRT-PCR. The expression and distribution of TLR-2 and MyD88 proteins were analyzed by western blot and immunohistochemistry, respectively. Results. Gross and microscopic examinations of the IND-treated rats revealed severe duodenal hemorrhagic necrosis, inflammatory infiltration, villus destruction, and crypt abscess, while LZD-treated rats manifested these pathological events to a markedly lesser degree. LZD significantly decreased UI and MS, increased CI, preserved the integrity of the villus and crypt, and normalized the tissue architecture of the duodenum of rats. The elevated TNF-alpha levels in the IND-treated rats were markedly diminished in the LZD-treated rats, while lower levels of IL-4, IL-10, and PGE(2) observed in IND-treated rats were significantly increased in LZD-treated rats. Interestingly, improvement of immune function in duodenal mucosa by reduction of mRNA and protein expression levels of TLR-2 and MyD88 was also observed in rats treated with LZD. Consistently, immunohistochemical analyses revealed a lower co-localization of TLR-2 and MyD88 proteins in the duodenal mucosa of LZD-treated rats as compared to the IND-induced rats. Conclusions. Our data demonstrate that LZD protects the duodenal mucosa from IND-caused lesions, which is at least partially attributable to the interaction of its potential cytoprotective and anti-inflammatory mechanisms together with enhancement of the mucosal immunity through TLR-2/MyD88 signaling pathway.

Background: The dried rhizome of Polygonatum odoratum (Mill.) Druce has been widely used in traditional medicinal preparations in China, Japan, and Korea. In China, it is distributed in Hunan, Guangdong, and Liaoning provinces, and its quality differs from habitat to habitat. In addition, P. odoratum has some adulterants, such as Polygonatum inflatum Kom, Polygonatum prattii Baker, and Polygonatum cyrtonema Hua. The morphological traits and chemical composition of the aforementioned adulterants have many similarities with those of P. odoratum. Therefore, it is possible that people often use adulterants instead of P. odoratum for clinical treatment. Objectives: We aimed to establish a reliable and accurate classification model of P. odoratum based on the support vector machine (SVM) and identify it from different habitats; we also aimed to identify its adulterants. Materials and Methods: In this study, we first determined the ultraviolet (UV) absorption spectrum of the water extract of the rhizome from 162 samples (including P. odoratum from Hunan, Guangdong, Heilongjiang, Yunnan, and Liaoning Provinces and adulterant species including P. inflatum, P. prattii, P. cyrtonema, and Disporopsis pernyi (Hua) Diels) by UV-visible spectrophotometry. The UV absorption data were preprocessed with the SVM model before establishing the habitat and other details. Results: According to our results, the SVM model showed a prediction accuracy of 100%. The model accurately identified five different habitats and four different adulterants of P. odoratum. Pretreatment of samples with UV spectrum might be useful in the accurate identification of P. odoratum. Conclusion: The SVM model seems very prospective in identifying herbs with multiple habitats and its adulterants.

The current research tried to explore the effect of Qiweibaizhu powder (QWBZP) on the bacterial diversity and community structure of the intestinal mucosa of dysbiosis diarrhea mice and provide a scientific basis for the efficacy of QWBZP on antibiotic-induced diarrhea. A dysbiosis diarrhea mouse model was constructed with broad-spectrum antibiotics through a mixture of cephradine capsules and gentamicin sulfate (23.33 mL center dot kg(-1)center dot d(-1)). Intestinal mucosa was collected, and DNA was extracted from each group. The bacterial characteristics in intestinal mucosa were analyzed by MiSeq sequencing based on the 16S rRNA sequencing platform. There were no significant differences in alpha diversity indices among the three groups. The sample distributions in both the normal and QWBZP groups were relatively concentrated, and the distance among individuals was close. However, an opposite result was obtained in the model group. Furthermore, the composition and abundance of species were similar between the normal group and the QWBZP group at both the phylum and genus levels. After treatment with QWBZP, the abundance of Lactobacillus increased, and Proteobacteria decreased, and the Firmicutes/Bacteroidetes ratio decreased to a normal level. Our results indicate that QWBZP can help repair mucosal bacterial structure and recover mucosal microbiota. Specifically, QWBZP increased the abundance of Lactobacillus and Bacteroidales S24-7 group norank.

OBJECTIVE: To dynamically observe the efficacy of Jieduan Niwan formula (JDNW) on a rat model of acute-on-chronic liver failure (ACLF). METHODS: Seventy Wistar rats were divided into control group (6 rats), model group (22 rats), JDNW group (21 rats), and SP600125 group (21 rats). 13 weeks' porcine serum injection followed with D-galactosamine and lipopolysaccharide joint acute attack was used to establish ACLF model. Rats in JDNW group were orally given JDNW formula for 3 days before acute attack; rats in SP600125 group were injected with SP600125 30 min ahead of acute attack. Rats were sacrificed respectively at 4, 8 and 12 h after model established. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TBIL), Creatinine (CR), blood urea nitrogen (BUN), prothrombin activity (PTA) were examined by biochemical process, Tumor necrosis factor-alpha (TNF-alpha), interleukin-1 beta (IL-1 beta) interleukin-6 (IL-6), interleukin-10 (IL-10), transformed growth factor-beta 1 (TGF-beta 1), High mobility group box-1 (HMGB-1), CD3, CD4, CD8 were analyzed by enzyme-linked immunosorbent assay, apoptotic index (Al) was detected by terminal-deoxynucleoitidyl transferase mediated nick end labeling staining, expression of Bad, phosphorylated Jun N-terminal kinases (p-JNK) and Cytochrome C (Cyt C) were detected by immunohistochemical analysis, Bax and Bid were detected by Western blot analysis. RESULTS: In model group, the levels of ALT, AST, TBIL, CR, BUN, IL-1 beta, IL-6, IL-10, TGF-beta 1 and HMGB-1 remarkably increased and PTA decreased compared with control group (P < 0.05), as time goes on, ALT, AST, TBIL, CR, BUN, continued to grow, while IL-1 beta, IL-6, IL-10, HMGB-1, TGF-beta 1 and PTA gradually decreased; massive necrosis could be seen; the levels of TNF-alpha, CD3, CD4, CD8, Al, p-INK, Bax, Bad, Bid and Cyt C increased at 4 h and peaked at 8 h, but decreased at 12 h (P < 0.05). JDNW group, by contrast, showed less pathological injury, increased PTA level, and reduced ALT, AST, TBIL, TNF-alpha, IL-1 beta, IL-6, IL-10, TGF-beta 1, HMGB-1, CD3, CD4 and CD8 levels (P < 0.05), moreover, the Al and expression of p-JNK, Bax, Bad, Bid and Cyt C were lower than model group at 4 and 8 h but were higher at 12 h (P < 0.05). Similar results were observed in SP600125 group. CONCLUSION: An ACLF rat model with low mortality can be established by porcine serum joint with D-galactosamine + lipopolysaccharide induction; JDNW decoction can effectively suppress the inflammatory reaction, improve the immune system, and protect the liver of ACLF rats, the mechanism might involve the inhibition of the JNK-induced mitochondrial apoptotic pathway. Voltage-gated sodium channels (VGSCs), which are abnormally expressed in various types of cancers such as breast cancer, prostate cancer, lung cancer, and cervical cancer, are involved in the metastatic process of invasion and migration. Nav1.5 is a pore-forming alpha subunit of VGSC encoded bySCN5A. Various studies have demonstrated that Nav1.5, often as its neonatal splice form, is highly expressed in metastatic breast cancer cells. Abnormal activation and expression of Nav1.5 trigger a variety of cellular mechanisms, including changing H(+)efflux, promoting epithelial-to-mesenchymal transition (EMT) and the expression of cysteine cathepsin, to potentiate the metastasis and invasiveness of breast cancer cellsin vitroandin vivo. Here, we systematically review the latest available data on the pro-metastatic effect of Nav1.5 and its underlying mechanisms in breast cancer. We summarize the factors affecting Nav1.5 expression in breast cancer cells, and discuss the potential of Nav1.5 blockers serving as candidates for breast cancer treatment.
